Malechová, Veronika ; Šulová, Lenka (advisor) ; Morávková Krejčová, Lenka (referee)
This theses deals with diagnostic of speech development of pre-school children. Its focus is on lexical-semantic language level. The goal of my work is to trace and map diagnostic possibilities in the lexical- semantic area of of pre-school children and to introduce the international PPVT test , considering its adaptation into czech language environment. Theses consists of two parts - the theoretical and empiric. Theoretical part is concern with the most important theories and personalities in the speech development and the process of experiencing language, characteristics of pre-school period, ontogenesis of speech and the ways to evaluate vocabulary and semantics in Czech and overseas. The empiric part examines the usages of PPVT in czech environment. The survey was realised by quantitative research, using datas from 236 tested individuals. The PPVT was adapted into czech language and given to preschool children, whose results were compared with the results of american children. The outcomes of the empiric part could be a solution for future research in the area, with the purpose of adapting a testing- language instrument into czech language and widening the limited possibilities in the field of language and speech development evaluation. Keywords: ontogenesis of speech, pre-school period,...

Titanium Dioxide - Phosphonate Assemblies as Medical Nanoprobes Ivan Řehoř PhD. Thesis Abstract: Multimodal imaging-therapeutic nanoprobe TiO2@RhdGd was prepared and successfully used for in- vitro and in-vivo cell tracking as well as for killing of cancer cells in-vitro. TiO2 nanoparticles, 12 nm in diameter, were used as a core for phosphonic acid modified functionalities, responsible for contrast in MRI and optical imaging. The phosphonic acid derivatives, used for surface modification, allows for grafting extraordinarily high loads of irreversibly adsorbed molecules of both types in one step. The prepared probe shows very high 1 H r1 relaxivity value as well as relaxivity density value, both crucial parameters for its use in MRI. The presence of fluorescent dye in its structure allows for its visualization by means of fluorescence microscopy. The applicability of the probe was studied, using three living systems - mesenchymal stem cells, cancer HeLa cells and T-lymphocytes. The probe did not exhibit toxicity in any of these systems and its long time storage in a lysosomal compartment was confirmed. Labeled cells were successfully visualized in-vitro by means of fluorescence microscopy and MRI. Consequent visualization of labeled cells in-vivo by means of fluorescence microscopy was also achieved....
Intravital diagnostics of neurodegenerative diseases
Baranová, Soňa ; Holada, Karel (advisor) ; Kaňovský, Petr (referee) ; Žáková, Dana (referee)
Prionopathies, also called transmissible spongiform encephalopathies (TSE) and synucleinopathies are neurodegenerative diseases that are associated with the accumulation of misfolded proteins (prion and α-synuclein) mostly in the central nervous system. To this day, early and definite diagnosis remains unavailable during the patient's lifetime, mainly due to the absence of reliable biomarker which makes clinical diagnosis more challenging. Therefore, the gold standard in diagnostics remains direct post-mortem evaluation of misfolded proteins within brain tissue by western blot and immunohistochemistry. In the recent years, seeding amplification assays (SAAs) like Real-Time Quacking-Induced Conversion (RT-QuIC) emerged for ultra-sensitive ante-mortem diagnosis of neurodegenerative diseases. SAAs exploit ability of pathological misfolded proteins present in patient's samples to change the conformation and initiate aggregation of native recombinant protein substrate by prion-like seeding mechanism. In the presented dissertation thesis, we exploited second-generation RT-QuIC assay (55řC, 700 rpm, cycles of 1 min double-orbital shaking and 1 min incubation) utilizing recombinant hamster shortened prion protein (rHAPrP90-231) to evaluate prion seeding activity in post-mortem TSE (n=38) and non-TSE (n=30)...

Fišerová, Tereza ; Malý, Tomáš (advisor) ; Bujnovský, David (referee)
Title: Objectification and comparison of lower limb strength characteristics in selected sports Objectives: The main objective of this study was to objectify and compare selected indicators of lower limb strength in players of team sports games (basketball, football, floorball) of category U19. The secondary objective was to determine the relationship (degree of correlation) between knee joint flexion/extension strength parameters and vertical jump parameters. Methods: The group consisted of 56 probands from three groups of different sports: basketball, floorball and football, the highest competitions in the Czech Republic in the youth category (16-18 years). An isokinetic dynamometer was used to investigate the flexion and extension forces in the knee joint at different angular velocities of movement (60ř/s and 300ř/s). Force plates were used to determine inversion dynamics parameters or lower limb strength during two types of vertical jump. Unassisted arm jump and squat jump were tested. Results: This study found different levels of lower limb strength adaptation in young athletes between the sports of basketball, soccer and floorball. Movement in sports such as soccer and basketball appear to achieve significantly higher absolute and explosive lower limb strength demands than in floorball. These...

Hodačová, Alžběta ; Vařeková, Jitka (advisor) ; Majorová, Simona (referee)
Title: Sensorimotor difficulties in children with speech impairments Objectives: The main aim of the thesis is to assess sensorimotor functions of children with speech disorders using standardized test batteries. Methods: 27 children were part of the study (21 boys, 6 girls), all with speech disorders from an elementary school in the age range 11-15 (12,4 years±1,05). Motor skills were tested with data collection testing batteries MABC-2, Unifittest 6-60. Furthermore, sensory functions were tested using the Nottingham Sensory Assessment. Probands were also provided with a supplementary questionnaire to gather additional information. In the next step a data matrix was created in Microsoft Excel where all data were input. Data analysis was conducted using programs Microsoft Excel and Jamovi. Results: Children with speech disorders showed higher prevalence of motor difficulties compared to Czech norms. According to the overall STS score in MABC-2 test, 63 % (17) were in the range of significant motor difficulties, which is 13 times higher than the norms. A statistically significant relationship between the Unifittest and MABC-2 was proven. In addition, a positive correlation was also confirmed between the BAL component of the MABC-2 and the long jump task in the Unifittest 6-60. Among the sensory...

Mezlík, Vojtěch ; Heřmánková, Věra (referee) ; Anton, Ondřej (advisor)
The subject of bachelor’s thesis is a structural-technical survey of a reinforced concrete frame structure. The theoretical part deals with the inspection of reinforced concrete structures, destructive and non-destructive diagnostic methods for the diagnosis of reinforced concrete structures, and the Czechoslovak architect Jaroslav Josef Polívka. The practical part conducts a structural-technical survey, focusing on the diagnosis of the reinforcement of selected structural elements and the determination of properties of the concrete. The reinforcement was determined using electromagnetic indicator Proceq Profometer 630 and the ground penetrating radar Hilti PS 1000 and verified by chiseled probes. The concrete strength class in the structure was estimated from core drills. In the conclusion, the condition of the structure is evaluated, and measures are proposed.
Modeling and simulation of PLC-BPL communication of smart metering in Network Simulator 3
Baranyk, Matěj ; Rusz, Martin (referee) ; Benešl, Lukáš (advisor)
n recent years BPL technology is becoming more used. Despite the technology not needing any additional cable installation it's important to make sure that using the technology itself makes sense or what the needed requirements will be. Due to this reason we opt to simulate real life scenarios and compare them with outputs from network simulator ns-3. This way we're able to recognize the usefulness of our efforts even before we make any plans to instal anything, which saves time and money. Furthermore we can use the same simulator for diagnostics of already installed BPL routes. Furthermore a laboratory measurement shall be conducted in order to increase dataset sizes and to have more data that can be compared with simulation outputs.
