Calculation of storage tank structure for sewage treatment plant

This diploma thesis is focused on finite element analysis of storage tank. The designed computational model in this work is based on existing storage tank documentation. Diploma thesis is divided into parts. Introduction part consists of various storage tanks types description and basis of finite element method. Next part contains methodical process of computational model designs, its characteristics and purpose. Designed models are loaded with set of specific border conditions. These tests are evaluated and described more closely. After that, the table of results is created and von Misses stress of all models is compared. The best model matching real construction is selected after this compare. The final part of this work consists of suggested construction changes based on the analysis results.

An introduction to sociology of theatre

Sociologie divadla je sociologickou disciplínou, která se začíná samostatně vyvíjet od poloviny 50. let především ve Francii, kdy Georges Gurvitch a Jean Duvignaud píší první sociologické práce o divadle, zejména z oblasti typologie výzkumu v divadle. V dnešní době se systematicky sociologickým výzkumem divadla a v divadle zabývá několik institucí v celé Evropě. Za všechny uveďme mezinárodní organizaci FIRT/IFTR (Fédération Internationale pour la Recherche Théatral/International Federation for Theatre Research) nebo mezinárodní asociaci SIBMAS (Société Internacionale des Bibliotheques et Musees des Arts du Spectacle, International Association of Librairies and Museums of the Performing Arts). Obě instituce sdružují výzkumníky mnoha oborů, jejichž průsečíkem zkoumání je divadlo se všemi svými aspekty. Divadlo je nazíráno z mnoha úhlů pohledu, teatrologického, psychologického, filozofického, antropologického, ale také sociologického. V rámci obou institucí se konají přednášky a workshopy, ve kterých má sociologie divadla své legitimní místo. Public finances reform in the light of pre-election proposal by the Civil Democratic Party

In this work I would like to evaluate government's reform about public finances, everything what was agreed upon and voted for by the end of 2007. At the same time I would like to compare that to the original pre-election proposal by the Civic Democratic Party and government's declaration by Mirek Topolanek coalition's party. In my work I will concentrate mainly on revenue of state budget, where the most impact manifest of the fiscal reforms is seen. The main differences are consolidation of tax brackets for individuals and also changes in rates of VAT, lowering corporate taxes and introduction of ecology taxes. Even though this reform appear like the straight taxes from the pre-election program from ODS, this form is only similar in tax brackets, but not by other structural tax provisions. On the side of current expenses was government less bold. On this issue, they enforced only small differences in the transfer expenditure, where they discontinue few pre-election populist statues, even though almost no changes were done in this section so far. This proposal in pre-election for negative tax was much more bold and interesting, because here together with the tax system the state could safe money and also be a motivation for people to work. Purpose of work is outlining other necessary steps in the reform process for public finances and also in area of financing of public medical system, retirement system and changes in budgetary allocation of taxes.

The development of cross-border cooperation of the Region of South Bohemia, the Free State of Bavaria, Upper and Lower Austria with a focus on Strategy for removal of consequences of the Iron Curtain.

The author of the bachelor thesis is going to handle with the development of the Region of South Bohemia, Free State of Bavaria and Upper and Lower Austria cross-border cooperation. The thesis concerns general problematic of cross-border cooperation, its historic development in the frame of the respective timeframe, concerning period from the year 1948 up to nowadays, further the projects administrated by the Region of South Bohemia and its neighbouring regions as well as the planning projects, not yet implemented. Introduction of The Strategy for Removing The Impacts of The Iron Curtain in the separate chapter enables deeper insight into the strategy as well as into its future utilization. Moreover, the author made questionnaire research among the citizens living in the border towns of the Region of South Bohemia that might feel the impacts of the iron curtain the most. In the conclusion of the thesis the author creates conclusions from the carried research and confirms or disputed the assumption of the thesis.

In the first one we present the necessary mathematical apparatus - selected topics from combinatorics, graph theory and theory of generating functions. The second one is an introduction to cluster expan- sion and abstract polymer model. Finally, in the third chapter we show a new resummation method for partition function of hard-core repulsive abstract poly- mer model. In this resummation we make use of cancellations of terms in partition function to rewrite the sum of clusters to a sum of quilted clusters, or alternati- vely as a sum of "bunches". The methods we use in this final chapter are original and may lead to some new results. Laboratory diagnostics of micrometastases in breast cancer patients

Introduction: The presence of circulating tumor cells (CTC) in the peripheral blood has been associated with worse prognosis and early relapse in breast cancer patients. CTC determination in the peripheral blood has been considered as a liquid biopsy. The aim of this project was to analyze the presence of CTC followed by their molecular characterization with the potential use not only as a new biomarker for real-time monitoring of therapy efficacy but also as a suitable tool for patient's stratification and individualization of treatment for breast cancer. Methods: A total of 54 patients with diagnosed early breast cancer were enrolled into a prospective study. Ten millilitres of peripheral blood were sequentially collected to test for the presence and characterization of CTC during the follow-up of patients. CTC isolation and detection was performed by AdnaTest BreastCancer™ (AdnaGen AG, Germany), which is based on the detection of EpCAM, HER2 and MUC1 specific transcripts in enriched CTC- lysates. cDNA from isolated CTC has been further used for newly optimized qPCR assays for breast tumor and therapy resistance associated genes: TOP1, TOP2A, CSTD, ST6GAL, KRT19 and reference gene actin. qPCR results have been analyzed by Genex software (MultiD Analysis). Hypertensive organ injury on the basic of 5/6 nephrectomy in REN2 transgenic rats

Hypertension is a major determinant of rate of progression of CKD. Treatment of systemic hypertension is now generally accepted as the most important clinical strategy to slow the progression of CKD to ESRD. Renin-angiotensin (RAS) and endothelin (ET) systems are important contributors to the pathogenesis of hypertension. The interaction between the RAS and ET systems also plays a crucial role in the development of hypertension- associated end-organ damage. Antihypertensive drugs that block the renin-angiotensin system (RAS) are now believed to provide class- specific renoprotective actions beyond their blood pressure lowering effects. Our first study was performed to compare the organ-protective effects of a RAS-dependent and a RAS-independent antihypertensive therapy in heterozygous (mRen-2)27 transgenic rats (TGR) after 5/6 renal ablation (5/6 NX). Normotensive transgene-negative Hannover Sprague-Dawley (HanSD) rats after 5/6 NX served as controls. RAS-dependent and -independent antihypertensive therapies induced equal reductions of BP and almost identical cardio- and renoprotective effects on the progression of CKD after 5/6 NX, in both HanSD and TGR. These findings suggest that the cardio- and reno-protection, at least in rats after 5/6 NX, is mainly mediated through BP- dependent mechanisms....

High resolution computed tomography - its role and position in radiodiagnostics

In the introduction to my bachelor´s thesis, I have described the historical background of HRCT, its origins, and evolution. The historical part includes information about the development of CT equipment and the principles of diagnostics using the instruments that preceded HRCT. In the following part, I have described the principle of how HRCT works and the conditions that are needed for HRCT examination. I focused on lung examination and partly also on the examination of temporal bone. The objective of my thesis is to determine the frequency of CT and HRCT application, for the purposes of which I compared two leasing hospitals in the South Bohemian region. I issued from the hypothesis of a higher frequency of HRCT use. In my bachelor´s work I used foreign literature and sources and also the method of comparing data from two hospitals. The preferred method was the quantitative one, used for numerical statistics in the theoretical part; for the practical part I used the mentioned comparison of the number of examinations performed at two hospitals in South Bohemia. When confronting the two hospitals, a significantly higher frequency of HRCT use was found in Nemocnice Tábor a.s. This fact is explained by the use of Aquilion 16, which is a lower ? quality appliance, and Aquilion 64 in České Budějovice, as well as by the monitoring of patients with fibrosing processes (so ? called cordarone lung, etc.), and patients with unclear small lesions appearing on summing images when only HRCT examination was performed. HRCT is also performed as a part of routine examination of the lungs and mediastinum if the indication requires it, while check ? up examinations run only under the HRCT algorithm. In conclusion it can be stated that the difference between the examinations in hospital České Budějovice a.s. and Nemocnice Tábor a.s. is based on the difference between their equipment and also the slightly different approach to small lesions in the pulmonary parenchyma.The hypothesis that the HRCT examination is used more frequently than the common CT examination of the lungs and mediastinum was not confirmed, which results from the fact that the number od patients needing the examination of just small or larger changes in the pulmonary parenchyma is substantially lower than the number of patients with indicated classical examination of the lungs and mediastinum.

Anti-oxidative stress response in Drosophila melanogaster - The role of adipokinetic hormone and adenosine

In this thesis, the phenomena of the oxidative stress (OS) and anti oxidative stress responses in insects are described in a comprehensive review, and the outcomes of the experimental work are presented. The focus of the work was on defence reactions and their putative control by the adipokinetic hormone (AKH) and adenosine in the Drosophila melanogaster model. For this purpose, we studied the effect of the paraquat (oxidative stressor) treatment on adult flies and larvae carrying mutations in Akh (Akh1) and adenosine receptor (AdoR1) genes, and in both these genes together (Akh1 AdoR1 double mutant). The initial mortality tests revealed the double mutant Akh1 AdoR1 was more sensitive to OS than either of the single mutants. The AKH synthesis under the OS condition seems to be out of the gene expression control, since the increase of an AKH amount in CNS was not linked with the stimulation of Akh gene expression after a paraquat treatment. Further, the gene expression of the antioxidant enzyme glutathione S transferase D1 (GstD1) increased rapidly with OS, though the enzyme activity increased negligibly regardless of both the OS and mutations. Interestingly, the relative expression of GstD1 gene was minimal in the double Akh1 AdoR1 mutant; thus, it was concluded that both AKH and adenosine are employed in the GstD1 gene expression control. Similarly, AKH and adenosine seem to act in tandem in glutathione (GSH) regeneration, since the GSH level was significantly lower in all untreated deficient flies with the maximal effect in the Akh1 AdoR1 double mutant; accordingly, the reduction in the GSH level was enhanced by paraquat treatment. Altogether, the important roles of both AKH and adenosine in the anti oxidative stress response in D. melanogaster were demonstrated.

Output-Inflation Trade-off After a Quarter of a Century of Inflation Targeting

This thesis estimates the treatment effect of inflation targeting adoption on inflation, inflation variability, output, and output variability for 25 explicit inflation targeting countries. I implement the propensity score matching methodology that takes into account the problems of non-experimental nature, such as selection bias or selection on observable, and allows me to effectively mimic properties of randomized experiment and compute unbiased treatment effect estimates. I introduce a variety of propensity score matching methods that were recently developed in the treatment effect literature, including Nearest Neighbor, Radius matching, Kernel matching, and Inverse Probability Weighting. The results indicate that both industrial and developing inflation targeting countries exhibit lower inflation levels and at the same time higher output growth than non-targeting countries. The estimates are however in most cases statistically insignificant. Moreover, it appears that both industrial and developing countries achieve combination of lower inflation variability and output variability compared to non-targeting countries. Nonetheless, majority of the estimates are again statistically insignificant. The results are to a small extent sensitive to the choice of propensity score matching method. Radius matching with tight calipers (r=0.005, r=0.001) tends to provide the most reliable estimates. Balancing properties of the models are reasonable and compared to the previous research the standardised biases are quantitatively better.
