The effect of biologicaly active substances on the structure and properties of collagenous substrates

Diplomová práce se zabývá přípravou 3D porézních kolagenových skafoldů metodou lyofilizace a jejich modifikací bioaktivními látkami. K modifikaci byly použity přírodní polysacharidy – chitosan, vápenatá oxidovaná celulóza a chitin/chitosan-glukanový komplex. Mechanické vlastnosti skafoldů byly upraveny síťováním pomocí karbodiimidů. Růstové faktory byly dodány formou destičkového lyzátu. Byl zkoumán vliv biologicky aktivních aditiv, siťovacího činidla a obohacení růstovými faktory na vlastnosti připravených skaffoldů a jejich bioaktivitu v tkáních živých organismů. Konkrétně byly studovány morfologické vlastnosti, struktura, porozita, botnání, stabilita, chemické složení, teplota denaturace a biologické vlastnosti. K charakterizaci byly použity metody rastrovací elektronová mikroskopie, infračervená spektroskopie, diferenční kompenzační kalorimetrie a konfokální mikroskop. Připravené kolagenové substráty obohaceny bioaktivním aditivem a destičkovým lyzátem mohou být využity v biomedicíně jako skafoldy pro růst buněk v systémech s nízkou mechanickou zátěží.

Prof. RNDr. Veronika Opletalová, Ph.D. Title of Thesis: Derivatives of Rhodanine as Potential Antifungal Drugs Rhodanine (2-thioxo-1,3-thiazolidin-4-one) forms the basic skeleton of many biologically active substances and potential drugs. Antifungal properties of rhodanine derivatives have been studied since the early 1950s. Theoretical part of the thesis deals with mycoses and drugs that are currently used to treat them. The aim of the experimental work was to find suitable reaction conditions for Mannich reaction of (5Z)-5-(subst.)benzylidenes-2-thioxo-1,3-thiazolidin-4-ones with various amines. The attempts to prepare Mannich bases using diethylamine as the basic component were unsuccessful. (5Z)-5-benzylidene-3-(morpholin-4-yl-methyl)-2-thioxo-1,3-thiazolidin- 4-one was successfully obtained by treating (5Z)-5-benzylidene-2-thioxo-1,3-thiazolidin- 4-one with formaldehyde and morpholine in dimethylformamide. This compound was reported previously and exhibited antibacterial activity and acceptable toxicity. Neopterin is used as a marker of immune system activation and inflammatory diseases. Its early detection in urine or plasma may indicate a pathological immune activity. Elevated concentration of neopterin is described in viral or bacterial diseases, in autoimmune diseases in HIV infection or in malignant tumors. 7,8 - dihydroneopterin is able to prevent proteins and lipoproteins from oxidative damage. Hydrophilic Interaction Liquid Chromatography (HILIC) is a chromatographic method that can be used to improve retention of very polar compounds. Education and development of children in lesbian family

The thesis deals with topic of lesbian families and specifics of children from these families. The theoretical part of the work is supported by studies from the USA and the Great Britain in particular. This part is focused on definition of homoparental family and its types, specifics of upbringing and relations within. It also deals with the topic of coming-out. It gives account of the family roles - biological mother, biological father, social mother / stepmother. The second half of the theoretical part describes psychological, gender and social development of the children from lesbian families. There are also mentioned some children's feelings and views of their nontraditional family and homosexuality of their parents. The thesis also deals with lesbian adoptive families. The research presented in the empirical part is based on individual interviews and projective techniques. The study group comprises 10 children between the age 4 and 15. In the study group there are children from previous heterosexual relationship as well as children from planned lesbian families. RNDr. Dagmar Solichová, Ph.D. Diploma Thesis Title: Comparison of extraction procedures for HPLC determination of liposolubile vitamines in serum In this thesis were compared extraction methods, solid phase extraction (SPE) and liquid liquid extraction (LLE) used for separation of human serum fat soluble vitamins: A (retinol), E (α-tocopherol), D2 (ergocalcipherol), D3 (cholecalcipherol), 25- (OH)D3 (calcidiol). Vitamins were then determinated by high performace liquid chromatography (HPLC). As a baseline method used the procedure that was developed and partially validated the thesis: Application of the SPE technology and monolithic columns in HPLC analysis of biologically active substances (Horčičková 2009). This procedure was further optimized in order to increase the extraction of target analytes 25(OH)D3, D2 and D3 and shortening the preanalytical phase. Characterization of Modern HPLC Columns and Their Application Potential

(EN) The aim of the dissertation thesis is characterization of modern HPLC columns from the point of their interaction possibilities and demonstration of their application potential. The first part of the thesis is focused on alternative reversed-phase HPLC columns based on zirconium dioxide. These packings offer excellent chemical stability and additional interactions that can be helpful in the development of analytical methods. A detailed study of the chromatographic behaviour of biologically active nonapeptides as model analytes confirmed the substantial effect of mobile phase composition on retention mechanism. Consequently, HPLC separation systems with zirconia-based polystyrene column were characterized by distinct approaches that allowed recognition of the interactions participating in the separation process. Empirically based chromatographic tests evaluated the fundamental properties of the system - hydrophobicity and polarity. The complex model of linear free energy relationship described the prevailing interactions in different separation systems. Application of a set of basic compounds revealed the contribution of ion-exchange interactions participating in the separation systems with zirconia-based column. Vanda Neradiová Biologically active substances in the genus of Rheum species used in traditional Asian medicine Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Specialist in laboratory methods Background The goal of this study was to determine content substances and their quantity in two different species of genus Rheum and compare to what extent these two species differ in a number of content substances. Another criterion by which these species were compared was their place of origin. I have compared a species growing in its natural environment in the Himalayas with a second species growing in the Czech Republic. Methods For determination of content substances and their quantitative determination high - performance liquid chromatography with UV and fluorescent detection was used. Samples were compared with retention factors and retention times of standards. Furthermore, isolation and identification of probable physcion was carried out using mass spectrometry. Results By qualitative analysis of antraquinones, aloe - emodin, emodin, chrysophanol, physcion and rhein were determined and by analysis of stilbenes piceatannol and resveratrol were determined. Negative body image and eating disorders

The aim of this thesis is finding out if and how much negative body image influences eating disorders. The body image is influenced by the contemporary ideal of beauty, which was historically formed. The influence of the contemporary cult of a skinny body has a biological argument, that slimmer people are healthier. Models, who are representing this ideal are often accused to be the reason of increase in frequency of present day diseases, like eating disorders. The most known eating disorders are anorexia nervosa and bulimia, but there are also other diseases which are not yet described in the International Classification of Diseases -- orthorexia, bigorexia and drunkorexia. What really causes these diseases? The essence of the practical part is, by using the interview, to find out risk factors for women and girls suffering from eating disorders, as well as satisfaction with their bodies and the impact of modelling. The conclusion presents results of a research, which shows whether or not there are biological, individual, family and sociocultural factors, which include the influence of the media as well, present in respondent's answers.
