Characteristics of expression vectors for Corynebacterium glutamicum and their use for studies of sigma factors of RNA polymerase

The aim of the thesis was to characterize chosen expression vectors used in biotechnologically important bacterial species, Corynebacterium glutamicum, and to test their use in studies of promoter activity control by sigma factors of RNA polymerase. Different properties of these vectors (level of expression of the cloned gene, leaky expression without inducer, dependence of expression level on inducer concentration and cell population homogeneity) were found by determination of expression level of the model gfpuv gene by fluorescence intensity assay of the produced protein and by gfpuv-expressing C. glutamicum cell population analysis using flow cytometry. The vector pEC-XT99A was chosen for testing the bi-plasmid system for assignment of a sigma factor to the chosen promoter. Although the level of expression provided by pEC-XT99A was not high, the vector showed no leaky expression, expression from the vector was comparable for a wide range of IPTG concentrations and the cell population was homogenous concerning the gene expression. John Dewey's Aesthetics and interactive art

This bachelor thesis is especially focused on John Dewey's theory. The first and the main part of the bachelor thesis deals with Dewey's conception of experience, with analysis of this process of experience and with aesthetic level of experience. Significant part of bachelor thesis is dedicated to aesthetic experience with art as well, which is discussed in the context of every day experiences. Continuity between both is highlighted. The second part of the bachelor thesis is an addendum to the interpretation of Dewey's theory. This part of the thesis deals with interactive art. Interactive art is interpreted using Dewey's theory. Interactive art is in this paper understood as artistic type which can emphasize continuity between art and everyday experience so as Dewey does.
New Names of Professions

The thesis New Names of Professions is divided into a theoretical and a practical part. In the theoretical part, basic terms from the world of work are defined; the essential term is profession. Furthermore, neologisms are defined in the first part in accordance with professional literature without being confined to denomination of persons. For the purposes of the practical part, new names of professions were excerpted from the list of professions of the employment office and from a list in Učitelské noviny. Their occurrence was further verified using the EDA and Newton media databases and the results of the research were compared with the created list. An alphabetical list of the excerpted professions and a thematically divided list of those professions are an integral part of this work.
The effect of selected endocrine disruptors on the cytochromes P450 1A1 and 2C

Many currently produced chemicals reveal specific properties which allow them to be referred to as endocrine disruptors (ED). These substances exhibit an exogenic hormone activity and usually act as antagonists or agonists of endogenic hormones. The exogenic EDs studied in this work were 17α-ethinylestradiol (EE2) and benzoapyrene (BaP). 17β-estradiol (E2), a typical endogenic hormone, was also included to the study. In the presented work, the effect of these EDs and their combinations on the expression and specific activities of cytochromes P450 (CYP) 1A1 and 2C was determined. First, the microsomal fraction (MF) of liver, kidney and lung of rats premedicated with these compounds or without premedication was isolated. CYP expression was assessed by the Western blot analyses in these MF samples. Moreover, CYP1A1 and CYP2C specific activities were evaluated. It was found that premedication of rats with BaP increased CYP1A1 expression in all above mentioned organs. Whereas BaP strongly induced rat CYP1A1, EE2 and E2 were almost without this effect. But, when these disruptors were administered to rats with BaP, they supported its potency to induce CYP1A1. Intuition and Meaning in Edmund Husserl

The submitted thesis deals with the relation of intuition to meaning in Edmund Husserl's early work. The structure of the thesis is given by Husserl's triadic structure of epistemic acts, which is formed by the realm of subjectivity, ideality and objectiveness. In this framework, we treat the meaning in its proper sense of meaning-intention, which is but also related to the objectiveness. This relation is made possible by meaning-fulfilment, which is intuitive. At the end, we also emphasize the role of intuition not only in the Logical Investigations, which are to our thesis the central work of Husserl, but also in the whole of Husserl's philosophy.

Miroslava Mayerová Title of the Bachelor's Thesis: Art group 12/15 and its place in the second half of the 20th century Czech art Abstract This thesis focuses on the evolution and influence of art group 12/15 on socio-cultural development of society in the second half of the 20th century. The theoretical part encompasses the socio-cultural environment and historical situation before and during the formation of the group, describes its formation, and presents the work of individual group members until their second joint exhibition. The didactic part teaches students the group's activities by utilising concepts used by 12/15. The practical part of the work carries out didactic task and by form of self-assess evaluates its feasibility and usefulness. The work includes historical documents and articles about group.
Study of effect of flavonoids on biotransformation enzymes expression

Flavonoids belong to the group of phenolic compounds included in the secondary metabolites of plants, which are represented mainly in fruits and vegetables. These compounds have antioxidant and anti-carcinogenic effects. Some of them can affect biotransformation enzymes, which include the cytochrome P450 and can interfere with the metabolism of xenobiotics. In the present work we studied effect of dihydromyricetine on the gene expression of cytochrome P450 1A1, 1A2 and cytochrome b5 in livers, lungs, kidneys, colon and small intestine of laboratory rats. The small intestine was divided into three parts namely a proximal, middle and distal. At first, isolation of total RNA was made using commercial reagents and isolated RNA was converted to cDNA by reverse transcription. Finally, using polymerase chain reaction in real time (RT-PCR), the relatively gene expression of genes observed in the organs of laboratory rats pretreated dihydromyricetine and control laboratory rats (untreated) to a reference gene β-actin was determined. It was found that dihydromyricetine did not significantly affect the gene expression of studied genes in most organs.
