[cze][eng] Retrovirus HIV-1 se během svého životního cyklu integruje do hostitelského genomu, ERVWE1 (syncytin-1) je v lidském genomu přítomen stabilně. Jako součást genomu podléhají epigenetickým regulacím jako jsou methylace DNA a acetylace a methylace histonů. U HIV-1 je deacetylace histonů a hypermethylace DNA v promotorových oblastech spojena s ustavením latentního stavu viru. Latentní rezervoár HIV-1 v paměťových CD4+ T-buňkách je zásadním problémem při antiretrovirové terapii (HAART), protože tento rezervoár zůstává léčbou nedotčen. V případě syncytinu-1 vede hypermethylace DNA v promotorové oblasti k jeho transkripčnímu umlčení. To je důležité při tkáňově specifické regulaci, protože za fyziologických podmínek dochází k expresi syncytinu-1 pouze v placentě, kde je výrazně snížená methylace DNA v promotorové oblasti. V dalších tkáních, jako je tkáň varlat, je zvýšená exprese syncytinu-1 spojena s nádory buněk germinální linie a s hypomethylací jeho promotoru. Epigenetická regulace retrovirů pomocí methylace DNA a chromatinových modifikací se výrazně podílí na regulaci exprese těchto retrovirů. Součástí bakalářské práce je i praktická část, která se zabývá klonováním kódující sekvence syncytinu-1 do expresního plasmidu, následnou transfekcí do buněčné linie HCT 1- 3B- a měřením hladiny exprese v...The human genome is the site of integration of the retrovirus HIV-1, ERVWE1 (syncytin-1) is its stable part. Thus as an integral part of the genome, they are under influence of important genome based epigenetics regulations, such as DNA methylation, histone methylation and acetylation. Promoter DNA hypermethylation and histone deacetylation leads to establishment and maintenance of latent state of the HIV-1 virus and formation of the latent reservoir in the CD4+ memory T-cells. This process leads to severe problems during HIV-1 infection treatment by antiretroviral therapy (HAART), as the HIV-1 latent reservoir remains unaffected. Moreover DNA hypermethylation in the promoter of syncytin-1 directs its transcriptional silencing, which is important in tissue specific regulation of this fusogenic protein. In physiological conditions syncytin-1 expression is observed only in placental cells, where the DNA methylation of promoter is decreased. Higher expression level of syncytin-1 was also observed in several other tissues, such as testes, where it is tightly coupled with germ-cells tumors and syncytin-1 promoter hypomethylation. In conclusion, epigenetic regulation of retroviruses by DNA methylation and chromatin modifications highly influence regulation of their expression. Presented bachelor thesis...
