Distinct eco-phenomena in Central Europe

The subject of essay is eco-phenomena and its influence on landscape diversity. The essay contains definitions of particular eco-phenomena types and attributes elaboration focused on extraordinariness of this effect. Each eco-phenomena type is described, his uniqueness is illustrated on examples of local vegetation types and eco-phenomena description is completed by concrete locations with this sort of effect. Next part of the essay contains information about distribution particular eco- phenomena locations on the Czech Republic territory. There are information about description mentioned locations and sorting according to bio geographic classification of the Czech Republic. There is map included. Amphibian habitat preferences in selected localities after mining moldavites.

This thesis pursues amphibian biotop preferences in locations where moldavites were quarried in the past. Field work took place in two localities near Trhové Sviny. The first one is situated near the village of Slavče where an illegal extraction still occures,.The second locality is found by the village of Dobrkovská Lhotka. This locality was partially recultivated in 2010. The aim of this work was to establish which factors do influence different amphibian species in the both studied locations. There were found five various species of amphibians near the village of Slavče: the Common frog (Rana temporaria), the Edible frog (Pelophylax kl. esculentus), the European fire-bellied toad (Bombina bombina), the European tree frog (Hyla arborea) and the Common toad (Bufo bufo). The results were statistically evaluated. Based on them it was established that the Common frog prefers the herb layer, the European Fire-bellied toad prefers semi shaded medium sized pools that do not dry up. The European tree frog and the Edible frog prefer large areas of pools and ponds. More over the edible frog demands ponds that are partially overgrown. The Common toad was represented only with one single individual. Therefore a statistical analasys could not be executed. The whole locality is negatively influenced by local inhabitants who gather here manure and organic waste from their gardens. On locality located near to Dobrkovská Lhotka was transformed from a mining landscape to a pond and three pools. All amphibian species that lived in the locality before reclamation were still present here. Occurrence of Common spadefoot (Pelobates fuscus) was confirmed by the unique finding of a tadpole. Statistical evaluation was not conducted due to the small number and variability of monitored sites.

The polymorphism of sericin 2 gene in silkworm (Bombyx mori)

In our study, we examined the polymorphism of gene Ser2 from domesticated silkmoth Bombyx mori and its closest wild relative B. mandarina. As a starting material for our work, we used the restriction map of allele C isolated from hybrid lineages 200 and 300 of European silkmoth B. mori (Michaille et al. 1990a). We also used the published sequence of allele D which was isolated from {\clq}qDaizo`` p50 strain of B. mori (Kludkiewicz et al 2009). Based on the published sequence, we designed PCR primers and performed sequence analysis of 4 different alleles of gene Ser2, including the almost complete genomic sequence of allele C (except for two short regions containing repetitive DNA). We then compared the sequences and found that the alleles differ significantly not only in intron lengths and the sizes of exon 9, but also in exon arrangements. While the allele C contains 12 exons (1, 2, 3, 4, 5, 6, 7, 8a, 9, 10, 11, 8b), the alelle D and other alleles analysed have 13 exons (1, 2, 3, 4, 5, 6, 7, 8, 9a, 10a, 9b, 10b, 11). Allele C showed very unique arrangement of exons, which suggested that it may represent an ancestral form of Ser2 gene. We analysed the arrangment of exons located at the 3´end of the Ser2 gene by PCR and electrophoresis in 70 available strains of B. mori and B. mandarina. We also tried to find a similar allele to the allele C and we amplified and sequenced 2 kb region from 55 Bombyx strains. Phyllogenetic analysis of gene Ser2 gene suggested that the C allele of now probably extinct European silkmoth hybrids 200 and 300 does not seem to be an ancestral form of Ser2 gene, but it rather seems to be the result of complex secondary rearrangements.

Analysis of the value of ecosystem services by stated preference methods: Case study of clearance cairns in the Eastern Ore Mountain

During last decade there has been developed concept of so-called ecosystem services (e.g. Millennium Ecosystem Assessment, 2005) in matured countries. This concept focus especially on identification of benefits resulting from intact ecosystem and also on various ways of taking all those benefits into account when dealing with decision-making process of market economy. Goal of this concept (among other things) is to keep currently freely available ecosystem services in nature as they are and also to ensure natural conditions for life won't be becoming worse. Landscape of eastern Ore Mountain has its specific character especially because of few unique ecosystems which don't occur in other locations at all; or they are presented there but not in such high volume. We can consider mountain meadows, natural streams and clearance cairns at the most important local unique ecosystems. Main goal of diploma thesis is to analyze recreational and mainly aesthetic values of clearance cairns. Even though clearance cairns are important biotopes there was quite small attention dedicated to them in existing analyses made in Czech Republic. That is in heavy contrast to attention dedicated to those biotopes in other areas, e.g. in eastern part of Ore Mountain belonging to Saxony where local inhabitants appreciate clearance cairns so much that every single section of them is completely documented (resulting into 1.000 kilometers of documented clearance cairns). Diploma thesis is based on methodology using analysis of primary data obtained from empirical research. Research was realized in eastern Ore Mountain during summer 2013 and author of this thesis was actively participated both in preparation phase and also in realization phase. Thesis also uses random utility theory, discrete choice models and also stated preferences methods (e.g. Bateman et al., 2002). Multinominal logit model and random parameter logic model are used for values estimation. Based on performed analysis thesis verifies or disproves hypothesis saying that willingness to pay for clearance cairns is statistically insignificant (i.e. people don't consider this landscape element as having significant value). Diploma thesis results will be used as a suggestion for obtaining clearance cairns management.

Václav Voska's theatre, film and television acting in Czech artistic and social context in the second half of twentieth century

SUMMARY: This thesis focuses on theatre, film and television acting of one of the important representatives of Czech dramatic art in Czech artistic and social context in the second half of the twentieth century. The aim of this work is to depict means of expression used by Vaclav Voska in his roles. It provides summary of actor's theatrical work and in the part dedicated to his TV acting tries to comprehensively describe compering television series "We invite you to the theatre". The following part is dedicated to film acting, especially to his cooperation with the Film Academy of Performing Arts in Prague. The work neither neglects his teaching at the Faculty of Performing Arts Theatre nor his political career. The essential part of this thesis is focuses on his role of Emile Magis in Félicien Marceau's game The Egg. The work purposely omits on Vaclav Voska's production of spoken word (radio, Poetic Cafe Viola, Lyra Pragensis) and it refers only to his major achievements only if they are relevant for analyzed topics. Characterisation of recombinant cathepsins B of the bird schistosome Trichobilharzia regenti

This study focuses on the recombinant cysteine peptidases - cathepsin B originating in the bird schistosome Trichobilharzia regenti that is unique across the whole family for its ability to migrate through the nerve tissue to the final localization. For invasion, migration, degradation of nutritional proteins and/or evasion of host immune responses, schistosome employs peptidases. This study follows the research done by researchers of Department of parasitology, Faculty of Natural Sciences, Charles University. The main goal of this study was to deepen the characteristics of recombinant cathepsins B originating in T. regenti. In T. regenti, two cysteine peptidases - cathepsins B1 (TrCB1) and B2 (TrCB2) - have been previously characterized. TrCB1 is located in the gut of schistosomula and involved in digestion. TrCB2 occurs in post-acetabular penetration glands of cercariae and probably facilitates penetration. The recombinant pro-cathepsin B (isoforms TrCB1.1, TrCB1.4 and also TrCB2) were expressed in Pichia pastoris yeast system. An attempt was made to produce in P. pastoris the recombinant isoform TrCB1.6, in which the active site cysteine is substituted by glycine. Wandering trought Genius loci (Cultural landscape of Bohemia)

The thesis focuses on landscape in the context of culture, or "cultural landscape" from the perspective of genius loci - "spirit of a place" - which on one hand consists of unique subjective feelings, yet also transcends subjectivity. At one level the atmosphere of a place is created by nature. However, at another level the presence of the human element is indispensable - "dead men" determine the overall spirit of each location. The overall spirit is thus an expression how landscapes appropriate us. (From this perspective the spirit of the place is not just a fleeting and purely subjective impression, but an expression of the fact that, in a manner of speaking, we are appropriated by the land ourselves.) This thesis discusses in fact two different points of view: the anthropological perspective (the subject produces nature, nature produces the subject) and the perspective of the landscape itself (landscape has its own quality - from nature's own point of view the Baroque period is a meaningless term). In order to study genius loci it is necessary to find some tools to help us describe it within the context of human history and culture. Title of the dissertation: Reconstruction of a criminal offence Key words: reconstruction, criminal offence, criminology Abstract This dissertation deals with reconstruction of criminal offence, its definition, the tactics used and the relevant aspects of the criminal law. The first chapter is concerned with the term of 'crime reconstruction'and the use of the crime reconstruction, discussing the historical development of crime reconstruction together with the development of the views and opinions dealing with the crime reconstruction. The second chapter considers specific types of crime reconstruction, that is a reconstruction of criminologically significant crime locations, significant objects and significant signs of a criminal and, finally, the reconstruction of criminologically significant actions and events. The third chapter deals with the legal aspects of the reconstruction of criminal offence, specifically its embedding in the criminal law. The fourth chapter discusses the difference between the reconstruction of criminal offence and the other methods of investigation, such as the initial investigation, verification of statements on the site and the inspection of the scene of crime. Sightseeing Tours - the Analysis of Transport and Accommodation Solutions

This bachelor's thesis deals with the analysis of transport and accommodation facilities from the random selection of sightseeing tours of more or less important travel agencies. In the first part there are defined various concepts related to the topic which are used in two ways, the analysis, and creation of own sightseeing tour. The aim of this bachelor's thesis is to create an analysis, and define the most frequently used accommodation, type of transport and related overall sightseeing tour. Another aim is to create an unique tour for the Slovak Republic, because Slovakia is very attractive country for tourists. It has a rich representation of different cultural, historical and natural sights which are worth visiting. The tour is interesting because of connections with hiking, cruises along the rivers and visiting various cultural sites.

Café - an architectural phenomena of the First Czechoslovak Republic´s society

This bachelor thesis deals with restored cafés in Brno and Prague that were established in the First Czechoslovak Republic. In the case of Brno´s coffee houses it is mostly emphasized their origin, historical context, history of coffee making and how the coffee houses returned to their former look and function. The author deals with the analysis of coffee houses and the purposes and goals of their restoration. The author focuses on Zeman´s café by the architect Bohuslav Fuchs and it´s replica from 1995 and on ERA coffee house by Josef Kranz. Then the focus is shifted to an elegant coffee house Savoy owned by Jan Nekvapil and designed by Jindřich Kumpošt and to a pastry shop and coffee house Kolbaba designed by Jan Víšek. To compare evolution of the coffee houses in Brno the author chose unique cubist coffee house Grand Café Orient in Prague located in Dům U Černé Matky Boží by the architect Josef Gočár. The thesis summarizes the circumstances of the formation of coffee houses during the First Czechoslovak Republic in Prague and Brno and examines different conditions for its development. It also looks into the circumstances of restoration and today´s activity of these coffee houses and evaluates its success rate.
