Reference gene selection for mRNA quantification in Haemonchus contortus
Růžičková, Michaela ; Matoušková, Petra (advisor) ; Zemanová, Lucie (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Michaela Růžičková Supervisor: Ing. Petra Matoušková, Ph.D. Title of diploma thesis: Reference gene selection for mRNA quantification in Haemonchus contortus Haemonchus contortus is a parasitic nematode, whose multiresistance to ant- helmintics means global problem, threatening mostly small ruminants farming. Genom and transcriptom have been published in 2013, allowing gene expression studies to be conducted. Reference genes for this studies have not been validated yet. Use of suitable reference genes is essential for accurate normalization of gene expression levels. Aim of this work was to identify and validate potential reference genes for gene expression studies in H. contortus adults. Eleven genes were chosen, stability of their expression was assessed in males and females of two H. contortus strains, one drug-susceptible (ISE) and one multi-drug-resistant (WR). Total RNA was extracted and reverse trancribed to cDNA. cDNA was diluted and analyzed using quantitative real-time PCR with SYBR Green I detection. Expression stability was evaluated by computer programs BestKeeper, geNorm, NormFinder and the comparative CT method. Ncbp, ama, sodc, gapdh and farb were found to be most stable...

Analysis of SAP NetWeaver Business Intelligence 2004s
Michna, Petr ; Novotný, Ota (advisor) ; Ing. Daniel Trhoň, Ph.D., MBA, (referee)
This diploma thesis deals with the analysis and evaluation of one of the most important Business Intelligence platform in the BI market -- SAP NetWeaver Business Intelligence 2004s. The main objective of the analysis is to provide reader with very comprehensive theoretical and practical platform overview that is based among others on valuable implementation of SAP NetWeaver BI sample application. The sample application not only allows better understanding of the platform, but also provides significant inputs to evaluation final phase. This evaluation is based on a metric system, which is defined as a part of this thesis as well. The evaluation result is then, apart from the quantitative quality interpretation of analyzed platform, included in a quality comparison (some areas) with BI platforms Oracle and Open Source Pentaho, whose evaluation and comparison have been made in diploma thesis [VÁLEK, 2008]. At the end of this thesis is then provided an overview of SAP NetWeaver Business Intelligence 2004s strengths and weaknesses.

Generating meaning of popular texts demonstrated on the czech television serial Místo nahoře
Fričová, Michaela ; Reifová, Irena (advisor) ; Bednařík, Petr (referee)
The starting point of this piece of work is proposition that television and its texts are an agent of the dominant ideology in society. Through the princip of hegemony the rulling class imposes its meanings and values on the subordinated classes. Hegemony is based on allowing resistent and alternative meanings which are nevertheless articulated in terms of the dominant ideology and are subsequently incorporated into the existing system. This thesis then follows John Fiske's understanding of popular culture and activated texts . These are texts which do not imp lose one limited meani ng on its readers, but which allow its readers by means of various strategies to interpret the text with regards to their social position. The second part of this work includes semiological analysis of a particular product of popular culture - the television serial Misto nahore. The emphasis is put on the process of encoding - how the dominant meanings and values are encoded and implied in the text. Besides, the attention is directed towards the 'opening' strategies of the text. This section also includes an analysis of the secondary texts (newspaper and magazine articles ) and terciary texts (viewers' reacti ons and opinions on the web). I examine whether the sec ondary texts promote dom inant or resistant reading strategies,...

Multifunctional building "Slatina"
Pifková, Erika ; Herman, Adolf (referee) ; Bantová, Sylva (advisor)
The subject of this thesis is to design a multifunctional building. The building object is located on the southeast outskirts of Brno, in the city of Slatina. The land chosen for the building is situated in the cadastral area of Slatina. The building has five floors without basement. Multifunctional house is suitable for housing and for commercial purposes. The house is projected with 13 apartments, with café – opening hours to 10 PM, healthy food store and drugstore. There are apartments with 1 bedroom, 2 bedrooms and 3 bedrooms. Each apartment is complemented by a balcony and a cellar. The main facade is located on the west side of the property where it is allowed access to the shops and cafe. Entry into the living area is on the eastern side of the property. Perimeter walls of the building will be made of sand-lime bricks KM Beta SENDWIX 5DF-LP. The ceilings are designed as monolithic. The house is covered by a flat single-roof, part of the roof over the 4th floor will be designed as a vegetation roof.

Accelerated Neural Networks on GPU
Tomko, Martin ; Zachariášová, Marcela (referee) ; Krčma, Martin (advisor)
This thesis deals with the implementation of an application for artificial neural networks simulation and acceleration using a graphics processing unit. The computation and training of feedforward neural networks using the Backpropagation algorithm are the main focus of this thesis, but the application also supports other network types, and it makes it possible to extend the application with different training algorithms. Next, the application allows us to create neural networks with structural anomalies, and thus, to test the neural network's fault tolerance. The application is implemented in the C++ language, using OpenCL to manage GPU computation. The Backpropagation acceleration results were compared with the free open source library FANN.

Distributed Processing of IP flow Data
Krobot, Pavel ; Kořenek, Jan (referee) ; Žádník, Martin (advisor)
This thesis deals with the subject of distributed processing of IP flow. Main goal is to provide an implementation of a software collector which allows storing and processing huge amount of a network data in particular. There was studied an open-source implementation of a framework for the distributed processing of large data sets called Hadoop, which is based on MapReduce paradigm. There were made some experiments with this system which provided the comparison with the current systems and shown weaknesses of this framework. Based on this knowledge there was created a specification and scheme for an extension of current software collector within this work. In terms of the created scheme there was created an implementation of query framework for formed collector, which is considered as most critical in the field of distributed processing of IP flow data. Results of experiments with created implementation show significant performance growth and ability of linear scalability with some types of queries.

SMV-2016-23: Molecular references for ultra-compact laser standards
Hrabina, Jan ; Holá, Miroslava ; Oulehla, Jindřich ; Pokorný, Pavel ; Lazar, Josef
The contractual research was oriented towards development of a set of optical frequency references intended for frequency stabilization of ultra-compact laser sources. These optical references were based on absorption cells filled with ultra-pure absorption gases and they allow precise locking of the lasers with the methods of laser spectroscopy.
