Teleworking Mother: Stategy for Reconciling Caregiving and Work

This thesis aspires to analyse unique strategies of teleworking mothers with childcare responsibilities. Teleworking as a relatively new form of flexible working arrangement may provide them with distinctive opportunity to reconcile their role within family and labour market at the same time, at the location of their home. In the theoretical part of my diploma thesis I pay special attention to the Czech labour market and family policies from gender point of view and I also introduce the concept of teleworking in relation to gender specifics. Afterwards in the analytical part I analyse three key areas to understand the whole reconciliation strategy - motivation to choose teleworking instead of parental leave, ways of workday organization and different forms of securing childcare. Equality of men and women in labour relations in the Czech Republic and the Nordic countries

The Equality of Men and Women in Labour Relations in the Czech Republic and the Nordic Countries Abstract The subject of the equality of men and women in labour relations is the key topic of this thesis. The main attention is paid to equality and non-discrimination of both parents in the performance of paid employment, especially in terms of opportunities to reconcile their work and family life provided by employers. The scope of this thesis is a comparison of family policies in the Nordic countries, i.e., Sweden, Norway, Finland, Denmark, and Iceland, with the rest of the European countries, especially the Czech Republic. The Nordic countries are generally considered to be the most progressive in many areas, but exceptionally in the area of equal access to both sexes as so- called gender-neutral states, where gender plays almost no role. This specificity was essential for their choice as role models in the field of family policy and the harmonization tools used. The comparison of Czech and German family policy enabling the harmonization of work and family life

The subject of this thesis is the comparison of family policy in the Czech Republic and Federal Republic of Germany in terms of measures adjustment enabling the reconciliation of work and care about the preschool children. The issue of work-life balance is further presented in the context of traditional sociological concepts and welfare state model theories, followed by a description of trends and current order of women's employment on the labor market of both countries and accompanied by the description of demographic trends. Thesis is specifically focused on the description of the governmental family policy of both countries in the perspective of parental leave entitlement settings, facilities of children day care facilities like nursery schools and creche, and formal involvement of fathers in the childcare. The methodological part of thesis is focused on the description of the family policy measures of the respective country. Comparison of both countries in terms of enabling the reconciliation of work and childcare is made on the basis of analyse and comparison of both the secondary quantitative data and the qualitative data drawn from the discussions placed on the free available web sites. Analysis of low-molecular proteins in barley by the SDS-PAGE method during malting

This diploma thesis deals with the analysis of low molecular weight proteins in barley during malting by SDS-PAGE method. Attention was paid to PR proteins, specifically LTP proteins and thionins, considered to be connected with gushing effect. Barley samples, malt intermediates and malt samples taken in 10 consecutive days to cover the whole malting process were used for the experiment. In total, 5 samplings were used for the analysis. Proteins extracted from the samples were separated by SDS-PAGE using a Tris-tricine buffer system. The protein lines of LTP proteins and thionins were identified in the resulting gels. The relative optical density values of the selected protein bands were obtained to assess the effect of malting on these proteins. A similar pattern of change in the content of mentioned low molecular weight proteins during the malting process was observed. This was confirmed by finding a statistically significant positive correlation between the relative optical density values of LTP proteins and thionins. Furthermore, the relationship between the low molecular weight protein content and the gushing potential and the microbiological contamination of the samples was investigated, but was not confirmed.
Barley Proteomic Studies Related to Beer Production

Tato práce se zabývá proteomickými studiemi ječmene v souvislosti s výrobou piva. Ječmen patří mezi nejvýznamnější plodiny na světě a je využíván hlavně pro sladovnické účely, nejčastěji pro pivovarnictví. Studium proteinů ječmene během sladování a výroby piva poskytuje informace o změnách v proteinovém složení nebo jejich posttranslačních modifikacích. Jelikož jsou proteiny v ječmeni a jejich změny zásadní pro kvalitu sladu a piva, proteomické studie ječmene mají potenciál pro zlepšení procesu sladování a pivovarnictví. Hlavním cílem této práce je studium ve vodě rozpustných proteinů ječmene a jejich změn, ke kterým dochází během sladování a výroby piva. Rozdíly v proteinovém složení byly sledovány pomocí gelové elektroforézy, kapalinové chromatografie na reverzní fázi, gelové chromatografie a MALDI-TOF hmotnostní spektrometrie. Během sladování se vlivem klíčení zrna zvyšuje množství některých proteinů a také jsou tvořeny nové proteiny. V průběhu vaření piva se naopak v důsledku vysoké teploty a enzymatické aktivity proteáz mnoho proteinů rozkládá. Těmto drsným podmínkám odolají jen některé proteiny, které přechází až do piva a mohou ovlivnit jeho kvalitu. Dále byly zkoumány různé odrůdy ječmene a jejich rozdíly. Byly porovnány odrůdy povolené pro výrobu certifikovaného Českého piva s jednou osvědčenou sladovnickou odrůdou a jednou nesladovnickou odrůdou ječmene. Kromě toho byly studovány v alkoholu rozpustné proteiny ječmene a jejich změny v průběhu sladování. Zvláštní pozornost byla věnována vybrané skupině posttranslačních modifikací proteinů: glykosylacím. Neenzymaticky glykosylované proteiny ječmene (neboli glykované proteiny) jsou tvořeny v průběhu sladování kvůli přítomnosti velkého množství glukózy uvolněné z rozkladu škrobu. Glykované proteiny ovlivňují stabilitu proteinů a kvalitu piva, obzvlášť pěnotvorný účinek. Enzymatické N-glykosylace představují nejčastěji studované posttranslační modifikace u rostlin, protože glykoproteiny hrají klíčovou roli v různých biologických funkcích. Glykoproteiny jsou často přítomny v malém množství, a proto je pro jejich analýzu potřebné obohacení glykoproteinů z komplexní směsi. Pro studium glykoproteinů byla využita afinitní chromatografie s lektinem concanavalin A. Kromě toho byla také optimalizována analýza sacharidové části glykoproteinů. Tato disertační práce přináší důležité informace o proteinech ječmene, jejich změnách a analýze, které budou užitečné pro další studium.
Proposal of Work-Life Balance in Selected Company

The diploma thesis deals with the issue of work-life balance of employees and especially managers in a chosen company. The theoretical part is about issues of gender, factors influencing managers’ careers and possibilities of work-life balance. In the practical part, there is an analysis of a present situation in a company. On the basis of theoretical findings, analysis of company’s documents, a questionnaire and interviews with chosen employees, there will be made some suggestions for improving a present situation in a company. There will be also a financial evaluation of new steps as a part of final suggestion.
The Proposal of Work-Life Balance in Selected Company

The diploma thesis deals with the issue of work-life balance in a chosen organisation. The work life balance instruments are described in detail and theoretical findings are compared with reality. The thesis identifies possibilities and barriers to work-life balance of employees of the observed company. On the basis of semistructured interview and questionnaire survey, measures for optimal work life balance are suggested.
Changes of protein profile in barley during malting

This diploma thesis is focused on studies of changing of protein profile during barley malting. Substantial part of this work is devoted to the proteomics identification of barley proteins which change during malting and so become more stationary and they influence quality of beer (haze and foam in beer). For this experiment was used barley variety Jersey. In the theoretical part of this thesis there is information about beer, manufacturing of beer with description of important commodities for manufacturing of beer and information about barley malting and information about malting process. Next there is description of methods for separation of proteins (1D gel electrophoresis and 2D gel electrophoresis), MALDI TOF/TOF mass spectrometry and this use for the analysis and identification of proteins, the use of matrices and ways of the sample preparation. In the experimental part of this thesis there was carried out the optimisation of the dosage of sample for 1D gel electrophoresis and the optimisation of staining. The 15 % TRIS-HCl gel was the best, this gel was stained by Commassie Brilliant Blue G-250. For illustration of changes was made 2D gel electrophoresis. With help of method peptide mass fingerprinting and MS/MS protein of barley – protein Z, -amylase subtilisin inhibitor, -amylase a peroxidase were identificated. The analysis of barley extract intact proteins was carried out, this analysis was focused on changes of important barley protein LTP 1.
Comparison of family policy of Germany and the Czech Republic

This bachelor thesis deals with the comparison of family policy of the Czech republic and Germany. The actual meaning of this topic is caused due to decreasing natality across whole Europe. Germany is the symbol of high developed economic country and very well maintaned family policy. Czech republic as a post-communist country is still exploring the best way of the family policy. The bachelor thesis evaluates demographic development of both countries and compares the family policy from the point of view of direct and non direct financial instruments, tax level, institutional background for the pre-school children and the harmonization of family and professional life. There are suggestions for improvements related at the end of the thesis to Czech republic.
Evaluation of storage protein profiles modifications in barley (\kur{Hordeum vulgare} L.) grains during malting procedure using gel electrophoresis

This bachelor thesis is focused on evaluation of storage protein profiles modifications in barley (Hordeum vulgare L.) grains during malting procedure using gel electrophoresis. It has been analyzed three varieties of spring barley. One nonmalting variety Herris and two malting varieties Radegast and Sebastien. There are significant proteolysis changes during the malting procedure. Theese changes of storage proteins were captured and evaluated by gel electrophoresis (SDS-PAGE) on the basis of changes in the molecular weight in the different phases of malting procedure. Next observation which has been done was the measurement of the thousand grain weight (TGW) before and after the malting procedure. Theese results have been evaluated by statistic program.
