23 7 SUMMARY This work deals with the role of adipose tissue-derived hormones - adipokines in the development of insulin resistance in pregnant women with gestational diabetes mellitus (GDM). The study consists of two parts. In the first one, the characterization of changes in concentrations of circulating adiponectin - the most abundant adipokine - during menstrual cycle in healthy women was performed. The second part was focused on the changes of gene expression in pregnant women with GDM relative to healthy pregnant controls (14 vs. 13 subjects). Specificial attention was focused on adipokines and their receptors (ADIPOQ, ADIPOR1, ADIPOR2, LEP, RETN, TNF , CFD a PAI1) as well as other genes potentially contributing to the etiopathogenesis of GDM in the samples of placenta, subcutaneous, and visceral adipose tissues obtained during surgical delivery. Psychological intervention in company management

Paper describes psychological intervention in organization based on Organization Development approach. Theoretical part presents principles, methods and techniques of this discipline, and relevant theory of organization. Key concepts of organizational character, organizational climate and organizational culture are amended with developmental aspect in several theories of organization growth. The principles imply practical advice for implementation of organizational change. This provides theoretical and practical backup for the following intervention. The practical intervention focused on transformation crisis in a production company. Intervention followed action research paradigm with certain practical limitations. Final results are supplemented with recommendations for company intervention derived from practical experience.

Caveolae and caveosoms

Caevolae are remarkably stable structures at the plasma membrane. They form specific domains distinct in lipid composition from the rest of plasma membrane. Many diverse functions are assigned to Caevolae. They play role in modulation of cellular surface, signalization and well regulated endocytosis. Caveosomes suppose to be large intracellular vesicular structures potentialy new membrane organels. They are derived from internalized caveolae. Tohether with caveolae they are proposed to form a separeted system of intracellular vesicles. However recent evidence suggests that caveolae can fuse with endosomes immediately after internalization. If this is true, then the system of vesicles derived from caveolae, including caveosomes, can be considered a regular component of endosomal system. Isolation of caveosomes from endosomes has been seen mainly in experiments where polyomavirus SV40 was used. Thus the question, if this isolation is not just a result of SV40 infection, arises. It has been shown recently that SV40 virus is capable of inducing caveosome-like structures even in the absence of caveolae. Consequently existence and properties of caveosomes are being questioned. Analysis of the career advisory system on the first level of ISCED in the Czech Republic

The main goal of my diploma thesis is to define some dimensions of career counselling. Primary and secondary source expert literature (aimed at individual psychology, pedagogy and career counselling) is analyzed in the theoretical part and the results of the study are presented in a synthetic form with the expression of my point of view about given thesis. For the research work I have used literature search, analysis, compilation and resumes of specialized literature. In the practical part of the work I have chosen with regard to the professional literature a questionnaire and individual guided interviews with career counselors, that revealed the findings about the respondent's approach to information and communication technologies, and their own professional experience. Consequently I created comparative tables and evaluated all given information. The survey revealed that ICT-oriented counselors who derive benefit from available modern technologies are more flexible than their colleagues. Counselors with a conservative approach are able to accept modern ICT only when they have adequate training and support.

The issue of the concept of criminal custody

Resumé The purpose of my thesis is to analyze legal regulation of custody in Czech law. Custody is a part of criminal law procedure, which is related to deprivation of fundamental human right of personal liberty. The aim of this thesis is also to explain some problems with the application of this legal measure custody and ilustrated disunited in interpretations of some provisions of a statute. The thesis is composed of six chapters. The thesis begins with a short introduction, which presents the main perspectives on the topic and the thesis. Chapter One describes the development of custody in Czech law focusing on the time period since 1918 and is divided into three subchapters by the historical backround of evolution of Czekoslovakia. Main attention is dedicated to changes after Velvet revolucion. Chapter Two discusses concept of custody in Czech law and EU. The chapter consists of three subchapters. Caspase inhibitors and regulation of the IL-1 family cytokines

Proinflammatory cytokines from IL-1 family play a key role in immune and immunopathological reactions and are involved also in initial phases of rejection mechanisms. One of their multiple functions is the induction of chemokines attracting immune cells into the site of injury. The aim of our study was to assess the effect of proinflammatory cytokines on the chemokine release and mRNA induction in epithelial cell lines of renal and lung origin. Furthermore, we tested the effect of caspase-1 inhibitors isolated from Streptomyces nodus (manumycin, asukamycin, MM273) on the release of IL-1 beta and IL-18 from human macrophages. These cytokines are cleaved into their active form by caspase-1 and inihibtion of the enzyme activity might be a perspective therapeutical approach to reduce inflammatory reaction. In our experimental model, we used cell line derived from renal adenocarcinoma (RA), A549 cell line (alveolar type II-like cells), and THP-1 monocyte/macrophage cell line. Chemokine mRNA induction was evaluated by an oligoarray and chemokine and cytokine levels were measured by Luminex or ELISA technology. Genetic interactions of PRP45 mutantion in S. cerevisiae

Human SKIP/NcoA-62/SNW1 ortholog proteins are nuclear factors which have been found in connection with the regulation of gene expression and subsequent modifications of primary transcripts. Human representative SKIP acts primarily as a transcription factor during the processes involving differentiation, carcinogenesis and other cellular signaling. It operates in the signaling pathways, eg TGFß/Smad, E2F/pRb, Notch/CBF1 and nuclear hormone receptors. SKIP protein is also essential for pre-mRNA splicing and recently its contribution during the elongation phase of RNA polymerase II transcription and contribution to some HIV genes alternative transcripts formation has been identified. The ortholog of S. cerevisiae FUN2O/Prp45 has been shown to participate in pre-mRNA splicing, others processes mentioned have not yet been associated with the function of this factor. Pre-mRNA splicing is a complicated process leading to the formation of native mRNA lacking non-coding regions, introns. The Role of Dipeptidyl Peptidase-IV in Glioma Cell Growth

Dipeptidyl peptidase-IV (DPP-IV, CD26, EC 3.4.14.5) is a widely expressed serine protease that by limited proteolysis regulates a number of biologically active peptides including a number of mitogenic peptides involved in cancer development. Deranged DPP-IV expression and/or enzymatic activity has been reported in a number of tumors, and could lead to altered signaling and biological function of its substrates. Changes in DPP-IV expression were observed in glioma cell lines in association with differentiation, and recently also in malignant gliomas in vivo. In addition, DPP-IV substrates substance P (SP) and stromal cell derived factor-1 (SDF-1) that trigger growth promoting intracellular signaling cascades in glioma cells have strongly been implicated in the pathogenesis of gliomas. The aim of this study was (i) to investigate the effects of DPP-IV on the signaling of its biologically active substrates that promote the malignant phenotype of glioma cells, and (ii) to assess the effects of DPP-IV on the growth, migration and adhesion of human glioma cells. Corporate communications and PR on the Internet

This paper deals with three basic problems, which also examines and analyzes. It is based on comparison of three company's efforts to effective public relations. Author derives benefit from his work experience from three different projects in the position of the person responsible for public relations and marketing. The Paper as a result compares options and strategies of making effective public relations in three different large companies. In addition, comparative studies of these three projects, which monitor the strategies of different companies and also the use of the Internet and other technologies for greater efficiency resulting effort, the paper will focus on analyzing the close relationship between marketing and public relations. Contribution of this paper is a new view on internet PR, in the light of term its relationship to an informative system used by organization, especially the part of Customer Relationship Management for managing relations with the public. By comparing the different strategies will also help to detect mistakes in the implementation of public relations as well as selecting appropriate means of internet PR. The third and fourth chapter describes the integration of public relations in the modern internet community and communication, and also in the marketing mix of organizations and companies. In next chapters, theoretical knowledge has passed to practical examples of Digital Sun music festival and music Club Na Rampě. The widest use of information technologies and IT/IS organizations will be seen on the example of accessing PR and marketing into the company Mironet, Ltd.

Leadership in a czech subsidiary of a multinational company

This diploma thesis is analysing the conditions of leadership development for potential leaders in Hilti spol.s r.o.. In this intention the theory of leadership is elaborated, further existing processes inside the company are described and finally data derived from Hilti internal research are evaluated. Information is supplemented by the findings derived from an e-questionnaire created for the purpose of clarifying uncertainties from the previous research data. To identify the objectives the thesis aligns existing processes with the theory of leadership and with the empirical researches. Exploiting these data within the company it is possible to identify strengths and weaknesses of leadership development processes for young leaders as well as propose areas which would be capable of implementing new trends with focus on higher effectiveness of these processes.
