Influence of employment politics on situation of people at the age of 50+ on the labour market: case of the Vsetín district

The thesis: "Influence of employment politics on situation of people at the age of 50 + on the labour market: Case study of the Vsetín district" deals with problems of older people's position on the labour market in the Czech Republic focusing on the Vsetín district and influence of employment policy on this position. This thesis characterizes the group of people at the age of 50 and more as labour power and deals with their strategies on the labour market. It also deals with employment policy of the European Union related to the group of people at the age of 50 and more on the labour market and analyses strategic documents of the Czech Republic solving these problems in detail. Furthermore this thesis focuses on the employment policy of the Czech Republic, mainly on analysis of functions, possibilities, limits and influence of chosen tools of the active employment policy on the lowering of the number of the unemployed people who belong to the above mentioned age-group on the labour market. It evaluates these tools in detail and shows which of these examined tools have the biggest influence on the lowering of the number of the unemployed older people. The corporate social responsibility is a voluntary concept of firm's embrace of responsibility for the impact of its activities on the environment and society. The corporate social responsibility is the brand and guarantee that firm runs its activities not only within the law and ethical standards, but goes far beyond the law and moral values. Firm demonstrates its social responsibility through engagement in social issues, substitute for the state in support of underprivileged or disadvantaged groups, but also develop activities which have positive impacts on the environment, advocate and meet ethical business principles, take care of their employees, to distance themselves of corruption and bribery, etc. This thesis deals with the objective introduction of corporate social responsibility concept from its origin to contemporary approaches. The individual types of domain names are associated with the effects resulting from the domain operation and other intangible assets that usually cooperate with the domain as a whole. Conclusions of the thesis are demonstrated in the analysis of intangible assets of the company ADSL s.r.o.

Fiscal Policy Rules

Předkládaná práce zachycuje komplexnějším způsobem problematiku fiskálních pravidel, včetně vybraných v realitě používaných typů, s cílem přispět na základě analýzy současného stavu fiskálních rámců v Evropské unii a v České republice k diskusi o volbě vhodného fiskálního pravidla pro české veřejné finance a českou ekonomiku. Stěžejní typy fiskálních pravidel byly komparovány jednak vzájemně, jakožto i ve vztahu k osmi fundamentálním atributům ideálního fiskálního pravidla. Pozornost byla rovněž věnována chování pravidel v rámci hospodářského cyklu. Ze srovnávaných pravidel bylo celkově nejlépe hodnoceno pravidlo výdajové. Jelikož primárním fiskálním rámcem po české veřejné finance je Pakt stability a růstu, byla spolu s Maastrichtskými fiskálními kritérii hodnocena jeho účinnost. Metodou pro hodnocení byla vybrána panelová regresní technika nejmenších čtverců s fixními efekty. Výsledek ekonometrické verifikace potom naznačil, že pravidlo Paktu účinné skutečně bylo, avšak že je funkční pouze tehdy, je-li skutečně vynutitelné (citelná ztráta z nečlenství v ?euro klubu?). Další část práce se teoreticky věnujeme otázce koordinace politiky fiskální a měnové. Analýza se ubírá směrem malých otevřených ekonomik cílujících míru inflace s deficitním omezením, čili k problematice relevantní pro českou ekonomiku. Sestavujeme tak teoretický model, kterým ukazujeme, jak tyto dva stěžejní elementy hospodářské politiky reagují na chování druhé autority sledujíce svůj zájem vyjádřený tzv. ztrátovou funkcí příslušné autority a kdy je možné dosáhnout bodu stálého stavu v prostoru nástrojů měnové a fiskální politiky. Řešíme rovněž otázku koordinace těchto dvou makropolitik po vstupu české ekonomiky do společného měnového prostoru a pomocí dedukce pak z nastolených premis modelu usuzujeme na důležitost fiskálního pravidla v malých ekonomikách pro jejich ?zdravý? fiskální vývoj. Na závěr práce syntetizujeme získané poznatky a vyvozujeme patřičné závěry pro českou hospodářskou politiku, které ústí do návrhu vlastního, resp. modifikace, fiskálního pravidla vhodného pro české veřejné finance v kontextu evropského Paktu stability a růstu.

Functional analysis of syntaxin 16 phosphorylation using yeast as a model

4 Abstract Mechanism of fusion of intracellular membranes in eukaryotic cells involves several protein families including soluble N-ethylmaleimide-sensitive-factor attachment protein receptor (SNARE) proteins and Sec1/Munc-18 related proteins (SM proteins). It is known that the transport is evolutionary conserved from yeast to man. Therefore for facilitating of the research, we can use simple eukaryotes Saccharomyces cerevisiae. Mammalian SNARE protein syntaxin 16 has a yeast homologue Tlg2p which is used in this study as a model for studying affects of phosphorylation to the syntaxin 16 function. Also their binding partners, SM proteins mVps45p (mammalian) and yeast Vps45p are homologous. Phosphorylation of SNARE proteins is known as a possible way of regulation of membrane fusion. Abolishment of one of the putative phosphorylation sites in Tlg2p protein, serine 90 leads to dominant effects on the exocytic and endocytic pathways. The work presented in this study shows some phenotypes of mutants based on this phosphorylation site of protein Tlg2p. Those mutants are S90A (cannot be phosphorylated) and S90D (phosphomimetic - acid carboxyl group mimics phosphate group). Pieta from Všeměřice and Lamentation from Korkusova Huť - the analysis of the works and changes in the iconography

This Bachelor Diploma deals with the motif of Piety and Lamentation. The thesis indicates the understanding of those two motifs from the iconographic point of view, follows their historical development during the Gothic period and examines several Christian writings, that could have influenced the establishment of an iconographic type of the motifs. The main chapters focus on the Pieta from Všeměřice form 1430 - 1440 and Lamentation from Korkusova Huť from 1510 - 1520. These chapters discuss the history of both works, their original placements, functions and analogies in their style. One of the important aspects is also an indication of the possibility of comparing the works of art with several other pieces of both Czech and foreign Gothic art of sculpture, that might become a guideline in determining the direct impacts on the final shapes of the works as well as their authors. The conception of Pieta and Lamentation undergoes a grand change during the Gothic period. The formerly widespread and famous motif of Piety withdraws as the motif of Lamentation overtakes its place. Therefore, another task of this thesis is to describe and explain this change of the iconographic motif of art. The thesis further describes and explains the main causes, that led to the withdrawal of the motif of Piety and the massive expansion of the motif of Lamentation.

Screening for the HCV IRES interacting proteins

Hepatitis C virus (HCV) is a worldwide spread pathogen infecting up to 3 % of the human population. Nowadays, research of new drugs against this virus is focused on the individual steps in its life cycle, including the translation initiation. In the case of HCV translation initiation is dependent on the internal ribosome entry site (IRES). Besides of components of the translational machinery also other components of the cell, so called IRES trans-acting factors (ITAF), contribute to its proper progress. This work continues in previous research of our laboratory focused on searching for new ITAF. In order to search for potential ITAF increasing HCV IRES activity new recombinant plasmid vectors and reference strains were prepared and selection conditions of the selection system were optimized. The differences in the growth characteristics of the reference strains were analyzed and quantified under selective and non-selective conditions. A set of pilot high efficiency transformations of the yeast strain pJ69-4A carrying bicistronic construct with HCV IRES were conducted using human expression cDNA library in order to optimize the efficiency of transformation and selection conditions and to attempt to identify new ITAF. Body Decoration with the Focus on Jewellery

This thesis focuses on the issue of body decoration, its meaning and function in culture and society. Main interest of this thesis is aimed on jewellery and its field of the visual state. Thesis divides main issue into following points: selection of the main functions of each kind of jewellery, material and technology, world point of view of cultures, main principles of its order, and main thematic fields. At the end of the thesis is briefly sketched changing approach to the genuine author jewellery. Thesis includes examples based on jewellery history accompanied by pictures and drawings. European tradition is stressed throughout all thesis. The thesis aims on the issue of cultural and social background and its influence on visual state of jewellery. The didactical part includes description of the possibilities in the field of pedagogical activity in the field described in the main part of paper.

This text is a part of my years long art work. Industrial architecture and mechanical parts, fascination to functions of mechanisms and the aesthetics of industry are for years my hobby. These phenomens became fundamental for my art work. In this work I introduce my creative basis. Their main themes are fundamental changings of human's development. Subsequently I have searched for similar contents used in the artistic world. Particulary I thought about the risks and dangers which the new technical age hides from our site. Technologie from a lot of points of view abbreviates our lives but on the other hand it is also a risk for people who do not esteem the values of our environment, nature and do not think much of their own ephemerality. The didactic part of my work I spent upon playfulness which is for all children natural. Mechanisms, constructions and technologies enable the creation of an abstract world and fantastic beings. The topic of "Industries" offers a rich scale of inspiring ideas for work with children. At the same time we save the concepts of how to combine the cultural and nature's world. It is necessary to educate all in the spirit of living, staying playfull and sensetive at the same time.
