Radka Vargová Comparison of results of two methods for laboratory diagnosing lyme borreliosis - PCR methods and ELISA Diploma thesis Charles University in Prague, Facutly of Pharmacy in Hradec Králové Specialist in laboratory methods At present time the Lyme disease is relatively wide-spread affection. Its agent is spirochete Borrelia burgdorferi sensu lato and its most eminent vector in Europe is the tick Ixodes ricinus. In the theoretical part of my thesis I decribed the basic characteristics of the genus Borrelia, its antigenic structure, epidemology and pathogenesis, disease vectors, clinical sypmtoms of the infection and possibilities of laboratory diagnostics. My thesis is aimed at comparison of dignostics of Lyme disease by using serological method ELISA and using method PCR. It was used 113 anonymized samples of patients for analyse. The samples were examined for the presence of the antibody against Borrelia in classes IgG and IgM and by using real-time PCR method they were examined for the presence of DNA pathogenic Borrelia. The experimental part of the thesis describes realization of the methods, which we have used for diagnostics of Lyme disease. Determination of the lipid profile in biological material by the method HPLC-ELSD

A method of high-performance liquid chromatography with evaporative light-scattering detector (HPLC-ELSD) has been optimalized for the determination of neutral and polar lipids. Column filled by silica with chemically bonded diol has been used as stationary phase. As mobile phase, a ternary gradient composed from A: hexan-tetrahydrofuran 99:1 (v/v), B: isopropanol-chloroform-acetic acid 82:20:0,01 (v/v/v), C: isopropanol-water-triethylamine 47:47:6 (v/v/v) was used. Calibration curves have been measured in the range 2-200 μg of the injected amount; for individual lipid classes, optimal interlay of experimental data corresponded to the following functions: triacylglycerols - third order polynom (R=0,998), cholesterol esters - exponential dependence (R=0,998), free cholesterol - third order polynom (R=0.9998), ceramid - exponential dependence (R=0,992), cardiolipin - square dependence (R=0,998), phosphatidylethanolamine - exponential dependence (R=0,999), phosphatidylcholine - square dependence (R=0,997), phosphatidylserine - third order polynom (R=0,9985), sphingomyelin - third order polynom (R=0,9997), lysophosphatidylcholine - exponential dependence (R=0,9986). Properties of branched oligoesters plasticized using 20 % of triethyl citrate

The aim of this graduation theses objective was the study of adhesions and rheologic properties of branched oligoesters and acyclovir release from these carriers. The experimental part introduces main targets for the mucoadhesion, natural and synthetic bioadhesives, their functions and properties. And further more there is the mucoadhesion in gastrointestinal tract and oral cavity with some examples of polymers and dosage forms. The practical part is regarded to the testing of dynamic viscosity and adhesion of the oligoesters of lactic and glycolic acid branched using dipentaerythrotol (0,5D, 1D, 2D, 3D, 5D, 8D) and linear PLGA. These oligoesters were plasticized by 20% of triethylcitrate. The acyclovir release from carriers 0,5D, 1D and 2D was studed. The dynamic viscosity was the highest by carriers 3D and 0,5D. The highest adhesion power was measured out by carriers 0,5D and PLGA. The longest period time of release was by carrier 2D, twenty-one days. Diploma Paper, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany and Ecology, 2007, p. 61 The work aimed at biotransformation of exogenous precursors of arbutin with in vitro cultures of Centella asiatica (L.). The precursors of arbutin were added into the medium (hydroquinone, tyrosine, p-coumaric acid, hydroxybenzoic acid). The precursors were used in a concentration of 200 mg/l and the period of their action was 6, 12, 24, 48 and 168 hours (1 week). The cultivation of calluses was created by two different ways - by suspension culture and by culture cultivated on the filter-paper bridge. The cultivation was prosecuted on the light. Positive results (both TLC and HPLC analysis) in arbutin production were obtained in all suspension cultures after an addition of hydroquinone. The largest amount of arbutin in callus cultures was measured after a 48 hours cultivation (0,043 %). After a 168 hours cultivation was amount of arbutin only 0,038 %. In this experimental variant arbutin was also released into the medium (0,096 %). Biotransformation experiments with the others precursors of arbutin were not successful. The way of cultivation by the filter-paper bridges was also unsuccessful.

Diplomová práce 2009 Abstract_ENG Jitka Vlčková ABSTRACT An investigation presented was concern to measure and evaluate the total antioxidant capacity (TAC) in samples of red wine clarified with four fining agents (polyvinylpolypyrrolidone and three different types of gelatin). All wine samples were treated by concentrations of 10, 20 and 40 g/hL of finings. The main aim was to discover how the particular agents in different dosages influence the TAC. The second part describes relations between changes in TAC in content of single antioxidant components (total polyphenols, anthocyanins, gallic acid and simple phenols). Total antioxidant capacity in red wine samples was evaluated using the Trolox equivalent antioxidant capacity assay (TEAC). A theoretical part of the thesis briefly summarizes basics about luminescence, focused on chemiluminescent reaction. Theory of free radicals and antioxidants, especially those included in red wine are also described. Process of winemaking aimed at fining is also contained in this part. An experimental part describes techniques of determination of the total antioxidant capacity, principle of chemiluminescent assay based on the luminol/peroxidase system. Influence of MDOCTM Administration on Atherogenic Process in the Experimental Model of Atherosclerosis. Mgr. Pavla Tomalová In this study we investigated prospective hypolipidemic and anti - inflammatory effects of modified oxidized cellulose (MDOC). In the therapy of hyperlipidemia and blood - vessel complications (atherosclerosis) there are used the most considerable substances - statins, today. However, it was also documented, that feed dietary roughage has positive influence on cardiovascular system, inclusive of cholesterol level. MDOC (polyanhydroglucuronic acid), because of it's properties can be classified probably as the soluble fibre. We used apoE deficient mice as the model of atherosclerosis, fed by atherogenic diet. We analyzed levels of total cholesterol and other lipoprotein fractions and serum concentrations of inflammatory markers (IL-6 and VCAM-1) in blood. Total cholesterol concentrations were assessed enzymatically by conventional diagnostic methods and spectrophotometric analyses. For the detection of VCAM-1 expression in aortic sinus and part of aortic arch we used imunohistochemical methods and for quantification of VCAM-1 expression we used stereological methods. Separation and determination of pharmaceutically important compounds using electrophoretic methods

This thesis deals with the use of electrophoretic methods in the analysis of pharmaceutically important compounds. In the theoretical part, the classification of capillary electrophoretic techniques is discussed. Capillary zone electrophoresis (CZE, capillary isotachophoresis (ITP), micellar elektrokinetic chromatography (MEKC), microemulsion elektrokinetic chromatography (MEEKC), isoelectric focusation (IEF), capillary electrochromatography (CEC) and capillary gel electrophoresis (CGE) are described. Further, on-line pre-concentration methods are described. Particular attention is given to the on-line combination of capillary isotachophoresis with capillary zone electrophoresis using column coupling arrangement. This combination enables the improvement of the selectivity of separation and the improvement of the limit of detection of analytes present in complex matrices of biological and environmental origin. Afterwards, the results of the experimental analyses are presented: 1. The first paper deals with the use of CZE in the analysis of ibuprofen and flurbiprofen in pharmaceuticals. Separation was carried out in a fused silica capillary (60 cm x 100 m I.D effective length 45 cm) at 30 kV with UV detection at 232 nm. Experimental Testing of Game-Theoretic Predictions: The Ultimatum Game

This thesis focuses on testing of game theoretical predictions in the ultimatum game by means of controlled experiments. This game has become one of the most scrutinized games from the area of bargaining game theory. The theoretical division of the reward, which the players bargain over, is such that one player gets virtually all the reward while the second player is left with nothing. Because of such an extreme division of the reward, the game represents a severe test for the theory. In fact, experimental results do not confirm to the theory. This thesis provides a survey of the experimental studies investigating different aspects that may affect the subjects' behavior in the game. Furthermore, some possible explanations for why the theoretical solution is not observed to be played by the subjects in the laboratory are presented. I show several new models, which try to capture the real nature of the subjects' behavior in the game. I also focus on the proposers' behavior from the income-maximizing point of view if the distribution of the responder's minimum acceptance thresholds is known to them. Outline of a new experiment examining such behavior is then presented.

Some Aspects of Communication with a Blind Patient

This diploma work deals with some aspects of the communication with sight disabled patients. The theoretical part is dedicated to the problems of a sight disability, general definition of a communicational idea and and communicational specialities of sight disabled patients. It describes some particular sight defects, psycho - social aspects of a sight disability and it also deals with a training of generally educated nurses, nurses with a diploma and health assistants for a work with sight disabled people. In an experimental part the diploma work makes an inquiry of sight disabled people when visiting Health Centers and on the contrary the experience of nurses with these patients. For this research work two questionaires were made and used. One of the questionaires was used for sight disabled people, another one for nurses. Two experimantal groups consisted of 104 sight disabled people and 100 nurses working in outpatients department. From this research it is perceptable that all the nurses working at all the health workplaces should know how to work with sight disabled people.The research proved that the problems appear in dealing nurses with patients, these problems concern especially the nurses lack of effort to solve the situation with a companion or an unsuitable accompaniment. Experimental Investigation Of Transonic Blade Cascade With Damping Device Tie-Boss

This paper is concerned with investigation of influence of two variants of Tie-Boss on\naerodynamic characteristics of a blade cascade. Conclusions are drawn based on results of\nexperiments and numerical simulations. The results show that one type of Tie-Boss exhibits\nlower losses of kinetic energy but higher flow turning than the other.
