The occurrence of the HW element in Welsh English

The aim of this diploma thesis is to investigate the accent realizations of the initial consonant in wh- words in Welsh English. The theoretical background of this study deals with the development of Welsh language and its interactions with the British standard variety. Together with the previously mentioned the phonological inventories of Welsh, RP, and Welsh English are described in the first part. We further analyzed the development of the <hw> element in the British Isles in general. The research part of this study is concerned with the analysis of the data from 20 speakers of Welsh English. Three speech styles were used for the final analysis: spontaneous speech (informal style), isolated words (formal style) and reading of Cinderella (formal style). Signal-to-noise ratio and voicing of the <hw> segments were measured in order to discover the occurrence and the nature of the <hw> element. The lowest levels of harmonicity were measured in segments that were obtained from the formal reading style. The highest levels were measures in the spontaneous speech tokens. Younger speakers showed higher levels of harmonicity than older speakers. The results of the male and the female participants were different in each type. Design and implementation of a Rezervacnik service

The main objective of this thesis is the design and the realization of the SaaS project Rezervacnik. The thesis itself consist of three parts. The first part defines the SaaS and Cloud Computing, and the establishment of basic advantages and disadvantages of SaaS services. The second part analyzes the information model of the SaaS provider. Introduce are the basic application billing and subscription software for the management of SaaS, which are researched and compared. The last part presents Rezervacnik project and defines the advantages of online booking. After finding a market assumption are designed all parts of the project. Finally, it is presented as part of the implementation.

V úvodu i dále v textu zmiňuji, že východiskem mé práce, jedním ze základních kritérií pro vytvoření výběrového souboru je kontext hodnotových orientací, a právě z tohoto důvodu uvádím teoretickou část své práce krátkým představením pojmů, jež mají zásadní postavení v sociologické teorii i empirickém výzkumu. Pojem hodnoty má svůj původ v oblasti ekonomie resp. filosofie; jakožto k základní sociologické kategorii se k hodnotě již od jejího vzniku váže mnoho různých významů a interpretací. "Podle Clyda K. M. Kluckhohna ( 1951) je hodnota explicitní nebo implicitní pojetí o tom, co je žádoucí, pojetí příznačné pro jednotlivce i pro skupinu, ovlivňující výběr přístupných prostředků a cílů jednání." City as a fictional character in the postmodernist novel: Alexandria Quartet and The Moor's Last Sigh

in English This Bachelor thesis engages with the topic of a portrayal of the city in the postmodern novel which is then studied on the example of two chosen novels: Alexandria Quartet by Lawrence Durrell and The Moor's Last Sigh by Salman Rushdie. Both novels present the urban space as a female character with a strong influence upon the events and the formation of the protagonist's identity. The second chapter is dedicated to a theoretical framework which helps to establish what we can understand under the term modernist literature and it traces the basic features such a literature possesses. For this framework I adopted the study of postmodernism presented by the work of Linda Hutcheon, mostly her A Poetics of Postmodernism. A special attention is dedicated to the aspects of postmodernism that can be applied on the portrayal of space and the relationship existing between postmodernism and modernism. The third chapter outlines the basic concepts of space established by modernism. The major concepts dealt with in this thesis are: subjectivity, perspective, palimpsest and myth. Cloning and expression of human carbonyl reductase CR-1

The purification of pOTB7 plasmid containing coding sequence of carbonyl reductase1 (CBR1) in cells of Escherichia coli (E. coli) was done by alkali hydrolysis. The sequence of CBR1 was multiplied by polymerase chain reaction (PCR) and synthesized primers with restriction sites were used. The left primer contained sequence for restrictive endonuclease NdeI and the right primer for restrictive endonuclease XhoI. Validation of the first step was confirmed by size measurement of the synthesized fragment with following restrictive analysis realized by restrictive endonuclease BamHI. Prepared sequence CBR1 was cloned into Topo vector, which was transformed into the competent E. coli cells. Topo vector was purified by alkali hydrolysis after innidation of E. coli cells. The size of cyclic Topo vector without CBR1 and Topo vector with CBR1 coding sequence was verified on agar gel by restrictive endonuclease BamHI. This was the validation of the second step. Subcloning of coding sequence CBR1 from Topo vector to express vector pET15b was the last step.

This master’s thesis is focused on the concept of the assessment centre method relevant for the positions of the guide, the expert salesman and the information centre employee, which are staffed in MOSER, a.s. company. The attention is located on the determination of the terms, the estimation of the specific positions through the project. The main aim was given on the realization of the assessment centre and especially to the proposals related to its implementation.

The Value of Touch in the Nursing Profession

This thesis concentrates on the issue of touch - based communication in general nursing in the area of emergency care. The theoretical part drala with the value itself, the importace of the touch in the life of a man, the role of physical contact within the scope of nursing profession, and the possible consequences of the touch. Further, the thesis is concerned with the issue of the therapeutic touch and its importance, versatility of means of non-verbal communication, and some selected dissimilarities of non-verbal communication within the field of multicultural environment. The last section of theoretical part of the thesis is engaged in the questions of burnout. The research project, which is presented in the practical part of the thesis, is aimed at the realization of the process of the touch-based communication between the nurse and the patient in selected departments and wards, and the variety of factors, which influence the process of communication. I have used two types of questionnaires in the research. The first one is my own question-form and the other one is the officially approved questionnaire. Using Stratego/XT for Generation of Software Connectors

Software connectors are used in component based systems as a special entities modeling and realizing component interactions. Besides this behavior, connectors can provide extra functionality and benefits (e.g. logging, adaptation, monitoring). This approach requires generation of connector code with respect to requirements of components, a target environment and features specified at the design stage. In this thesis we show how to extend the existing connector generator [33] by the Stratego/XT transformation engine, which includes a language for implementing program transformations and a collection of transformation tools. We use the toolset to realize a simple method of defining connector implementation, which is use as a template for a process of generation source code.

Interactive Java applet used for 3D optic nerve head visualization

The aim of this thesis was to design and implement the interactive Java applet used for topography 3D optic nerve head visualization. Primary purpose of the 3D vizualization is ophthalmology, especially for diagnosis of glaucoma. More over should serve as a training material, which enables to study the transformation of 2D figures to 3D model and to test the effects of adjustments and regulations to imaging of the model in 3D space. Two program versions were designed in this work using the development system NetBeans version 5.5. Their final realization is implemented as .html application working in web browser window. First version of the designed applet is simple intuitive application with several default settings, so that the figure is opened automatically to 3D model. Subsequently, the applet enables to set the size of the image, 3D depth of the image, smoothing of the surface and selection of monochrome or coloured image. The second version is the extension of the first version of the designed applet, and provides the possibility to display x, y, z axis, depiction of the 3D model surface by dots, curves or grid, and illumination of the surface. Software was tested on available topographic data acquired by HRT equipment and in different web browsers. Technical documentation and user’s manual are also involved in this thesis.

Monolog as an authentic confession in the context of the contemporary drama

The thesis deals with the notion of monologue and monologicity as a peculiar means of expression of the modern drama. The concept of monologue is defined within the frame of its relation towards dialogue, in the context of theoretical thinking about drama and theater, and in terms of the application of these concepts in everyday life.It is based on the theoretical works by Jan Mukařovský and Jiří Veltruský. The concept of monologue as a lack of dialogicity is being challenged with understanding monologue as a haven of authenticity, a space for authentic testimony. Through the general reflection, thoughts are put in connection with modern drama and illustrated on examples from particular drama works. The thesis leads into a reasoning about the dialogue between the stage and the auditorium as a way-out from the monological situation on the stage.
