Analysis of candidate transmembrane adaptor proteins

Transmembrane adaptor proteins (TRAPs) function as a scaffolding anchor for numerous signaling molecules and facilitate formation of large signaling complexes near the plasma membrane. They regulate spatio-temporal organization of signaling events in order to transduce fast and effective signal from surface receptors into the cell nucleus. The importance of described TRAPs in the development and proper functioning of immune cells in relation to their localization within lipid rafts is still under debate. The simple reason is the lack of a profound phenotype in knock-out mice. The described phenotypes observed in animals deficient in ITAM-containing adaptors of immunoreceptors as well as lipid raft adaptor Lat all lead to the block in the T-cell development and impaired TCR signaling. The subject of my diploma thesis is to perform bioinformatic search and filter candidate TRAP genes according to predicted and published data. Selected TRAP candidates were subjected to functional characterization. I checked for their localization to the plasma membrane and correlate their ectopic overexpression with the expression of CD69 surface Tcell activation marker. From 14 candidate TRAPs, only a single Pdzk1ip1 protein is probably localized to the plasma membrane but its expression does not influence CD69...

Simulation of nesting of birds

Theory (Storch and Frynta, 1999) suggest that in a process of the evolution some species acquire ability to select better habitats upon presence of habitat features which correlate with expected tness. As far as we know this theory was not yet tested with computational agent-based model. In this thesis we introduce computational agent-based model describing life cycle of autonomous agents in their habitat for studying evolution of habitat selection according to the theory. Results of the simulation show ability of the agents to adopt on differenct habitats by acquision of proper cognitive clues and formation of separate groups of agents within species.

Types of word-formation in modern standard Chinese and the effect of word-formation on the typology

Tvoření slov rozumíme vznik nových slov na základě už existujících forem v daném jazyce. Tento způsob je nejdůležitějším zdrojem obohacování slovní zásoby. Proces vzniku nového slova probíhá na principu hledání odpovídajícího pojmenování pro určitou myšlenkovou bázi, přitom se vychází od konkrétní známé formy. Nová slova vznikají rozličnými postupy. Nejrozšířenější způsoby tvoření slov jsou afixace a skládání (kompozice). Okrajově se v odborné literatuře uvádí i zkracování (abreviace), nastavování slov, mechanické krácení atd. V čínštině se běžně uplatňují afixace, kompozice, abreviace a navíc i zdvojení významových jednotek (reduplikace). Při tvoření slov derivací a reduplikací se zpravidla vychází z jednoho základu a při kompozici ze dvou a více. Naším úkolem je seznámit čtenáře s těmito způsoby tvoření slov v moderní čínštině. Přitom nemáme za cíl podat vyčerpávající výklad o možných existujících slovotvorných typech, ale budeme se zabývat jen těmi produktivními a představíme ty nejběžnější neproduktivní. Při popisu jednotlivých způsobů slovotvorby si budeme všímat zejména funkcí odvozovacích morfémů (prefixů, infixů a sufixů), významových změn při tvoření slov a struktur a slovnědruhové příslušnosti vzniklých slov. Formation of carbonyl compounds by non-enzymatic protein glycation

Carbonyl groups are formed by effect of protein oxidation or glycation. As a model protein, I used bovine serum albumin for spectrophotometric and aspartate aminotransferase for electrophoretic method of assessment, which were glycated by several glycating agents (glucose; glucose + copper; fructose; fructose + copper; methylglyoxal). I optimized different parameters of spectrophotometric determination: solvent, concentration of reagent, amount of protein, and duration of incubation. I managed to find suitable conditions of this determination, which I have used in subsequent experiments with different glycating agents. At electrophoretic determination, I performed SDS-PAGE with following silver staining or immunoblotting with chemiluminescent detection. Formation, incorporation and invalidity of a business company

The main purpose of my thesis is to analyse and clarify the process of foundation, incorporation and invalidity of the business company. A business company may be defined as a private association of persons established under an agreement, whose purpose is to undertake business. Members contribute their own money to the business and divide among themselves the profits attained. Regulation of all types of business company is contained in Part II of the Commercial Code. Our Commercial Code provides six types of the business company: Unlimited liability company, limited partnership company, limited liability company, joint-stock company, European public limited company and finally European economic interest grouping. It is necessary to be careful with the translation of the concrete types of the company, because this is only approximate translation of the terms, because of the distinct legal institutions in each legal system. The thesis is composed of seven chapters, each of them dealing with different aspects of the above mentioned process of foundation, incorporation and invalidity of the business company. The case of "State Transformation in Postcolonial Mozambique" is set into the context of African postcolonial state research in the introductory part of the thesis. The concepts of africanists Mahmood Mamdani, Patrick Chabal and Jean François Bayart are presented and used as an conceptual framework for the analysis of the postcolonial state in Mozambique. The origin and development of the postcolonial state in Mozambique is analysed in the wider historical context. The pre-colonial political systems in Mozambique are presented, as well as the first contacts between the pre-colonial Mozambique and the Portuguese and the establishment of the Portuguese presence. The description of the formation and consolidation of the colonial state includes the issue of colonial border demarcation, conquest of the territory and establishment of the colonial administration. The Mozambican independence struggle is analysed as well as its impact on the colonial administration. The development of the leading force of the struggle, the FRELIMO party, is analysed including its origin in the union of three nationalist movements, its internal disputes and divisions and its gradual radicalisation. GCC: economic relations and perspectives of next integration

The target of the thesis is to describe the integrative process of six countries from the Gulf as a chain of reactions to former challenges, wishes and needs of these countries, with the insistence on analysis of actual economic situation and mutual relations. The first chapter pursues the economic and safety situation going along with the rise of GCC and introduction of cooperation areas and its results. The second chapter describes the economic situation after the formation of GCC and impacts of integrative process, but the main point is the detailed analysis of actual economic situation in the region. The last chapter deals with the topic of next expansion of GCC or next continuing of the integration and presents the possible future development of GCC.

The issue of constructions on the land of another

Diploma thesis deals with the issue of constructions on the land of another. At first the thesis focuses on definition of basic terms, i.e. the construction and the building plot. In the following text is concerned about permission to build. Attention is focused on three types of titles of use - lease, easement and of peripheral importance, loan of land, which is not given a full explanation, but only focusing on issues related to the topic of work. To the previous issues link two institutes which deal with third and fourth chapter - authorized and unauthorized construction. Authorized construction is current problem in the future and therefore it is very noticeable lack of explicit rules that must be compensated for general rules of unjust enrichment. The chapter on unauthorized construction is divided into sections according to the character of each "type", i.e. whether the construction is movable thing or realty and whether the authorization to construction lacked from outset or dropped out after the construction was built.
