Tůmová, Marie ; Sochorová, Ludmila (referee) ; Štěpánová, Irena (advisor)
The main theme of the presented dissertation is life's work of Anna Regina Husová (1857 - 1945). The autor, ethnology student Marie Tůmová (Department of Ethnology Charles University in Prague), prowides a comprehensive overview of many aspects of her life which hasn't been mapped in the academic scene of the Czech republic yet. The author proposes first, that she was an outstanding personality in ethnology of our regional history. The work itself is divided into two main sections (life and life's work of Anna Regina Husová) and subsections which analysing the topic in a specific context: a brief introduction, structure, proposition, conclusion and full bibliography. There are also seven appendices (activities, friends, a brief look into her prose and poetry, illustrations, this English summary and CD disk), literature and sources. The methodological backround of the presented thesis was grounded in the professional qualifications, archive's sources, literature and recent literature as well. Cardinal method used in this thesis is a dense and holistic case study: a mohography. Although the thesis of graduate student Marie Tůmová, led by lecturer Irena Štěpánová, concerns many aspects selected regional personality exceeding 180 pages in lenght, it still raises several unanswered questions.

Pretransfusion testing, important red cells antibodies and transfusion in alloimunized recipients.
EBROVÁ, Jana
The topic of this thesis is to analyse the pretransfusion testing focusing on red cells transfusion, important red cell antibodies and transfusion in aloimmunised patients. This thesis is supposed to sum up the present knowledge of aloimmunisation in modern transfusion medicine, to provide a list of clinically significant antibodies and to determine the aloantibodies incidence in blood samples in pretransfusion testing in hospital Nemocnice České Budějovice, a.s. The thesis informs about the pretransfusion testing procedure, its requirements and recommendation for laboratory praxis. Furthermore, it describes immunohematologic laboratory methods that use pretransfusion examination. Then, it lists group systems with short description and clinically important irregular antibodies. The summary of actual state of the topic is concluded with the explanation of the term alloimunisation, causes of its emergence and recommendations for phenotypically suitable RBC transfusion preparations choice for aloimmunised patients. In the patient samples before transfusion of preparations containing RBC, we did the pretransfusion testing, i.e. blood type examination in AB0/RhD, screening of irregular antibodies against RBC antigens and compatibility test. If the antibody screening was positive, we identified them and eventually processed them through some additional RBC antigens test. After those tests, we used column (gel) agglutination and tube methods. In 7 % of the samples, the irregular antibodies were positive. More than 50 % of them came from women. Aloantibodies were found in 57,6 % of positive samples. Altogether, we identified 52 antibodies types: anti-E (23,08 %) > anti-D, -CW (13,46 %) > anti-c, -K, -Kpa (9,62 %) > anti-C, -Wra, -Jka, -S, -e, -Fyb, -P1 (1,92 %). The majority of identified aloantibodies came from the Rh (68 %) and Kell (19 %) system. Combinations were found in aloantibodies of Rh and/or Kell system. Acording to the results, polytransfused patients and fertile women can be recommended to use this ideal algorithm in the choice of transfusion preparations containing RBC: to test Rh and Kell system antibodies in the pretransfusion testing and to provide the recipients with transfusion preparations of maximal compliance of RBC antigens in above mentioned systems.

Survey of stage interpretacion of chorus in Sofokles's tragedy Oidipús Tyrannos on czech stage
Mrázek, Vít ; Topolová, Barbara (advisor) ; Sarkissian, Alena (referee)
The dissertation is trying to form a panoramic survey of interpretational possibilities of an ancient chorus in the process of the Czech theatre on the basis of the detailed analysis of chosen productions in Sofokles's tragedy Oidipús Tyrannos from the end of 19th century to the beginning of 21st century. Chronological summary of the interpretations follows evolutional trends from the original version of the choir of the old men that prevailed in the first half of the 20th century over the variants ancient people that began in the fifties of the last century to metaphorical transformation of choirs that was brought by some stage conceptions from the beginning of 21st century.

Epigenetic regulation of retroviruses.
Dobšová, Martina ; Trejbalová, Kateřina (advisor) ; Drda Morávková, Alena (referee)
The human genome is the site of integration of the retrovirus HIV-1, ERVWE1 (syncytin-1) is its stable part. Thus as an integral part of the genome, they are under influence of important genome based epigenetics regulations, such as DNA methylation, histone methylation and acetylation. Promoter DNA hypermethylation and histone deacetylation leads to establishment and maintenance of latent state of the HIV-1 virus and formation of the latent reservoir in the CD4+ memory T-cells. This process leads to severe problems during HIV-1 infection treatment by antiretroviral therapy (HAART), as the HIV-1 latent reservoir remains unaffected. Moreover DNA hypermethylation in the promoter of syncytin-1 directs its transcriptional silencing, which is important in tissue specific regulation of this fusogenic protein. In physiological conditions syncytin-1 expression is observed only in placental cells, where the DNA methylation of promoter is decreased. Higher expression level of syncytin-1 was also observed in several other tissues, such as testes, where it is tightly coupled with germ-cells tumors and syncytin-1 promoter hypomethylation. In conclusion, epigenetic regulation of retroviruses by DNA methylation and chromatin modifications highly influence regulation of their expression. Presented bachelor thesis...

Study of plasma protein binding of receptor-specific peptides
Křivková, Šárka ; Lázníček, Milan (advisor) ; Trejtnar, František (referee)
SUMMARY: Drug binding to plasma proteins is one of the primary pharmacological parameters. The binding rate has significant effects on drug distribution and drug elimination. In vitro investigation of plasma protein binding helps us foresee their destiny after its administration to a living organism. This dissertation focused on studying plasma protein binding of DOTA-NOC marking by 90 Y and 111 In in term of interspecies comparison. The binding was determined by methods of equilibrium dialysis at 37řC. Results showed binding of 90 Y-DOTA-NOC increases respectively: human < rabbit < bovine < rat plasm. Binding of 111 In-DOTA-NOC increases respectively: pork < human < rat < rabbit plasm. KEYWORDS: protein binding, DOTA-NOC, 111 In, 90 Y, interspecies comparison
