Regulation of alternative splicing

Alternative splicing is an important cellular mechanism. It allows to produce multiple protein isoforms from a limited number of genes. Regulation of alternative splicing involves cis-acting elements on pre-mRNA and trans-acting splicing factors (SR and hnRNP proteins). Because splicing occurs co-transcriptionaly, chromatin structure appears to have a role in the regulation of alternative splicing. We have studied the effect of histone acetylation on alternative splicing. We have prepared splicing reporter for alternative EDB exon, which is part of the fibronectin gene. We have shown, that the inhibition of histone deacetylases affects splicing pattern of EDB exon from the reporter in the same way as the splicing of the endogenous EDB exon. Furthermore, we have shown, that the structure of the promoter affects splicing of alternative EDB exon from splicing reporter. Currently we have found out, that the structure of the promoter influences the degree of histone H4 acetylation. Inclusion of alternative EDB exon in mRNA was inversely proportional to histon acetylation on the reporter. This work might explain why various promoters have different splicing patterns of alternative exons.

Novel Approaches to the Application of Electrochemical Volatile Compounds Generation in Spectrometric Methods

Various possibilities for the use of electrochemical generation of volatile compounds for the determination of trace and ultratrace quantities of elements using atomic absorption spectrometry and optical emission spectrometry, respectively, were studied in this work. The Thesis is divided into four thematic sections. Electrochemical cold mercury vapor generation is common to the first two parts. The third part is devoted to electrochemical generation of the volatile form of cadmium and the last part deals with the use of the electrochemical hydride generator as a derivatization unit for speciation analysis of arsenic compounds. The optimization of each individual determination was carried out including, for example, various designs of the generator of volatile compounds (forms) of the analyte, the changes in the design of the gas-liquid separator, optimization of the concentration and the flow rate of all the solutions and the carrier gas. However, the value of the electric current employed was the most important parameter. In the first section, the in-situ trapping system of the cold mercury vapor generated in the graphite furnace (with the inner surface covered with a gold foil) of the atomic absorption spectrometer was also designed and optimized. The Social Transformation of Former "Closed Cities" in Estonia. Case-study of Paldiski and Sillamae

The project will focus on analytical and comparative studies of social and economical structure transformations in "closed cities" (ZATO) and in the former military cities in Estonia. The main subjects of this comparative study are former <closed cities> Paldiski and Sillamae. Closed cities were characterized by limited communication with outside environment and by the lack of exchange of information. The state institutions, such as police or Public Prosecutor's Office, had extended legal power there. These and other restrictive boundaries were offset by state benefits and by better abundant supplies for the city. The risk factor of local economy was the military focus in exchange for self sustainability. After the collapse of the USSR and the loss of state fundings these cities fell into a deep social and economical crisis, which is needed to overcome until now The main question of that project is if the post-soviet development of those cities has different or common elements and if in a specific way of transformation of those cities exist.

The thesis "The Policy with an emphasis to reduce the long-term unemployment of people with primary education in selected region - Ústecko" deals with the labour market and educational system within the local level. The current labour demand prefers workers with qualification, expert knowledges and skills. Therefore, the possibilities of people with primary education to find a job are decreasing. Regarding these facts, people with low qualification are threatened by long-term unemployment. Contemporary services of state employment policy in Ústí nad Labem do not focus on people with primary education. As well as the educational system within this region is deficient. Despite the fact that some applicants for a job are not motivated enough or willing to study, the education system is not flexible and thus, applicants have only limited opportunities for further education and for receiving vocational certificates or graduation diplomas. According to above mentioned facts, this thesis deals with an educational policy as fundamental element for prevention of the long-term unemployment of people with primary education. Assembled structure of hall in Vysoke Mýto

The diploma thesis is focused on design and assessment of mounted storage hall. I have designed prestressed truss, reinforced truss, beam, column and foundation pad. I have made calculations for consideration of ultimated limit state as well. I have made analysis of internal forces for beams just with manual calculations. Analyses of internal forces for column and foundation pad were found with Scia Engeneer. All calculations are following the Eurocode 2. All designed elements have formwork drawing and reinforcement drawing.

Statistical aspects of nuclear collective dynamics

The current PhD thesis presents a collection of selected articles related to the theoretical and numerical study of low-energy collective dynamics of atomic nuclei. The articles were published or recently submitted to international physics journals and were authored or co-authored by the author of the thesis. The eects in collective dynamics have been studied within the framework of two common models-the interacting boson model (IBM) and to a lesser extent the geometric collective model (GCM). The "statistical aspects" in the title relate predominantly to the interplay of ordered and chaotic behavior observed in properties of quantum eigenstates as well as in the classical limits of the models. The main attention was devoted to correlations between the measures of regularity/chaos and the presence of exact and approximate dynamical symmetries. An important subject of the studies were also the relationships between the properties of the classical and quantum solutions of the models both in the integrable regime as well as in the mixed regime containing elements of regularity and chaos simultaneously.

Corporate identity of HC Slavia Praha and its utilization in commercial activities

CORPORATE IDENTITY OF HC SLAVIA PRAHA AND ITS UTILIZATION IN COMMERCIAL ACTIVITIES Objectives: The aim of this work is to assess the corporate identity of HC Slavia Praha and its utilization in commercial activities. The next object is to propose new instruments to reinforce existing components and integrate new elements. Methods: Case study, document research and interview were methods used for the elaboration of this thesis. We applied the acquired information to assess current corporate identity and to compile a SWOT analysis. Following corporate identity improvements are based on SWOT analysis's results. Results: Thesis described general signs of corporate identity of HC Slavia Praha in its complexity and together with its utilization in commercial activities. Through the SWOT analysis we emphasized main assets and limitations of HC Slavia Praha in this field and suggested the improvement of actual state of affairs. We can summarize all suggestions into one fundamental point which is to focus on the growth of audience during home games and as a result the building of a strong brand of HC Slavia Praha. Word order in Czech subordinate clauses

This diploma essay contributes to the studies of the Czech word order. Its theoretical background is provided by the findings of the Czech school of functional sentence perspective (FSP), represented by Jan Firbas and Aleš Svoboda. Based on the material excerpted from the present-day Czech corpora SYN 2005, this paper presents a broad scale of word order varieties in Czech subordinate clauses. Special attention is paid to the word order position of the theme elements and the most common adverbial complements. In comparison to primary clause some differences are pointed out: The transit centre (or transit proper or theme proper) position in front of the theme centre and rheme is a characteristical feature of subordinate clause, whereas in the primary clauses it applies to rather limited number of clauses. The emotional (subjective) word order is often caused by unfitting allocation of information instead of emotional movement. The initial theme in the subordinate clause seems to be of higher degree of communicational dynamism (CD) compared to the primary clause. It proved useful to differentiate premedial theme (in front of transit centre), which has higher degree of CD than postmedial theme (behind transit centre). Governing bodies of General and Limited Partnerships The subject of my thesis is to analyse process of acting on behalf of a corporation and to depict specific bodies which take part in this essential process, their competency, powers and also their responsibility. The reason for my research is based on fact that not a single corporation stands alone in this world which makes interactions with other corporations, customers and state authorities not only inevitable but more importantly necessary for a corporation to survive in business world. Just to clarify the subject of this thesis does not include process during which bodies of a corporation form will to act based on internal proposals and opinions but it concentrates solely on the acting itself. The thesis is composed of two complex chapters, each of them dealing with different aspects of acting on behalf of a corporation. Chapter One is not only introductory even though it defines basic terminology used in the thesis but it also analyzes common elements of governing bodies.
