The Function of Speech in Husserl and Merleau-Ponty

The Function of Speech in Husserl and Merleau-Ponty The submitted doctoral thesis is an attempt to describe the development of the intentional function of speech in Husserl and Merleau-Ponty. The intentional function is defined as the change of expressed meaning that is engendered by the expression itself. We trace Husserl's position from the Logical Investigations and the first book of his Ideas pertaining to a pure phenomenology and to a phenomenological philosophy, where he describes speech as the non- productive mirroring of other kinds of intentionality, to the late text The Origin of Geometry, where he discerns two functions of speech: it provides thought its ideality, which is different from the ideality of species; and it provides thought its objectivity, i.e. the form of object that lasts in history as identical. In The Phenomenology of Perception, Merleau-Ponty adopts Husserl's late position with several profound modifications. The starting-point ceases to be the linguistic sign, and speech becomes a kind of gesture. As a consequence, the difference between linguistic and non-linguistic ideality disappears. Aesthetic Factor in John Dewey's Conception of an Experience Aesthetic Relevance demonstrated by Example of Snowbording

This diploma thesis examines John Dewey's concept of experience, focusing especially on his notion that an aesthetic factor is present in every complete and consummatory experience. It deals with the differences between a common experience and "an experience" and examines how an aesthetic factor works in them. The indispensable role of the aesthetic factor is intentionally demonstrated on the non-artistic example of experiences of snowboarding. The most important findings are developed through the approach of Richard Shusterman's somaesthetics. The theories of these two philosophers support the main idea of this thesis, namely that dynamic movements of the body can bring aesthetic experiences to their actor. Thus, some of the reasons that explain the attractiveness of this modern sport come to light that are not apparent at first glance.

Preparation of the 14-3-3 Protein Binding Partners for Structural Studies.

Tyrosine hydroxylase belongs to the group of hydroxylases of aromatic acids and catalyzes a key step in the biosynthesis of catecholamine neurotransmitters. The tyrosine hydroxylase possesses the homotetrameric structure and contains three structural domains: the N-terminal regulatory domain, the catalytic domain and the C-terminal tetramerization domain. The activity of tyrosine hydroxylase is regulated by phosphorylation and through the regulation of its expression. Phosphorylation at Ser-19 induces binding of the 14-3-3 protein, which affects the structure of the regulatory domain and protects it against both dephosphorylation and degradation. Since the structure of the regulatory domain is still unknown, we decided to perform its structural characterization using NMR techniques. First, the expression and purification protocol of the regulatory domain of tyrosine hydroxylase was optimized. The protein was expressed as a His-tag fusion protein and its purification is composed from two steps: the chelating chromatography and the size-exclusion chromatography. The dynamic light scattering and the 1 H nuclear magnetic resonance were used to verify its monodispersity, and hence its suitability for further experiments.

Etela Kouklíková Supervisor: RNDr. Eva Novotná, Ph.D. Title of diploma thesis: Study of the effect of cyclin-dependent kinase inhibitors on the expression of selected AKR and CBR enzymes in human cell lines Cyclin-dependent kinase inhibitors (CDKi) are considered as a suitable treatment especially in patients with wrong prognosis or advanced stage of cancer. It has only recently been discovered that CDKi are able to influence the activity of some enzymes from aldo-keto reductase (AKR) and short-chain dehydrogenase/reductase (SDR) superfamilies. AKR and SDR enzymes belong to a group of carbonyl reducing enzymes that are involved in the metabolism of endobiotics and xenobiotics. An important group of drugs that are metabolized by these enzymes to less efficient compounds are anthracyclines. The aim of this diploma thesis was to find out whether purvalanol A, roscovitin, dinaciclib, AZD5438 and R547 can affect the expression of the most important anthracycline reductases (AKR1A1, AKR1B10, AKR1C3, AKR7A2 and CBR1) in human HepG2 and HL-60 cell lines. Perception and expression of non-verbal vulgarity in context of gender

Vulgarity is becoming to be an increasingly discussed and interesting topic, although not deeply explored by science. This thesis includes a survey focused on vulgar gestures. The main objective is to compare differences in perception and expression of non verbal vulgarisms- gestures among 1) boys and girls 2) two different teenage groups, in terms of frequency, form and purpose of using the gestures. I mainly use a closed, semi-closed or open questions in the questionnaire, which also includes a parallel presentation of each gesture. Preparation of expression vectors for the production of ZIKA and Dengue NS3 helicase

Zika and Dengue viruses have spread, due to globalisation, to all continents which lie at least in part in the subtropic and tropic climatic zones. This spread of these viruses is a reason of an increasing number of severe diseases caused by them. New drugs, which would be effective against these infections, could be an answer to this challenge. Various viral proteins, among them also viral helicase, which is the topic of this bachelor thesis, can be a suitable drug target. The task was to prepare expression constructs for production of recombinant helicases of Zika and Dengue viruses via the suitable bacterial strain Escherichia coli. Several constructs derived from plasmid pET-16b were prepared with inserted helicase of Zika and Dengue viruses. One of them was used for the preparation of recombinant purified helicase of Zika virus, that will be used for further research. PharmDr. Martina Čečková, Ph.D. Title of diploma thesis: Expression of selected membrane transporters in placentas of pregnant women diagnosed for preterm rupture of membranes Placenta is a key organ for pregnancy maintenance. One of its main functions is transport of compounds between mother and her fetus. The transplacental penetration is ensured due to membrane transporters that are present in the apical or basal side of trophoblast. Their expression level is affected by many physiological and pathological factors, among others it can be influenced by infection and inflamatory reaction. Inflammation is also one of the risk factors of preterm deliveries and it can be therefore assumed that these pathological states are accompanied by changes in expression of placental transporters. This study was performed using 51 placentas obtained from Faculty hospital in Hradec Králové from women who underwent preterm delivery and on 15 placentas delivered in term. The study employed quantitative RT-PCR approach.
