Beer proteome analysis by mass spectrometry

The aim of presented diploma thesis was to characterize recent knowledge in the field of beer proteomics. The main part of this work was focused on modern instrumental methods of protein analysis, especially on protein identification by mass spectrometry. In experimental part proteins from selected beer samples were isolated, purified and separated by 2-D electrophoresis. The identification was performed by MALDI MS/MS and LC-MS/MS. Identified proteins were divided into 6 groups - serpines and protein Z, trypsine/-amylase inhibitors, yeast proteins, LTP protein, hordeins and other proteins. Proteomic analysis provided identification of proteins important for final analytical and sensory characteristics of the beer - for final beer quality and taste

Analysis of low abundant lipids in vernix caseosa using chromatographic methods and mass spectrometry

The aim of this work is to develop a separation method for characterization of unknown lipids in vernix caseosa. We focused on very low abundant lipids which have not been previously described in vernix caseosa. The total lipid extract was fractionated using low pressure normal phase liquid chromatography to obtain sub-fractions. The sub-fractions were further separated in high performance liquid chromatography mode to isolate classes of lipids. This procedure enabled us to characterise lipid classes by mass spectrometry and identify one of the class as acyl-ceramides with non-hydroxylated fatty acids.

Strategic partnership between Germany and Russia

This diploma thesis focuses on analysis and evaluation of the German-Russian strategic partnership from the German point of view. The aim of this thesis is to answer a question, whether Russia had ever been or still is a strategic partner of Germany, or whether the establishment of the German-Russian strategic partnership remains rather a political goal than the reality. The theoretical part of the thesis deals with the theoretical definition of the strategic partnership and indentifies key characteristics of this concept. The analytical part is divided into separate chapters analysing individual dimensions of the German-Russian relations: economic, energetic, security and political. The final chapter sets the goal to summarise and to evaluate the main findings arising from the previous chapters and to answer the research question, whether Russia can be or in the past could be considered a German strategic partner.

Causative Constructions \kur{faire+inf.} and their Czech Translation.

This thesis provides a comparison of causative mechanisms between two languages: Czech and French. The aim of this research is to reveal expressions that contain a causative meaning in Czech and to analyze which of them are truly equivalent to the French causative construction faire + infinitive. This work classifies general causative mechanisms, according to their nature, between synthetic (prefix, lexical expressions) and analytic (French complex predicate faire + infinitive, periphrastic constructions, separate clauses) and focuses on the French construction by describing its syntactical and semantic specificity. This causative construction is the basis of a parallel research in corpora InterCorp, a technical tool which helps excerpt authentic texts. Additionally, it is attempted to clarify and classify the usage of the Czech equivalents and search for factors that influence their choice with an eye on the source language.

Supreme Audit Office -- the status and organization

This thesis focuses on the status and organization of the Supreme Audit Office (SAO), which carries out the external audit of public administration in the Czech Republic. For easier orientation in the area offers this work in the first part the theoretical foundation of control in public administration. At the beginning of the analysis is presented the historical development of the highest accounting controls in the territory of today's Czech Republic. In three analytic blocks are listed the findings, which were based on research resources, particularly in the area of constitutional, administrative and financial law and on the study of the legislation and professional publications. SAO status as an independent supervisory authority in the Czech legal system is examined in terms of its inclusion in the Constitution and further discussed is the scope SAO and its relation to local government. Linked to this is especially the current proposed change of the Constitution and the Law on the Supreme Audit Office, which is also reflected in the work. This work also introduces the issue of the status of the SAO in the system of public power. The relation of the Supreme Audit Office to other public authorities is partially analysed. Investigated are also the position and competences of authorities within the SAO. In a separate chapter, based on the analysis presented and evaluated, there are the conclusions and suggestions for possible modifications of the law de lege ferenda.

Economy and efficiency analysis of electronic toll in the Czech Republic

Diploma thesis presents information about electronic toll system in the Czech Republic with the gist being aimed on economy and effectiveness of the system in use. The theme is discussed in separate parts, where the first part consists of explanation of basic terms about road infrastructure, toll system and its functionality in the Czech Republic. Next part is revealing information about public sector and its economy, efficiency and effectiveness that are essential for measuring public sector, and its administrative costs. In the analytical part is evaluated economy of electronic toll with the usage of findings from administrative costs of electronic toll and with its earning. Electronic toll is analyzed as well from the view where are put into comparison domestic and foreign and their weight on the toll and as well from the view of the amount of truck carriers that were using Czech road system before and after electronic toll was introduced. Part of the analysis compares the administrative costs of electronic toll with administrative costs of other taxes in the Czech Republic and it compares systems of electronic tolling in chosen neighboring countries.

c) Analysis of the personnel function in the city government in Ostrov

This thesis contains an analysis of the personnel function in municipal government, specifically in the city government in Ostrov. It involves a mapping of the current human resources function along with a recommended new system of managing human resources. The method used is a comparison of knowledge gained from current city government officials with that obtained from academic literature. The recommendations are divided into separate analytical categories, and are given in the form of practical steps to be applied in human resources management.

The comparison of different types of stationary phases in HPLC analysis of medicament Diclofenac emulgel

The aim of this rigorous work is a comparison of different types of stationary phases for the analysis of the drug Diclofenac emulgel. It is necessary to take into account the many specialized materials from the pharmacopoeias, ICH guidelines and professional scientific journals in solution of the development of HPLC methodology, validation and choice of stationary phase analytical columns. Several types of stationary phases were tested, with the testing of conditions of the separation of active substances from the pharmaceutical formulation - diclofenac emulgel. The best results were placed on Discovery HS-F5 (15 cm x 4.6 mm, 5 m), time analysis was less than 4 minutes; Zorbax SB - CN (4.6 x 150 mm, 5 m) time of analysis was less than 5 min; Chromolith Performance RP - 18e (100 - 3 mm) time analysis was less than 3 minutes.

Application of UHPLC with various types of detection in the field of environmental and food analysis.

The use of UHPLC with various types of detection in the field of environmental and food analysis Abstract The aim of this work was to inform about possible use of UHPLC instrumentation in connection with various types of detection when chromatographic separating of substances in the field of environmental and food analysis is performed. The next aim was to search and study a number of articles referring to UHPLC methodology, which are at present available in the internet databases ScienceDirect and Web of Science. On the basis of the information from the articles there is a graphic summary in this work relating to the number of found articles and the groups of analysed substances within years. In the introduction of the work van Deemter's, Gidding's and Knox's theories about the efficiency of the chromatographic system are mentioned. These theories are substantial for understanding the principle of chromatographic separation. Following chapters deal with the problems of the development of chromatography in the last decades, especially concerning the length and type of analytical columns, the size of filling particles or the temperature and pressure in the chromatographic system. Visualization of Multivariate Statistical Data

The thesis deals with the possibilities of visualization of multivariate statistical data. Since this is a very broad area the thesis is divided into four sections, two of which are theoretically and two practically oriented. The first section is devoted to theoretical aspects of data visualization. It contains information about the building blocks of graphs, and how the brain processes graphs in various stages of perception. The second section charts the available chart types that can be used to display data. Selected types of graphs for continuous and discontinuous multidimensional data are described in detail. The third section focuses on available software tools for creating graphs. The section describes several programs, with focus on STATISTICA, R and MS Excel. The knowledge gained in previous chapters was sufficient source of information to perform a graphical analysis of multidimensional continuous and discrete data and using advanced analytical methods in the last section. This analysis is performed separately on the data file with continuous variables and on a data file with discontinuous (categorical) variables.
