The Production and Ecological Potential of Pine Stands on Anthropogenic Soils of Post-Mining Sites
Dragoun, Lukáš ; Merganič, Ján (advisor) ; Konopka, Bohdan (referee)
Extraction of mineral resources constituted the development of industrial companies worldwide. But it has a major impact on the landscape and environment. Restoration of areas disturbed by mining is current issue especially on large surfaces after surface mining. In the Sokolov region, the forest reclamation was carried out on the Antonin spoil bank in the years 1969-1972, where forest stands of different species composition were established under the same conditions. The aim of this work is to compare the mixed pine stands on anthropogenic soils more than 40 years after their foundation, to assess their suitability for reclamation and to describe the influence of admixed deciduous trees on the production and ecological potential of the site. On the plots the basic mensurational and production quantities of the stand were measured. The dendrochronological analysis was performed and the diameter increment was detected for the main species Scots pine. Based on the diameter increment, the tolerance analysis was carried out for the highest risk factor on these sites - drought. The most appropriate in local conditions seems to be a structure of pine stands with an admixture of lime trees, which is characterized by the highest production, soil formation and ecological potential. The work also points to two important factors that significantly influenced the silviculture on the Antonin spoil bank. It is the neglect of stand tending, and thus significant disruption in stand stability, and limiting intake of water, causing significant declines in increment regardless of an admixture or character of the stands.

Endoparasites of horses
Scháňková, Štěpánka ; Langrová, Iva (advisor) ; Bejček, Vladimír (referee)
Nine ponies were randomly allocated to one of three groups (A, B, C). Gradually, these groups of ponies were experimentally infected with third stage cyathostome larvae. Each pony was infected with 30,000 infective cyathostome larvae stored 4 weeks under the natural environmental conditions of the Czech Republic. These animals were examined post--mortem for small strongyle infections from October 2011 to January 2012. Seventeen species of the following Cyathostominae genera were identified: Coronocyclus, Cyathostomum,Cylicocyclus, Cylicostephanus, Gyalocephalus and Petrovinema. Of the 17 species of Cyathostominae recovered, Cyathostomum catinatum, Cyathostomum alveatum and Cylicocyclus nassatus were the most prevalent (100 %). From the entire population of Cyathostominae, 65,6 % were located in the caecum and only 34,4 % in the colon. Only 1,736 (24,6 %) females and 5,329 (75,4 %) males were observed. In the first and second phase (group A, B) of the experiment, 98.57 % and 95.48 % of adult nematodes were found, respectively. However, only 29,07% of adults were found in the third phase (group C) of experiment, and the proportion of arrested larvae (EL3) was 67.73%. All ponies were treated orally with ivermectin at a dosage of 400 ug /kg. However, adult Oxyuris equi were present in six horses. This is the first European study to demonstrate anthelmintic resistence in Oxyuris equi to macrocyclic lactones.

The importance of legumes in the human diet
Střelková, Tereza ; Hejtmánková, Alena (advisor) ; Martina, Martina (referee)
Legumes are part of human diet since Early Years and they have important role. In Czech Republic the most commonly consumed legumes are beans, peas and lens and in recent years even cicer (commonly chickenpea) and soya. Yearly consumption of legumes in human diet in Czech Republic is about 2,5 kg per person, which is less than in other countries but the consumption is slightly increasing. Being very good source of protein, legumes as source of non-complete protein should be combined with other sources of protein preferably those based on cereal. Legumes are also source of carbohydrates, from which big part is favourable mix of starch and fiber. Present minerals are e.g. calcium, phosphorus, magnesium, iron and zinc. From vitamins mostly B complex can be found. Legumes, especially soya, also contain phytoestrogens and other biologically active substances. Phytoestrogens emulate behaviour of human estrogenes and raise interest in areas of research. There are series of scientific foundations for their potential to be used as part of a treatment for worldwide spread disease like cancer or diabetes, or other common health difficulties e.g. post-menopausal symptoms. Legumes are often refused in human diet, because of antinutrients they contain. Those are mostly oligosaccharides that are fermented in large intestine which causes flatulence and indigestion. Antinutrients are e.g. enzyme inhibitors or phytic acid. Antinutrients can be removed by variety of culinary treatments. The easiest are soaking or boiling. The harder, yet very effective treatments, mostly because of time consumptions are e.g. germination or fermentation, or because of technological demands - extrusion. However, heat treatment can degrade proteins and thus lower their usefulness and also reduce biological activity. Biologically active substances are nowadays intensively researched and studies bring new perspective advantages of consuming legumes.

Molecular mechanism of hydrogen sulfide action during meiotic maturation of porcine oocytes
Veselá, Andrea ; Hošková, Kristýna (advisor) ; Tomáš, Tomáš (referee)
At present reproductive biotechnology methods are on the rise, but their development and application in the broader management of reproduction is dependent on obtaining a sufficient number of quality oocytes cultured in vitro. The prerequisite for this requirement is the creation of the optimal conditions in the course of culturing oocytes. Understanding and knowledge of the processes that occur in oocyte during maturation is an important and necessary condition for optimizing the process of culturing oocytes in vitro and gaining a sufficient number of good quality oocytes in metaphase II of meiotic division. A large number of mechanisms that affect and control oocyte maturation are known, however it cannot be claimed that this process has been fully explained and studied. One factor which has a potential role in the regulation of meiotic maturation of oocytes is gasotransmitter hydrogen sulfide (H2S), a critical signaling molecule of endogenous origin. The study of H2S led to the hypothesis that H2S actively influences the course of meiotic maturation of pig oocytes by regulating key signaling cascades. The aim of this work was to determine the involvement of H2S in the regulation of the MEK1-MAPK signaling cascade, responsible for the initiation and progress of the meiotic maturation of oocytes and the MEK1-PARP-1 cascade as signaling that supports cell viability. For this purpose, pig oocytes cultured in modified media were used, supplemented with a specific combination of enzyme inhibitors (3Ki) or in a culture medium with donor H2S. The ocytes were then subjected to immunocytochemistry staining, fluorescence microscopy and image analysis. The results show that H2S is involved in the regulation of meiotic maturation. It confirmed the hypothesis of the endogenous production of H2S in the course of the meiotic maturation of pig oocytes and the influence of the MAPK signaling cascade. Based on the results, it is however likely that the MEK1-PARP-1 signaling cascade is not affected by H2S, unlike MAPK signaling, comprising the mentioned MEK1 as superior kinase. MAPK kinase activity is significantly lower in oocytes after treatment 3Ki. Further experiments are for a detailed understanding of these regulatory pathways and for the proper verification of the mechanism of the effects of H2S necessary, in particular for a full understanding of the target control factors by the post-translational modification of S-sulfhydration.

Economic Development of Czechoslovakia in the Years 1945-1948
Pazdera, Lukáš ; Skřivan, Aleš (advisor) ; Szobi, Pavel (referee)
The diploma thesis is focused on the economic development of Czechoslovakia after 1945 up to the political takeover in 1948, when communists finally assumed power. The work offers the view of Czech economy influenced by the experience of the Great Depression in the 1930s, occupation and the transformation of economic thinking in Europe. The main concern is to answer the questions to what extend the post-war reconstruction was successful, what the political conditions of the changes in our economy were and which economic results they produced. The land reform, final solution of German question by the expulsion of Germans, nationalisation of industry, codification of two-year economic plan and refusal of the Marshall plan are considered to be the key points. The allience with the Soviet Union played an important role too. The main conclusion of the work is that, in Czechoslovakia, the successful transformation from war economy to a mixed economy, in which the main role of the state was supplemented by a private sector, was accomplished. The existence of different alternatives of development, which could not be realized because of the increasing influence of communists in the government, is considered to be the key factor for the economic development in the years 1945-1948.

Þorr's voyage to Geirrøðr
Kozák, Jan ; Antalík, Dalibor (referee) ; Starý, Jiří (advisor)
The thesis analyzes eight versions of a narrative of thórr's journey to the dwelling place of thurs (giant) Geirrodr. These variant versions are preserved in several distinct genres and come down from different ages - the oldest is a scaldic poem from the time before the christianization of Scandinavia (10th century) whereas the other versions are written down by Christian Icelander Snorri Sturluson and Danish chronicler Saxo Grammaticus (both 13th c.). The youngest version is a fairy-tale short story from 15th c. The fact of diachronic and genre variation is used to track and analyze the changes of an originally religious narrative after the christianization, i.e. after the dissolution of the original pagan symbolic universe into the medieval (or even post-medieval) one. Although the basic pattern of the story and the names of the protagonists are preserved quite well (in respect to the time span and indirect transmission), the meaning and genre setting of the story is significantly altered: in the case of Saxo Grammaticus into euhemerized pseudohistory and in the case of 15th century short story into fairy-tale loosely connected to historical king Ólafr Tryggvason. This observation illustrate and confirm the process of demythization of pagan myths and show two genres where those myths are to be "stored"...

Smejtek, Lubor ; Bouzek, Jan (advisor) ; Podborský, Vladimír (referee) ; Jiráň, Luboš (referee)
The work is a general analysis and evaluation of the results of an extensive rescue excavation (2.939 features, out of which 1.470 were post and column holes), that was carried out by the Archaia company at the Kněževes site (Praha-západ district) in the year 1998. In an area of more than 10 ha, evidence of settlement in several prehistoric and historic periods was recorded (Neolithic, Eneolithic, Hallstatt period, Early La Tène, Hillfort period), however, it was particularly the Bronze Age, where the majority of the finds belong. An analysis from different viewpoints, supplemented with selective descriptive and visual documentation, represents the focal point of the submitted treatise. In the introductory part, there are described the progress and methods of excavation, the geomorphologic conditions, the subsoil and soil coverage on the site, as well as the expected vegetation in the area of interest on the basis of scientific analyses (macroremains, charcoal). Attention is paid also to the history of archaeological research within the cadastre of the current village of Kněževes and the neighbouring Kněžívka. From the beginning of the Bronze Age (the early phase of the Únětice culture) date 24 inhumation graves equipped with pottery and occasionally also with copper/bronze earrings. 16 features of...
