Karel Caslavsky's documentary aktivities on TV

The submitted paper deals with the work of the significant film historian Mr. Karel Caslavsky and focuses on his auctorial series Hledání ztraceného času (Searching the Lost Time). In the first part, I will briefly describe the early phase of his life and the beginning of his professional work in the Czechoslovak Television. Further, I will write about the show Videostop in which he was recognized by the public for the first time. The main intention is the detailed analysis of the series Hledání ztraceného času which will be supported by the solitaire episodes, mini-series and, especially, two unique cycles Co viděly americké kamery v Čechách (What the US Cameras saw in Bohemia) and Vltava v obrazech (The Vltava River in Pictures). I will focus on the preparation process, the work with the archive materials, the way of realization or the importance of his work for the Czech public as such. There are interviews with the close colleagues of Mr Caslavsky as part of the study.

Image analysis of fluorescently labeled nuclear structures: DNA replication in focus

In this thesis, we expanded the knowledge about some aspects of DNA replication in human cell nucleus with the help of microscopic techniques and advanced mathematical approaches. DNA replication was studied from several perspectives: Fluorescence detection of newborn DNA on the stretched DNA fibers, ultrastructural mappings of individual replication domains, fluorescence analysis of replication foci re-distribution in early S-phase and cold-dependent immunodetection of replication-coupled chromatin modulation. We started with measurements of average speed of a replication fork movement during S- phase. In agreement with previous studies on DNA spreads with radioactive labeling, the replication speed was two to three times slower in early S-phase than in late S-phase. Moreover, the exogenous dNTP supply accelerated the replication in the early S-phase while additional dNTPs had no effect on the speed of replication forks in late S-phase. Therefore the availability of dNTPs seems to be a rate-limiting factor for replication speed at the beginning of DNA synthesis. We then focused on in situ replication pattern at the EM level. The aim of the study was to create a new animal model of schizophrenia based on a neurodevelopmental hypothesis. The central idea of the project has been that a transient insult at early postnatal age will reveal itself in the form of delayed distinct behavioral changes that can be related to schizophrenia. The experimental design used intracerebroventricular (icv) infusion of N-acetyl-L-aspartyl-L-glutamate (NAAG) to rats at postnatal day 12, followed by combination of histology and quantitative morphology in brain tissue obtained within 24 and 96 hours of the NAAG administration and used to investigate possible neurodegeneration. Finally, a battery of behavioural tests was performed several weeks later (late adolescence/early adulthood). NAAG is the most abundant neuropeptide and interacts with the active site of metabotropic glutamate receptors (mGluR II), however, it is also an agonist at NMDA receptors. Neonatal icv infusion of NAAG resulted in detectable damaged neurones in gyrus dentatus. The damage appeared greater at 24 hours, as compared to 96 hours, after the infusion. The presence of damaged neurons was easily demonstrated by Nissl stain, Fluoro Jade В staining combined with Hoechst 33342 and by TUNEL technique. Cluster analysis of more dimensional data by a neural network

The topic of the present thesis is an analysis of a sample data archive containing measured values of real and reactive power. The measurement in question took place in late 2006 and early 2007 using MEg40 recording measurement devices disposed in a station for transforming high voltage to low voltage in the Pražská energetika distribution network. The procedure of processing measured values, the preparation thereof for a subsequent processing by a neural network, and a final statistical evaluation of determined individual clusters -- typical daily take-off diagrams -- will be described. The results of the present thesis may be applied in the making of predictions of electrical energy consumption at a particular transformer station.

Study of Constructed Treatment Wetland Reconstruction for Dražovice Village

The constructed wetlands, which have been being created in the Czech Republic since early 90s, are designed so they do not allow successful removal of ammonia nitrogen. Almost all of these 300 registered constructed wetlands suffer from the colmatage which leads to the lowest effectiveness of treatment in nearly all parameters. The main effort of the Institute of landscape water management is to get rid of bad reputation and to clarify the reason that leads to unsatisfactory conditions of constructed wetlands. The diploma thesis focuses on the selected constructed wetland in Dražovice (850 EO), which removes ammonia nitrogen with the effect just below the limit values in the long term. The pressure of the Morava river basin and Department of environment (Building Authority in Bučovice) has led to the necessity of taking measures leading to the increase of treatment efficiency especially ammonia nitrogen. The diploma thesis includes both, actual project documentation concentrating on the adjustment of watercourse to nitrify ammonia nitrogen, and effective choice of suitable filtration material supported by testing method in situ. The practical part of the thesis deals with the implementation of the technical solution according to the project documentation. The adjustment of the constructed wetland is now nearly before completion. Last adjustments will be taking place during the spring 2017, the final inspection as well as the evaluation of the reconstruction will take place at the end of this year.

Cultural and Historical Specificities of the South Bohemian Region

This thesis is focused on culturally historical specifics of South Bohemian Region. To get the aim of the thesis there is undertaken a contents study of documents as well as questionnaire survey of proprietors of historic sights. Contents study of documents is focused on documents of South Bohemian Region in the field of protection and support of culturally historical potential on region´s area. Questionnaire survey gets information about financing and promotion of cultural sights in the region of South Bohemia from the view of the sights proprietors. Undertaken study of questioned spheres provides requested information. Whole thesis is targeted on culturally historical potential of South Bohemian Region, as well as their financing and promotion and includes final solution proposals of those matters.

Eating disorders in adolescence

The bachelor degree work deals with disorders food intake, in particular, mental anorexia and mental bulimia in maturing period. The theoretical part is aimed at maturing problems and food intake disorders generally. There is characteristics and division of maturing period into early and late adolescence including psychological and physical changes which are typical for this period. Then food intake disorders, their causes, effects and their possible treatment are specified. The aim of the practical part is the evaluation of today youth´s awareness of food income disorders, then the evaluation of their appropriate reaction ability in case such disorders occur in their class. At the end of the work primary prevention in this field in our system of education is mentioned.

Effect of drought stress on selected winter wheat yield formation components within pot and field experiimental design

The object of this study was to find out what is the behaviour of the same winter wheat variety (Bohemia) plants cultivated within pot and field experiment. Therefore, the main aim of this study was to verify (based on the pot experiment results) whether the pot experiment (that is limited by the soil area) does not substantially affect plant reactions. The pot experiment was carried out in growth chambers where daily temperature course, relative humidity (RH) and photosynthetically active radiation (PAR) were set via protocols. The pots were exposed to the drought stress for 14 days with the daily maximum temperature 26 °C from noon to 2 p.m. The pots were split into 2 groups: (1) Dry where the soil moisture within pots were maintained below 30% of the maximum water holding capacity, (2) Wet where the soil moisture did not decrease below 70% of the maximum water holding capacity. The plants within Wet variant were considered as a control group. The pots were placed onto the concrete floor of a vegetation hall (where the plants were exposed to the weather conditions) prior and after stress regime exposition. The field experiment was conducted within experimental station in Bystřice nad Pernštejnem belonging to the Bohemian-Moravian Highlands in the Czech Republic. The drought stress was established through the transparent roofs installed above plants’ tops level in the field. The control experimental plot without roofs was nearby there as well. The plants were harvested when the full maturity was reached and the selected yield formation components were evaluated.

Settlement area from the Late Bronze Age in Hluboka nad Vltavou

This thesis summarizes results of comprehensive processing two implemented rescue excavations settlement area from the Late Bronze Age in the cadastre town Hluboká nad Vltavou (Křesín). Performed analysis selected ceramic assemblage and other artefacts, together with the evaluation all settlement objects, provided the results for dating site and interpretation of its purpose. Location was also compared with other sites in south Bohemia and set in a settlement framework of microregion Českobudějovicko.

Three year old children in schools working according to the principles of Marie Montessori

The objective of the bachelor thesis is to ascertain what conditions and environments are the most favourable for children who begin to attend preschool establishments before reaching the third year of age or even at two years of age, which has currently been a growing trend that requires looking for appropriate methods allowing their optimal development. The thesis also surveys the approach to the children of the above stated age bracket, and monitors the educational conditions and readiness of the environment in Montessori establishments. The thesis is divided into a theoretical and practical part. The theoretical part describes the development specifics of the late toddler age and the early preschool age, and the conditions of education. The conclusion of the theoretical part explains basic principles of Marie Montessori's method. The practical part describes the research methods used in the thesis, namely observation in Montessori establishments, interviews with teachers, and a survey.
