Philosophical approach to life and death of people who were between life and death

This dissertation thesis goes into the depths of philosophical reflection on the nature of human existence and its relationship to life and death, especially for individuals who have gone through extreme life situations. The thesis explores attitudes towards free will, life and death in people who have found themselves on the brink of life, whether due to addiction, illness or other life-threatening situations. The aim is not only to analyse changes in these individuals' attitudes towards life and death, but also to explore the intellectual and philosophical development that may be prompted by this existential limit. Regardless of the cause of this confrontation with the limit of life and death, we seek to uncover how this experience affects the deeper dimensions of human existence. The work is based on the conviction that illness, although challenging on the physical plane, has an extraordinary effect on the spiritual sphere. It also highlights the impact of illness on the physical and spiritual aspects of the human being and seeks to show this by examining the works of great philosophers whose ideas have shaped the world. Charles University, Faculty of Pharmacy in Hradec Králové Department of Biological and Medical Sciences Title of Diploma Thesis: Effect of endoglin blockage on inflammation-induced endothelial dysfunction in human aortic endothelial cells Author: Andrea Škubová Supervisor of Diploma Thesis: PharmDr. Katarína Tripská, Ph.D. Aim: The aim of this thesis was to describe the effect of TNF-α and LPS-induced inflammation on the expression of endoglin and its transcription factors, the expression of biomarkers of endothelial dysfunction, the levels of soluble endoglin, and the adhesion and transmigration of monocytes through human aortic endothelial cells. We further investigated the effect of endoglin blockage by the monoclonal antibody TRC105 (carotuximab) on these processes. Methods: In this thesis, human aortic endothelial cells (HAEC) were used, and they were exposed for 16 hours to TNF-α (10 ng/ml) and LPS (100 ng/ml) and for 12 hours to TRC105 (300 µg/ml). The mRNA expression of endoglin and its transcription factors (KLF6, RelA, NR1H3), mRNA expression of inflammatory adhesion molecules (ICAM-1, VCAM-1, SELE) and mRNA expression of MMP-14 enzyme were measured by real time qRT-PCR. Feasibility Study of the Reconstruction of the Building Construction

The aim of this work is the explanation of the issue of passive houses and renovation of existing buildings to passive energy standard. Another task of the work is a feasibility study of building renovation of the "Tuzex" for the needs of Center of sustainable construction in Brno. Part of this study is to create a marketing analysis, design of organizational project management, a description of technical solution, impact assessment project on the environment and in particular evaluating the economic efficiency of investment. The result of this work is to evaluate the feasibility of the project under current conditions.
Sentence representations with similarity interpretation

Sentence representations - embeddings - obtained from neural network models are the core part of many applications in both academia and industry. Although embeddings reach great results in correlation with human sense of sentence similarity, there is often a lack of explanation for why models choose sentences to be similar. In this thesis, we strive to increase the interpretability of model embeddings by incorporating different semantic sentence level annotations in the learning process. We introduce a model called SBERTslice that produces embeddings that can distinguish nuanced semantic variations in text, including elements like negation, sentiment, named entities, emotional tone, and verb-oriented relation between words in a text. We evaluated SBERTslice embeddings in various text classification and semantic sim- ilarity tasks and for a majority of them, SBERTslice outperformed the original SBERT. 1
Detection of soluble endogline in Lab-in-Syringe

The main target of the thesis was to develop an automated method for the isolation of endoglin from a sample using a magnetic immunosorbent in Lab-In-Syringe with subsequent immunodetection by affiblot. The theoretical part deals with endoglin, in its membrane and soluble form, especially with their connection to endothelial dysfunction. The principles of anti-endoglin therapy using the monoclonal antibody carotuximab or TRC105 are also briefly described here. Furthermore, the theoretical part summarizes the basics of the automation in Lab-In-Syringe and of the affiblot technique, their principles, and their applications. The last chapter deals with the biofunctionalization of magnetic particles by antibodies in order to prepare a magnetic immunosorbent which is used for the isolation of proteins from a complex sample. The experimental part describes methods for testing the affiblot prototypes. The main goal was to develop a method for the preparation of anti-endoglin magnetic immunosorbent and to increase the capacity of the method by upscaling it from a 1 ml to a 5 ml syringe. Furthermore, methods for the isolation of the soluble form of human endoglin from the culture medium containing the drug TRC105 were developed for 1 ml and 5 ml syringes. The dissertation The Phenomenon of Responsibility in the Postmodern Era discusses the phenomenon of responsibility in the context of the postmodern era. The issue of responsibility is viewed here primarily from a philosophical point of view. At the same time, the dissertation attempts a partial thematic overlap, in connection with the meaning of the selected concept of responsibility, within the ethics and particularly even other related social science disciplines. In the beginning, the basic problematics is set here. This discusses the etymological point of view and responsibility is subsequently given here in the connection of the context of terms such as freedom, response or ambiguity. Furthermore, the basic relationship is presented, in the context of the phenomenon of responsibility, and responsibility is also here confronted with the present. Subsequently, the approaches and theses of an important author of the 20th century, who systematically dealt with the phenomenon of responsibility, the French author Emmanuel Levinas, are particularly emphasized. Threats and opportunities in motivating senior workers in education with experience in further education.

The diploma thesis deals with the motivation of managers in the field of further education. The aim of the diploma thesis is to find out how kindergarten principals with practice perceive their own motivation to continue their education, or to identify a certain key critical period during their career as a kindergarten principal. Attention is paid to the characteristics of managers, their motivation, personal development and further education. Issues related to the burnout syndrome, possible factors causining the resigns of a manager, fundamental obstacles in motivation and critical moments of managers in their careers are also elaborated. The thesis includes qualitative research based on personal interviews with educational leaders, which took the form of semi-structured interviews. Qualitative research was chosen on the basis that appropriate methods could be used to achieve the stated aim of the thesis. A research sample was selected and specified for the qualitative research work. The research sample for this thesis was selected as the headmistresses of kindergartens who have long experience in the position, six years or more. The interviews obtained were then analyzed using the coding method. The coding was divided and identified into five main categories to correspond as closely as possible...
