The role of mitochondrial metabolism in initiation and adaptation to hypoxic conditions.

We can meet pathological hypoxia in the cases of hearth attack, ischemic stroke, but also during tumor invasion, thanks to insufficient angiogenesis. The activation of HIF- 1 factor during hypoxic conditions is crucial for the cell survival. This factor modulates energetic metabolism in favor of fast progressing glycolysis (with the contribution of glutaminolysis) which provides to cell enough ATP and "building blocks", while suppressing Krebs cycle and respiration because of shortage of oxygen. The thesis studies energetic metabolism of HepG2 cells (derived from liver carcinoma) which are cultivated in the media with various energetic substrates, i. e. glucose or galactose (always together with glutamine and pyruvate) under the hypoxic conditions (5% O2). HepG2 cells use particularly oxidative metabolism for ATP and "building blocks" production under the normoxic conditions while hypoxic environment causes metabolic shift in glycemic condition. Interestingly, cells cultured in galactose (glutamine) didn't switch the energy metabolism from oxidative to aerobic glycolysis such as cells cultivated in glucose, although HIF-1 factor was stabilized. Severe lightning activity over the Czech republic

This work comprises an introduction to study of lightning and presents data about daily amount of lightning flashes over the Czech Republic (CR) in period 2002-2012. For selected days in this period, there are introduced and examined some meteorological characteristics. There are presented an evolution of lightning discharges and contemporary options for lightning detection which contain ground-based systems and satellites systems. Main advantage of ground-based systems are location accuracy <1 km and additional information which could be provided about every single stroke which is e. g. polarity or peak current. The benefit of satellite lightning detection is the large covera- ge which is useful for a global view of lightning activity on the Earth. After that, the general climatology of lightning in the world is introduced. The land-ocean contrast in lightning activity is presented (lightning activity over land is up to two orders higher than over the ocean). There is also information about maximum amount of flashes over equatorial Africa. The hypothesis explaining the land-ocean contrast in lightning activity and African maximum are included. Studies on structure and biological functions of NKR-P1 receptors

Natural killer (NK) cells play a significant role in the detection and destruction of virally infected and tumor cells. The NKR-P1 receptors regulate NK cell function by an alternative missing-self recognition system. Although the NKR-P1 receptors were among the first surface NK receptors identified on rodent NK cells more than 20 years ago, there is still very little known about their biological function and their physiological ligands. Furthermore, no three-dimensional structure of any of the NKR-P1 family receptors has been published so far. To understand the functional architecture of mouse NKR-P1 receptors, we developed a simple and efficient protocol providing large amounts of pure soluble NKR-P1 proteins. The crystal structure of mouse NKR-P1A, determined at 1.7 A resolution, is the first structure of a representative of the NKR-P1 family. Crystal structure is formed by a compact C-type lectin-like domain and an extended loop that participates in domain swapping. A potential role of the swapped loop has been suggested in natural ligand binding by in silico studies. Soviet Near East Policy 1917 - 1921 between Revolution and Pragmatism

This work addresses the beginnings of Soviet policy towards the Middle Eastern countries Afghanistan, Persia, and the Ottoman Empire/Turkey between 1917 and 1921. The author addresses the manner in which Soviet leaders formed their perceptions of the Middle East and the place and role assigned to the countries in question by Soviet leaders. At the same time, the work investigates how real Soviet policy towards the countries mentioned above was formulated, what the aims and motivations of the policy were, and the factors which influenced its formulation and realization.
Study on conformational changes in proteins using mass spectrometry.

Some proteins and enzymes require presence of their specific ligand, cofaktor or prosthetic group for their activity. Binding of this specific molecule causes conformational changes, which permits to perform their function. In some occasions the identification of conformational changes is difficult. Using chemical cross-linking coupled with mass spectrometry perform complex tool for searching and low resolution visualization of this changes. The aim of this thesis is study of conformational changes induced by binding of calcium ion to calmodulin protein molecule. Calmodulin is a secondary intermediate messenger, which can interact with various proteins. This feature associates with wide dynamical range of calmodulin. Thus calmodulin is the suitable target for identifying conformational changes. After reaction of protein with chemical cross-linkers with different arm length (DSG and DSS) were products of reaction digested by trypsine. Formed linked peptides were separated by high-performance liquid chromatography and analysed followed mass spectrometry. Seven unique intramolecular cross-links were identified. Using isotope unlabeled cross-link reagents in the presence of Ca2+ in combination with using isotope labeled reagents in calcium free conditions we quantified formed lysine-lysine cross-links....

75 English summary Lien of the share in a limited liability company The main goal of my thesis "Lien of the share in a limited liability company" is to analyze provisions of the new Czech Civil Code and Corporation Act, which were passed in the spring of 2012. Both statutes come into effect on 1 January 2014. At first I briefly describe the historical development of this issue since 1906, when first a statute concerning the limited liability company was passed in our country. Then I describe the main issues in the provisions of Czech commercial Code. Current legislation concerning the pawn of the share is a very brief one. The main problem, which I introduce, is that the legal possibility of pawning the share depends on the legal possibility of transferring the share. This provision neglects the fact, that the commercial Code defines two types of transfer of the share, that is to another shareholder or to a non-shareholder, i.e. to a person outside the company. The new Czech Civil Code contains a comprehensive legislation of the pawn. Its main assets are wide contractual freedom and protection of the lien debtor. Proposal for Development of the Manager

The aim of my bachelor thesis is to make a proposal how a manager should develop himself, what abilities are necessary to emphasise and what the key features of the right manager are. The theoretical part deals with the manager issues and manager development. It solves and tries to answer the question how the manager should develop himself and what he should do for his personal development. The result of this thesis is my own proposal and recommendation for a young ambitious manager. This proposal has been based on the combination of qualitative and quantitative research.

The Bachelor’s thesis deals with the optimization and the designof the e-shop communication mix. The thesis is divided into three main chapters. The firts one is dedicated to the theoretical knowledge of the area od marketing, the second one contains basic data and analysis performed. In the third chapter, the improvement of the existing communication mix of the company is suggested, based on the theoretical knowledge and subsequent analysis of the problems and the actual condition. It includes the suggestion of the new promotional methods which will lead to a better communication with the public and the consequent increase of the number of the customers.
