The Repair of Oxidative DNA Damage in the Chinese Hamster Cell Line Deficient in Nucleotide Excision Repair

The repair of oxidative DNA damage in the Chinese hamster cell line deficient in nucleotide excision repair Abstract The aim of our work was to specify DNA oxidative damage repair in the Chinese hamster ovary cell line. Comet assay was used to detect the damage consequences - DNA breaks. We were capable of achieving high method sensitivity due to application of specific endonucleases and specific inhibitors of excision repair - inhibitor of poly(ADP- ribose)polymerase PARP 3-aminobenzamide (3-AB) and cytosine arabinoside with hydroxyurea (AraC/HU). PARP-1 is an important factor in the base excision repair (BER) process. PARP-1 deficient cells show recovery impairment both in LP-BER and SP-BER way. AraC/HU is known as the long-patch excision repair inhibitor. The results of our experiments showed the inhibiting effect of 3-AB on the repair of hydrogen peroxide-induced DNA damage. But in the same setting the AraC/HU effect on repair was not observed. To make the value of experimental results more significant and clear, we decided for additional experiments with monofunctional alkylating agent methyl methanesulfonate (MMS). Damage caused by MMS showed slower repair both when 3-AB or AraC/HU were present during the repair period. Based on all available results we suppose that the MMS and also hydrogen peroxide...

The integration of MS Excel and BusinessObjects

The main goal of this work is to analyze the change in reporting mortgage processes in the GE Money Bank from the solution based on the application Microsoft Excel into a state, where the mortgage reporting is integrated into the solution based on BusinessObjects platform. And at the same time to carry out the comparison of new and previous solution. To partial purposes belongs the quantification of time savings and benefits resulting from the integration and adumbration of the next integration step "Generation 2". These aims were achieved by obtaining theoretic knowledge of Business Intelligence, by my participation in the real project practically and by consultations of knowledge with specialists in practice. Main contribution of this work is in suggestion and analysis of the reporting solution in the firm. It enables speed up and automatizes processes of data measure, which were ineffective and time-consuming capacities of workers. The work is divided into the four main chapters. The teoretic forepart introduces the role of Business Intelligence in a firm and acquaints readers with contemporary trends in BI. The second part has practical background. The Business Intelligence architecture in GE Money Bank is analyzed in this part. Awareness of BI architecture and data sources is important due to understanding connection of the BusinessObjects with the database. In the third part the project "Integration of mortgage measure from MS Excel to the BusinessObjects" is introduced and it is also suggested, how the integration would be proceed. At last part my personal findings are summarized, benefits of integration are formulated and future view is described. Last but not least an advice resulting from the project is showed or what would be possible to do better next time.

Versioning compression of text documents.

In the work we study e ective management of di erent versions of text les. Outcome of the work is an application managing such archives. The application is able to maximally exploit a similarity between versions of les to avoid redundancy of stored information. Application uses the method of Diff erential File Comparision (diff ), improved by additional smaller dividing entities. Contrary to the the classical di it recogizes also word and character diff erences. Precision of the di vary along a text le according to density and character of differences. Due to that a smaller archive is made and moreover in shorter time. Extra reduction of the archive is achieved by the use of the zip compression method implemented by freeware library Info-ZIP.

Tax Optimalization of The Business Subject

In my bachelors work I focus on a particular company{\crq}s tax optimization. The aim of the work is to find an optimal tax solution, especially the Juristic Persons´ Income Tax and to accomplish that the company has the lowest tax duty possible. The particular proposals for tax duty reduction are explained gradually and calculated in the practical part. In this work I came out of precise data and numbers that I had obtained from the chosen company. This company, Epigon A Ledenice is one of the best agriculture companies in the České Budějovice district. I have chosen this company due to my good contacts with the owner of the company and therefore the possibility to get to know well its internal functioning, economical situation and property structure. The work is divided into twelve chapters. The first chapter is an introduction where the essence of this work is summarised, the Czech republic tax system and Juristic Persons´Income Tax general data are stated in the following chapters. In these chapters, I came out especially of laws, regulations and specialised publications. The chapters are divided gradually into particular subchapters dealing with specific problematic. These introducing chapters that are to acquaint the readers with the problem in theoretical round are followed by a chapter where possible cases of tax optimization are solved practically along with the calculation of advantageousness. The results I have achieved in this work and that are offered to the company as its tax duty solutions variants are summarized in the final chapter.

Internal Development of Macedonia (1991-2005)

The Republic of Macedonia gained its independence peacefully in November 1991 and was the only one ofthe former Yugoslav republic that got its independence peacefully. The peacefully transformation of Macedonia was preceded by an uneasy period of democratic consolidation. Among the different factors that negatively influenced succesful consolidation were - the struggle for the international recognition of the country, the Greek diplomatic and economic pressure for the republic to change its name, the disruption of the economy due to the UN sanctions on Macedonia's main trade partner Serbia, and the Kosovo refugee crisis. None of these factors hadn't a such strong importance for the consolidation of Macedonia as the disputing of character of the state by Macedonian Albanians. Ethnic Albanians were according to Constitution relegated to the category of a minority, just like Turks, Vlachs, Roma and others. Ethnic Albanians disagreed with these status of minority. Transatlantic trade and investment are intense and the two economics narrowly interlinked. Despite the rapid increase of the global trade in recent decades, transatlantic economic relations remain the dominant force in international economic relations for both the United States and the European Union, particulary if one considers not only trade but also foreign direct investment. Today American and European economies are the world's largest, and they are likely to remain so for foreseeable future. Overall bilateral commerce approaches 2.5 trillion of dollars per year and employs directly or indirectly some twelve million workers in Europe and the US (2000). With such intensive commercial relations trade disputes have arisen with some frequency whereas the range of trade conflicts is so wide that a complete accounting is difficult. During the cold war foreign economic objectives were often subordinated to security concerns. The US fathered European integration primarily to achieve political rather than economic objectives. Dynamic Loewenstein occupational therapy cognitive assessment in individuals after obtaining brain injury in working age

The thesis Dynamic Loewenstein occupational therapy cognitive assessment in individuals after obtaining brain injury in working age deals with a comparison of the level of the brain injured and non-brain injured people's cognitive functions with the aid of the Loewenstein Occupational Therapy Cognitive Assessment battery (DLOTCA). The theoretical part is focused on an issue of the acquired brain injury, a description of cognitive areas and as well their pathology due to this injury. The aim of the practical part is to test the cognitive functions with the aid of the Loewenstein Occupational Therapy Cognitive Assessment. The survey's main object is the applicability of the working version of the Loewenstein Occupational Therapy Cognitive Assessment battery (DLOTCA), its translated version. The testing was carried out on seventy probands devided into three groups. Healthy people (n=50), cerebrovascular accident patients (n=10) and traumatic brain injury patients (n=10). Test results confirm all the stated hypothesis because it did not meet the prerequisites of the total evaluation. Prohibition vs. decriminalization - two different approaches to the drug abuse problem

The main subject of this study is a comparation of two different policies which are used to cope with the drug abuse problem - prohibition and decriminalization. Example is set on the U.S. and the Netherlands. First, each policy is described in detail. Subsequently, both policies are compared according to criteria of the drug use prevalence and the direct costs related with them. The Netherlands, which applies decriminalization, has maintained lower drug use prevalence than the U.S for long time and in almost all kinds of drugs and age groups. Moreover, Netherlands it achieves at the same or lower relative costs. Yet, the higher drug use prevalence does not prove ineffectiveness of policy prohibition. It seems that the U.S. is more likely to have higher prevalence due to more general tendency of using drugs in population. The study also shows that the decriminalization can be linked to increase of drug use that followed its introduction in Netherlands.

Utilization of endophytic microorganisms for growth promotion of Miscanthus

Increased survival and aboveground biomass yield of Miscanthus was achieved due to the inoculation of Miscanthus seedlings by a mixed inocula based on the selected endophytic microbes (bacteria and fungi). Inoculated treatments produced 20-90% higher yield of biomass compared to the controls. Importantly, fungal inoculum increased also production of rhizomes and the same trend was observed in the treatment inoculated by the bacterial mixture. Bacterial endophytes also strongly augmented overwintering of the plants on one of the tested model plantations. Because the price of the inocula production is economically feasible and does not substantially increase the plantation establishment costs we hope for fast implementation of the methodology both in CR and beyond.

New Approaches in Airborne Thermal Image Processing for Landscape Assessment

Letecká termální hyperspektrální data přinášejí řadu informací o teplotě a emisivitě zemského povrchu. Při odhadování těchto parametrů z dálkového snímání tepelného záření je třeba řešit nedourčený systém rovnic. Bylo navrhnuto několik přístupů jak tento problém vyřešit, přičemž nejrozšířenější je algoritmus označovaný jako Temperature and Emissivity Separation (TES). Tato práce má dva hlavní cíle: 1) zlepšení algoritmu TES a 2) jeho implementaci do procesingového řetězce pro zpracování obrazových dat získaných senzorem TASI. Zlepšení algoritmu TES je možné dosáhnout nahrazením používaného modulu normalizování emisivity (tzv. Normalized Emissivity Module) částí, která je založena na vyhlazení spektrálních charakteristik nasnímané radiance. Nový modul je pak označen jako Optimized Smoothing for Temperature Emissivity Separation (OSTES). Algoritmus OSTES je připojen k procesingovému řetězci pro zpracování obrazových dat ze senzoru TASI. Testování na simulovaných datech ukázalo, že použití algoritmu OSTES vede k přesnějším odhadům teploty a emisivity. OSTES byl dále testován na datech získaných ze senzorů ASTER a TASI. V těchto případech však není možné pozorovat výrazné zlepšení z důvodu nedokonalých atmosférických korekcí. Nicméně hodnoty emisivity získané algoritmem OSTES vykazují více homogenní vlastnosti než hodnoty ze standardního produktu senzoru ASTER.
