The measurement of total alkaloids in tobacco
Pipková, Renata ; Vránová, Dana (referee) ; Omelková, Jiřina (advisor)
The aim of this diploma thesis is the assay of total alkaloids in different tobacco types. To measure the content of total alkaloids are utilized types of tobacco used for preparation of tobacco blends and consecutive production of cigarettes. The samples of analyzed tobacco include various countries of cultivation and different crop years. There is described tobacco as a plant, in it contained alkaloids and the utilization of tobacco in the theoretical part. In more detail it deals with the production and construction of cigarettes which is closely related to the content monitoring of total alkaloids in tobacco. The contents of total alkaloids are determined by the continuous flow analyzer- SKALAR. On this spectrophotometer are set the contens of reducing sugars as well. The comparison of total alkaloids contents and reducing sugars from the perspective of single tobacco types, countries of cultivation and crop years is clearly summarized in the experimental part of the thesis that was performed in the company Philip Morris ČR, a.s.

Význam rostlinných proteinů z podrodiny ABCB pro transport auxinu
Kubeš, Martin ; Zažímalová, Eva (advisor) ; Hejátko, Jan (referee) ; Reinöhl, Vilém (referee)
Polar auxin transport provides essential directional and positional information for many developmental processes in plants. At the cellular level, it is realized by both passive diffusion and the active transport through the membrane proteins - AUX1/LAXes, PINs and ABCBs. The aim of this thesis was to characterize the role of ABCB1, ABCB4 and ABCB19 proteins in polar auxin transport using transformed tobacco BY-2 cell lines. It was shown that the plasma membrane (PM) localization of the ABCB1, 4 and 19 is not polar. The ABCB4 was also more stable on PM after the treatment with auxin influx inhibitors; making use of ABCB4-cell line helped to uncover new characteristics of markers of endocytosis - the FM- dyes. The induction of ABCB19 has led to a decrease in 3 H-NAA accumulation with characteristic auxin starvation phenotype, similar to PIN7 overexpressing cell line, that could be rescued in case of PIN7 cell line by application of the auxin efflux inhibitor NPA; however, the accumulation of auxin in ABCB19-overexpressing cell line was less sensitive to NPA and the rescue of the auxin starvation phenotype was ineffective. Importantly, unique property of the ABCB4 was demonstrated: It displayed dual, auxin-concentration-dependent auxin transport activity in Arabidopsis roots, tobacco BY-2 and yeast cells. The...
Public attitudes towards substance use and drug addiction - April/May 2023
Spurný, Martin
In a survey conducted during April and May 2023, the The Public Opinion Research Centre (CVVM) asked a set of questions about public attitudes towards substance use. Respondents over the age of 15 rated the extent to which they considered it acceptable to consume certain addictive substances, specifically tobacco, alcohol, painkillers, sleeping or tranquillisers, cannabis drugs (e.g. marijuana, hashish) and drugs such as ecstasy, methamphetamine and heroin. They commented on selected statements about the problem of drug addiction, whether drug use is a problem in the Czech Republic as a whole or in their place of residence, and whether the relevant institutions in the Czech Republic deal adequately with the problem of drug use.

Prokešová, Miroslava ; Kulhánek, Adam (advisor) ; Kočvarová, Linda (referee)
Introduction: A relatively new trend that has been gaining popularity in recent years is heated tobacco, which has been on the Czech market since 2017. Although the number of users of heated tobacco products in the Czech Republic is increasing, the phenomenon is still relatively under-mapped. There is also very little information on the gender specificities of the use of heated tobacco. Therefore, this study focuses on mapping and describing the patterns of use of heated tobacco products among women who have switched from using conventional cigarettes to heated tobacco products. Aims: The main aim of the research is to map the patterns of use of heated tobacco products among chosen women in the adult population of tobacco users in the Czech Republic. The sub-objectives are to describe the nature of heated tobacco product, find out what motivated female users to switch to heated tobacco products. And what are the typical situations when women use the device. Sample: the research sample was selected by simple intentional (purposive) sampling method, combined with snowball sampling method. The final sample consisted of 7 female respondents. The mean age was 25,7 years (youngest 21 years and oldest 34 years). Methods: The research was conducted using a qualitative method. Data was collected by...

The decrease in alcohol consumption and smoking in the Czech Republic between the years 2010 and 2018 in relation to the family environment
Krásná, Anna ; Soukup, Petr (advisor) ; Petrúšek, Ivan (referee)
Since 2010, sociologists have noted that teenagers drink less alcohol and smoke a smaller number of cigarettes each year than in previous years. This paper analyses the association between family background and the decline in alcohol and cigarette use in the Czech Republic between 2010 and 2018. The data are from the international HBSC (Health Behaviour in School-aged Children) study, from the years 2010, 2014, and 2018. The bachelor thesis only discusses cases from the Czech Republic, the data have thousands of cases, so they are robust enough to perform quantitative analyses. The analysis focuses only on alcohol and conventional cigarettes as the two most common addictive substances consumed by adolescents. Family background is examined in terms of parental presence in the household, ease of communication between parent and child, and family wealth. Secondary analysis showed an association of family wealth with the amount of alcohol consumed and cigarettes smoked. Children from wealthier families drink alcohol more often than children from the opposite end of the spectrum, and conversely, children from poorer families smoke more cigarettes. The data does not have the potential to observe causality, only correlations could be observed. The main conclusion of the paper is that the decline in...
Spatial Differentiation of Tobacco Smoking Rates in
TOMÁŠKOVÁ, Tereza
The bachelor thesis examines the spatial differentiation of tobacco smoking rates in the world at the national level. The thesis is divided into theoretical and practical parts. It aims to analyse tobacco use rates in different countries of the world and to produce more general outcomes at the macro-regional level based on the results obtained. The sociopathological phenomenon under study is analyzed using nine socio-economic indicators, which are divided into three groups - economic, social and other. Using correlation analysis of the selected determinants, the dependence of each indicator with the proportion of adult smokers is observed. With the help of the created map outputs, some spatial differences, certain specifics and regularities of the observed phenomenon are clarified, which in the conclusion of the work confirm or refute the established hypotheses.
Effect of Nicotine Products´ Usage on Human Health.
DLOUHÝ, Aleš
This bachelor thesis deals with products that contain nicotine with a focus on electronic cigarettes. The aim of the work was to find out whether there are changes in the behavior and level of their use during the transition of users from classic tobacco products to electronic cigarettes. The work is divided into two parts. The theoretical part deals with the general history of tobacco. Furthermore, individual types of tobacco products are characterized here, especially products heating tobacco. This chapter is followed by a section on the treatment of nicotine addiction, including nicotine replacement therapy and pharmacological treatment. Much of the work is devoted to electronic cigarettes, their development, the principle of operation, division and fill into them. The end of the theoretical part focuses additive components, addiction its division and withdrawal symptoms. The last chapter deals with nicotine. It characterizes its chemical-physical properties and describes its effect on the human body. The aim of the second practical part of the work was based on a questionnaire, which contained 21 questions, the collection of answers and their evaluation. The questionnaire was completed by users of electronic cigarettes who had experience with tobacco products. A total of 435 respondents took part in the questionnaire survey. The results of the questionnaire survey were evaluated using the implemented tools of the Survio questionnaire platform, while important results were displayed graphically. The results of the research show that the behavior of electronic cigarettes is changing behavior and increasing the frequency of use compared to conventional cigarettes. This in turn causes a deepening of nicotine addiction. The practical benefit of this work should primarily consist in a comprehensive overview of all aspects that play a role in the transition of users to electronic cigarettes. Furthermore, it could be used as a basis for designing solutions that would facilitate the transition to electronic cigarettes and reduce the risk of increased nicotine dependence.
