Developement and validation of method for ferulic acid isolation from wheat bran

This master thesis deals with an optimization of processes of isolation of ferulic acid from wheat bran. Process of isolation based on alkaline hydrolysis from the substance and its adsorption was tested. In the theoretical part were described wheat bran, adsorption proces and used methods. For adsorption were tested five types of adsorbents. Especially these adsorbents: activation carbon (2,53 ± 1,16 mg.l-1), Amberlyst A-21 (105,73 ± 11,87 mg.l-1, Amberlit XAD-16 (241,55 ± 10,42 mg.l-1), Amberlit IRA-900Cl (5,90 ± 0,68 mg.l-1) and Amberlit IRA-96 (189,16 ± 6,49 mg.l-1). As the most efficient adsorbent was determine Amberlit XAD-16. The whole process has underwent detailed optimization in order to maximize the purity of the final product. The isolate was characterized by HPLC and FTIR techniques for ferulic acid and related phenolic acids. Based on FTIR analysis was discovered that the isolate was contamined by proteins. A purification procedure based on deproteination of the raw material before the hydrolysis itself was proposed. As the ultimate solution that eliminated the presence of proteins in the isolate was determine ultrafiltration with capture of molecules with Mw > 10 KDa. Ferulic acid yield from one kilo of wheat bran was 1,00 ± 0,22 g.
In vitro biodegradation of biopolyesters exposed to synthetic gastric juice

This bachelor’s thesis is focused on the study of in vitro biodegradation of biopolyesters exposed synthetic gastric juice and phosphate buffer saline with lipase for 81 days. The theoretical part summarises the necessary information about characteristics of biodegradable biopolyesters, deals with their possible applications in medicine. The experimental part deals with the biodegradation of prepared biopolyesters films. in this work, poly(lactic acid), amorphous poly(hydroxy alkanoates), poly(3-hydroxybutyrate), poly(butylene adipate terephthalate), poly(3-hydroxybutyrate-co-3-hydroxyvalerate), poly(3-hydroxybutyrate-co-3-hydroxyhexanoate), filament poly(lactic acid) and filament poly(lactic acid)/poly(hydroxybutyrate) were used for the experiment. The films were prepared by dissolving of the biopolyesters in hot chloroform. The dissolving agent was evaporated, and prepared films were dried. Prepared films had different properties depending on the used biopolyesters. Their appearance was formed transparent to milky cloudy. Also, trey differed in their plasticity. During biodegradation, the morphology changed, and the samples gained higher fragility. The effect of biodegradation was monitored by changes in 1) molecular weights (SEC-MALLS), 2) chemical structure (FTIR) and 3) weight loss changes (gravimetrically). The analysed biopolyesters were demonstrably susceptible to biodegradation in simulated body conditions. The degree of biodegradation depends on the type of biopolyester. The most significant weight loss was recorded for the sample of amorphous polyhydroxyalkanoate (PHA). The PHA´original weight was reduced about 84.28 ± 0.03 % after the 81 days of biodegradation in the phosphate-buffered saline with lipase.

Acidic Hydrolysis of Waste Feather Biomass - Processes Scale-Up.

The paper present novel data on acidic hydrolysis of waste chicken feathersin presence of water solution containing carboxylic acid catalyst with the pKa value lower than 4, instirred batch reactors, in the temperature range from 114°Cto 150°C and at the pertinent vapour pressure of reaction mixture.The process was scaled-up from laboratory bench scale to pilot plant dimensions using batch high pressure autoclaves. Process conditions were applied according to patent application1. Knowledge of kinetic aspects of the featherhydrolysis and process parameters operation window are inevitable for asuccessful process scale up2,3.

UTILIZATION OF FOOD PROCESSING WASTE FOR LACTIC ACID AND ETHANOL PRODUCTION

The doctoral thesis is focused on the microbial production of lactic acid and ethanol using food processing waste as substrate. Coffee processing waste (spent coffee grounds), wine production waste (grape pomace) and orange processing waste (orange peel) were chosen as substrates for experiments. The theoretical part is dedicated to summarizing current knowledge about waste from food production and possibilities of its processing. It also deals with selected metabolites (lactic acid, ethanol) to which these wastes can be used. Part of the experiments was focused on the characterization and optimization of hydrolysis to maximize the amount of fermentable saccharides. Different combinations of chemical, physical and enzymatic hydrolysis of selected substrates have been tested. Subsequently, a suitable strain for lactic acid and ethanol production was searched for. In the case of lactic acid production, 7 bacterial strains were selected (Lactobacillus casei CCM 4798, Bacillus coagulans CCM 2013, Bacillus coagulans CCM 2658, Lactobacillus rhamnosus CCM 1825T, Lactobacillus delbruckii subsp. bulgaricus CCM 7190, Lactobacillus plantarum CCM 7039T, Streptococcus thermophilus CCM 4757). These strains were first cultivated on the synthetic media containing different kind of saccharides. Afterward, the cultivation on the waste biomass hydrolysates were tested. In the case of ethanol production, 2 yeast strains kmeny (S. cerevisiae CNCTC 6646 a S. cerevisiae CNCTC 6651) were cultivated on hydrolysates of individual waste substrates. Subsequently, the experiments focused on the production of lactic acid and ethanol on hydrolysates of waste biomass in bioreactor were done. The last part of this doctoral thesis deals with the microaerobic pretreatment of lignocellulosic biomass to increase the production of organic acids during the acetogenic phase of anaerobic digestion.
Characterization of different bran species in terms of ferulic acid content

This bachelor’s thesis works with an alkaline extraction of a ferulic acid from different kinds of cereal bran. Another researched issue was extraction of nutritionally interesting elements extracted from bran and from hydrolysed waste. Those extracts has possible usage as a mineral fertilizer for plants. In the theoretical part, this thesis deals with problematice of cereals, composition and differences in a cereal grain depending on kinds of cereal and an enviromental conditions. Chemical composition of cereal materials, focused on phenolic acids are also described. Another part deals with theoretical background of methods for the extraction of phenolic acids from bran with description of advantages and disadvantages. Methods for alkaline hydrolysis and extraction of minerals from different kind of bran bran are described in experimental part of this work. Sodium hydroxide (0,5 M) was used for the hydrolysis of bran material. For the determination of the yields of ferulic acid, HPLC method with UV/VIS detection at wave-length 330 nm with usage of mobile phase mixture of methanol and 0,5 % solution of formic acid was used. Acidified deionized water was used for the extraction of nutritionally interesting elements from bran and also from the hydrolyzed material. For the determination of the elemental composition of extracts (Cu, Fe, Zn, Mn, P, Ca, K, Mg), ICP-OES method was used. It was found, that differences in wheat bran species in terms of yields of Ferulic acid are satistically important. The biggest yields of ferulic acid was obtained from the sample of spelt. The smallest yields of this acic was obtained from the sample of oat. Content of micro and macro-elements showed statistical important differences in different kinds of bran and their hydrolyzed waste. Most abundand elemets were obtained from rice bran and from spelts hydrolyzate. The smallest abundand elemens were obtaned from oat bran and hydrolyzate.
Hydrolytical stability of poly(3-hydroxybutyrate)

The bachelor thesis deals with the study of hydrolytical stability of poly(3-hydroxybutyrate) in the hydrolysis with excess of water and in the hydrolytic degradation in melt. Theoretical part summarises knowledge about the process, mechanism and kinetics of hydrolysis and contains an overview of further decomposition reactions of poly(3-hydroxybutyrate). Experimental part is focused on degradation of poly(3-hydroxybutyrate) in the presence of water and on comparing the rate of hydrolytical degradation with acidic and alkaline catalysis. In order to comparing the range of hydrolysis, hydrolytic degradation of poly(3-hydroxybutyrate) was investigated in extruder at the temperature of 170 °C. Samples were analyzed to determine the melt volume index, melt temperature, degree of crystallinity, distribution of molecular mass and the structure of poly(3 hydroxybutyrate) using infrared spectroscopy.
Hydrolysis and subsequent processing of poultry feathers

The goal of this thesis is to introduce and explain technology of hydrolysis and subsequent processing of poultry feathers and make calculation of line for processing poultry feathers. This thesis also includes technological scheme of processing line, design and final evaluation of chosen processing technology.
QM/MM výpočty a klasické molekulárně-dynamické simulace biomolekul

Systematic calculations were performed to uncover the free energy surfaces for hydrolytic reactions of methyl-diphosphate (in vacuum and implicit solvents) and GTP in EF-Tu active site. Density functional theory and ONIOM extrapolative QM/MM scheme were adopted for the assay. In accordance with experiments, the catalytic effect of the sodium cation was mild. It changes the conformation of GTP attracting its negatively charged oxygen atoms. hydrolýze GTP. The Na+ also equilibrates the charges of all phosphate groups of the GTP mostly by transferring electrons from gamma to beta-phosphate group, which is characteristic for the intermediate states during the hydrolytic reaction.
Preparation and characterization of the photocatalytic active TiO2 nanoparticles

Bachelor thesis Preparation and characterization of the photocatalytic active TiO2 nanoparticles Lenka Volfová 2015, ABSTRACT The present work concerns the preparation of photocatalytically active TiO2 nanoparticles, their characterization and study of their photocatalytic properties. The colloidal solution was obtained by hydrolysis of aqueous solutions of TiOSO4. The samples were lyophilized and the product was annealed at 500 řC, 650 řC, 800 řC and 950 řC. The microstructure, morphology and other characteristics of the products were analysed by several physical and chemical methods: X-ray, EPR, SEM, HRTEM and BET. The measurements of IR, UV/VIS and DTA spectra have also been carried out. Photocatalytic activities were investigated by measurement of kinetics of degradation of 4-chlorophenol or methylene blue aqueous solutions under UV light. Measurement of photocatalytic activity of the samples was compared with the standard Degussa P25. It was found that the efficiency of the samples was higher than the standard.
