Evaluation of ecological education in Ecocentre Vlašim

The main topic of this thesis is the CUNC basic organization unit and Podblanické Ecocentre CUNC in Vlašim. The first part of the thesis presents the Czech Union for Nature Conservation (CUNC) followed by an overview of its Environmental Centres. The next section contains a summary of all the activities carried out by the CUNC basic organization unit and Podblanické Ecocentre CUNC in Vlašim from their establishment to the present and recapitulates the formation of these organisations themselves. The thesis also covers a characteristic of all the one-day as well as residential teaching programmes for secondary schools, technical schools and grammar schools carried out by the Ecocentre. The greatest emphasis is laid on four selected one-day teaching programmes, for which a research was conducted by using a questionnaire method. Its aim was to determine the influence on pupils and students of the educational programmes from secondary schools, technical schools and grammar schools.
Nursing Care for Patient with Tuberculosis

Nursing Care for Patient with Tuberculosis Tuberculosis is a disease accompanying human beings since immemorial time. Its originally epidemic occurrence has changed into endemic one. Presented bachelor thesis is devoted to the issue of nursing care for patients with tuberculosis. Its objective is to outline the course of the illness as well as measures necessarily taken to avoid its further diffusion, and to describe principal nursing care for patients. The empirical part of the thesis is based on a sample survey focused on nurses' knowledge on tuberculosis and hygienic-epidemiological measures applied during prevalence of this disease. The level of knowledge in two respondents groups - nurses in pulmonary and infection departments and nurses working in other ones - are compared there.

This bachelor thesis is dealing with synthetic food dyes and their impact on human health. Synthetic dyes are suspected of having a toxic effect on living organisms, which is proven for some of these substances by experiments on animals. The experimental part of this thesis is focused on the determination of the quantity of these synthetic food dyes in 12 selected samples of commercially available products by means of spectrophotometric multi-component analysis. The amount of synthetic food dyes was determined in 3 samples of al-coholic beverages, 6 samples of non-alcoholic beverages, 2 samples of food and in one food supplement. Subsequently, these results were compared with the maximum allowable quanti-ty (MPM) of synthetic food dyes for each food category set by the legislation. All beverages, including one food supplement, which is recommended to be consumed in aqueous solution, have been complied with given numbers of the MPM. However, two samples of eatables (specifically chewing gum) did not meet these numbers and the amount of synthetic dyes was above limits in them. In the case of these samples, it would be appropriate to carry out an additional verification by another method and, if the high quantity of synthetic food dyes would be confirmed in them, to recommend their withdrawal from the market.
Interaction between hydrogenosomes and endoplasmic reticulum in Trichomonas vaginalis

Endoplasmic reticulum-mitochondria encounter structure (ERMES) is a protein complex tethering ER and mitochondria. ERMES consists of four core subunits - Mmm1, Mmm2 (Mdm34), Mdm10 and Mdm12. It was first discovered in Saccharomyces cerevisiae and most functional information is based on studies of this organism. ERMES affects mitochondrial distribution and morphology, participates in lipid trafficking and is important for homeostasis of the cell. In Trichomonas vaginalis, the human urogenital parasite, three genes for putative, highly divergent components of ERMES complex were predicted. However, the cell localization of these proteins and their function is unknown. This thesis is focused on investigation of ERMES components in T. vaginalis, their cellular localization, interactions between components and identification of their possible interacting partners.
Adhesion and growth of adipose tissue-derived stem cells on fibrin assemblies with attached growth factors for tissue engineering of heart valves

Currently used xenogeneic biological heart valve prostheses are decellularized and crosslinked with glutaraldehyde. These grafts usually undergo degeneration and calcification. Pericardium-based heart valve prostheses, re-seeded with autologous cells, i.e. Adipose tissue-derived cells (ASCs) and endothelial cells, could have longer durability and biocompatibility. In order to improve the adhesion of cells and their ingrowth into decellularized pericardium, various fibrin (Fb) layers were developed, i.e. Fb, Fb with covalently bound heparin (H), Fb with either vascular endothelial growth factor (VEGF) or fibroblast growth factor 2 (FGF) in various concentrations (1 ng/ml, 10 ng/ml, 100 ng/ml) or with both VEGF and FGF (100 ng/ml). Growth factors were attached onto Fb via heparin or were adsorbed. ASCs were seeded on theses layers in a DMEM medium supplemented with 2 % of fetal bovine serum, TGFβ1 and BMP-4 (both 2.5 ng/ml), and with ascorbic acid. Cell adhesion and growth/viability was assessed by counted cell number/MTS evaluation. ASCs were stained for differentiation markers of smooth muscle cells, such as alpha-actin, calponin, and myosin heavy chain. On day 7, ASCs on Fb-H-VEGF layers produced both calponin and alpha-actin. An increased FGF concentration caused reduced calponin staining of ASCs. Lack of heparin in fibrin assemblies with growth factors inhibited the production of both alpha-actin and calponin in ASCs. The highest ASCs density/viability was found on Fb-H-VEGF-FGF layer. The proper formulation of fibrin coatings could be favorable for ASCs growth and differentiation and could subsequently support endothelialization of cardiovascular prostheses with endothelial cells.
Tobacco use and its consequences among children 8-12 years old. Prevalence and forms of smoking, brand and source awareness, smoking initiation.

Objectives: Within the Czech Republic there are missing reports on smoking prevalence, forms of smoking, sources of cigarettes or the influence of tobacco marketing on children younger than 12 years of age. Methods: Between 2009 and 2012 we conducted a cross sectional survey with 3 887 children from Prague and 592 from the Usti Region, age 8-12 years old, from 51 primary schools. The survey was part of a school based smoking prevention program at participating schools. Results: Among children, 23.3% reported some experience with smoking. Smoking experience was higher among boys, and children with family members who smoked. Cigarettes were the most common form of smoking (about 17 % of all respondents), followed by water-pipe (prevalence in Prague was 7.4% and 4.9% in Usti Region (p = 0.030)). Less than 1% of all children reported experience with marijuana. Relatives were the most commonly named source of cigarettes for first smoking experience (43.1%). Among children that reported smoking at least once a month, relatives were the most common source of cigarettes (32.5%), and 15.5% reported purchasing their own cigarettes. Comparation of a consument loans on Czech market

Needs of people are still increasing and with that is evenly increasing need of financial possibilities. I chose this theme for my diploma because it is present topic and it is affecting all areas of Czech Republic. Goal of my bachelor's diploma "Comparison of consumer's loan on Czech market" was mainly analysis of situation at market of consumer's loan and comparison of non-purpose consumer's loans offered by financial institutions at Czech market. This goal led me to determination of which banks offers the most favorable loans. Theory part of my diploma is mainly focused on characterizing basic terms of this theme and categories of consumer's loans. Then the term of compound interest, types of compounds interests and their calculations of compound interests are according to formulas. Practical part of my diploma is occupied with analysis of current condition at financial institution market. For this work I chose 5 institutions offering loan services and then I chose products of those institutions based on the similarities of terms for granting and drawing loans. To accomplish the goal is important to know theoretical knowledge of consumer's loans and current state of drawings consumer's loans, that are offered by financial institutions to Czech people. A main criterion for the comparison of loans was rate of compound interest due to duration of repayment of consumer's loan. I decided to add multi-criteria comparison by method of order. It compared chosen consumer's loans according to rating criteria like APR, compound interest, high of repayments monthly, total number of repaid amount and fee for early repayments. In the end conclusion is shown as implementation of results. In what order those chosen financial institutions are placed and how favorable is any of those loans for consumer.
