Neural and Genetic Basis of Epilepsy

This thesis describes neural and genetic basis of epilepsy, addresses the distribution of epileptic sezuries, which genes affect epilepsy and the connection between epilepsy and the nervous system. The thesis is based on data collection from scientific books, partly from data published by hospitals. Its focus is getting from the general expressions to the cellular level of the problem, which is necessary to clarify the epileptic principles. It also deals with the genetic nature of epilepsy, as it is with the inheritance of the disease and deals in more detail with early infantile epileptic. The beginning of work is about basic information of epilepsy. Next, it looks into the mechanisms that influence the emergence of epilepsy. Continues to look at the structures in the brain associated with epilepsy. And at the end, it deels with genetic nature of epilepsy. A brochure was created for this work, where basic information about epilepsy can be found, first aid for an epileptic seizure and early infantile epileptic encephalopathy is discussed in paragraphs.

ŠIMKOVÁ, D. Reaction of Horse´s Neuroendocrine System to Stress. Mendel University in Brno, 2017. Bachelor thesis. The bachelor thesis deals with the issue of stress in connection with neuroendocrine systems. The work is focused on the comparison and cooperation of these two systems depending on the stress. The first part describes general characteristics of the endocrine system and hormones, a general description of the nervous system and the definition and view of stress. In the next part are given the specific organs of the given systems and their main function. End of the chapter is devoted to the pathological processes of both systems. In conclusion the effect of stress on these systems and the impact on possible desorders and diseases of the neuroendocrine system are summarized.
Evolution of the air navigation services in the European open sky concept

This bachelor thesis deals with technologies and methods provided by air navigation services. This work aimed to create a cross-section of the history of the development of such technologies and methods and to present new and developed technologies and methods designed for European airspace. The result is an overview that serves as study material for understanding the problems of insufficient airspace capacity and for understanding the directions taken by the EUROCONTROL.
Magnetic resonance imaging of central nervous system lymphoma.

Background: The aim of this study was to describe the morphological signs of the central nervous system lymphoma (CNSL) in magnetic resonance imaging (MRI). We compared morphological characteristics of primary CNSL (PCNSL) and secondary CNSL (SCNSL) and also of PCNSL and glioblastoma (GBM). Methods: We included 64 patients with PCNSL (ten of them were immunocompromised), 21 patients with SCNSL and 54 patients with GBM. The diagnosis was confirmed histologically in all patients. We evaluated morphological signs on the first MRI examination. Additionally, in patients with PCNSL, we evaluated the development of the disease on follow-up examination before histological confirmation of the diagnosis, if available. Results: In most patients with PCNSL (62.5%) the tumor was localized supratentorially and presented as multiple lesions (53.1%) or as a diffuse infiltrative lesion (23.4%). In 87.5% of the patients the lesions reached the brain surface. Infiltration of ependyma was seen in 56.3%, infiltration of meninges in 39.1% and infiltration of cranial nerves in 48.5% of patients. The main goal of this bachelor thesis was to characterize changes of adipokinetic hormone (AKH) production in the brain and corpora cardiaca of the firebug Pyrrhocoris apterus during the infection caused by entomopathogenic nematode Steinernema carpocapsae. The effects of the pathogen were investigated by determination of AKH level in the CNS using competitive ELISA test and immunohistochemical localization with a specific antibody against the P. apterus AKH (Pyrap-AKH). The results confirmed presence of majority of AKH in corpora cardiaca and smaller amount also in brain. The infection increased AKH production in both monitored organs, however, in brain the elevation was more intensive than that in corpora cardiaca.
Production of cytokines in mice infected with bird schistosome Trichobilharzia regenti

The neuropathogenic trematode Trichobilharzia regenti (Schistosomatidae) infects the central nervous system of birds and mammals. During its migration through the spinal cord, the parasite causes tissue damage and triggers inflammation which is likely responsible for the elimination of the parasite. In this thesis, the proinflammatory cytokines IL-1β and IL-17 were detected by immunohistochemistry in the affected spinal cord of C57BL/6J mice during the infection. Additionally, IL-4 and IL-6, participating in the regulation of the inflammation and tissue repair, respectively, were noticed. Astrocytes, microglia and other, yet unidentified cells, produced these cytokines. Furthermore, splenic T-lymphocytes were phenotyped by flow cytometry to characterize peripheral immune response. At the peak of nervous tissue inflammation, mixed (Th1/Th2) cellular immune response was observed. Taken together, this thesis extended the knowledge of cytokine immune response of mice infected with T. regenti and also confirmed that inflammation in the nervous tissue influences the polarization of peripheral immune response. The effect of the canonical Wnt singalling pathway on the differentiation of polydendrocytes after ischemic brain injury

Polydendrocytes, or NG2 glia, are fourth type of glial cells in mammal central nervous system. In the adult brain, NG2 glia represent important cell type with respect to their role in gliogenesis and nervous tissue regeneration following injury. Ligands from the Wingless/Int (Wnt) family play key role in proliferation and differentiation of NG2 glia and they can also influence regeneration of nervous tissue after ischemia. The aim of this thesis was to elucidate the role of NG2 glia in neurogenesis and gliogenesis following ischemic brain injury and investigate the impact of Wnt signalling on the reaction of NG2 glia to this type of injury. To fulfil these aims, transgenic mouse strains with tamoxifen-inducible recombination, that enabled simultaneous expression of red fluorescent dye and either activation or inhibition of the Wnt signalling pathway in NG2 glia, were employed. To induce ischemic injury, middle cerebral artery occlusion model was used. Changes in differentiation and electrophysiological properties of NG2 glia were analysed using patch-clamp technique. Activation of the Wnt signalling pathway under physiological conditions and 7 days after ischemic injury led to increased differentiation of NG2 glia toward astrocytes, while 3 days after ischemic injury activation of this signalling...

During inflammation, dying or temporarily damaged cells release ATP, as a danger-associated signal molecule, that contributes to the induction of astrogliosis and promotes clearance of the debris by immune cells such as microglia. Adenosine that results from ATP metabolism also stimulates astrogliosis. However, the effects of adenosine on astrogliosis may be more complex, since it also modulates microglia phenotype and microglia have been shown to prevent excessive astroglial proliferation mediated by nucleotides. In this context, ATP and adenosine are assumed as relevant signalling molecules in the control of astrogliosis and its modulation by microglia. However, it is still unknown whether and how microglia modulate adenosine-mediated astrogliosis.
