Study of the novel phtalocyanines for the vascular-targeted photodynamic therapy of tumors
Brázdová, Petra ; Macháček, Miloslav (advisor) ; Šimůnek, Tomáš (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Petra Brázdová Supervisor: Mgr. Miloslav Macháček Title of diploma thesis: Study of the novel phthalocyanines for the vascular-targeted photodynamic therapy of tumors Cancer is one of the leading causes of death in developed countries, therefore a big effort is devoted on research of novel anticancer drugs. Photodynamic therapy (PDT) is currently a well-established method in this area, but targeting its effect on tumor vasculature has not been considered until recently. In comparison with PDT, vascular-targeted photodynamic therapy (VTP) takes the advantage of a much shorter interval between administration of the photosensitizer (PS) and irradiation (drug-light interval) and thus turning the effect of cytotoxic agents to tumor vasculature. It is intended to disrupt the oxygen and nutrients supply to malignant cells and thus cause their damage and death. Aim of this work is to evaluate the efficacy of newly synthesized PSs from the group of phthalocyanines and azaphthalocyanines using the VTP protocol under in vitro conditions. The PSs used in this study have been investigated recently with classical PDT protocol, in which they have demonstrated very promising activity against tumor cells...

Molecular simulations in structure analysis of intercalates
Veteška, Marek ; Pospíšil, Miroslav (advisor) ; Beneš, Ludvík (referee)
The samples of hydrotalcite intercalated with pyrenetetrasulphonate acid were obtained. Their structure cannot be precisely determined by the analysis of X-ray data. The amount of water in the interlayer space and the structure of the samples differed considerably in dependence on humidity. Convenient procedures and minimization strategy were established on the base of X-ray data in the molecular mechanic and the molecular dynamic simulations. A procedure for using of the supramol software was made. This software enables deterministic searching of conformational space. Initial models from supramol were minimized in the Cerius2 software using the Universal force field and then they were eventually optimized by the molecular dynamics. The arrangement of molecules of pyrenetetrasulphonate acid and water was determined in the interlayer space of hydrotalcite for three different samples with different interlayer distances: The sample 1 prepared by RH = 0%: 9.83A; the sample 2 prepared by RH = 84%: 13.63A; the sample 3 prepared RH = 40%-50%: 11.74A a 12.81A. The results of these structures proved a large variability of the arrangement of the molecules in the interlayer space. The sample No. 3 seems to be unstable.

Harmonizace Účetní Úpravy Pozemků, Budov, Zařízení a Nehmotných Aktiv
Babinec, Matúš ; Krupová, Lenka (advisor) ; Mejzlík, Ladislav (referee)
The globalization of world economy has inevitably affected also the movements in harmonization process of accounting standards around the world. International Financial Reporting Standards have become the ultimate accounting and reporting tool of most developed countries. The only remaining counterpart -- the US GAAP, is slowly subsiding to the enormous spread momentum that IFRS gained in recent years. In this setting I examine the application of International standards on accounting treatment of Property, Plant and Equipment, and also the Intangible Assets. Subsequently I compare this treatment to US GAAP and document all major differences still persistent at the end of year 2008. Considering that there are still many divergences between the two sets of standards, I try to document their impact on the reporting and presentation of financial statements of several blue chip companies. My analysis suggests that differences in reported numbers depending on standards chosen can distort comparability of financial statements and performance assessment, thus potentially biasing the decision making process of involved stakeholders.

Dream of Great France in Transatlantic Perspective
Paggio, Viktor ; Dubský, Zbyněk (advisor) ; Nechvátal, Martin (referee)
This text focuses on the French self-peception in the modern world. French consider their concept of state and citizenship universal, as Americans do. But the two universalisms are built on a different basis in terms of religion, language and many other aspects. I analyze the clash of the two visions of the world and the French reactions to the American rise to power in the 20th century.

UNCITRAL Arbitration Rules and their application in practice
Halada, Martin ; Růžička, Květoslav (advisor) ; Pfeiffer, Magdalena (referee)
UNCITRAL Arbitration Rules and their application in practice The purpose of the thesis is to analyse the UNCITRAL Arbitration Rules and the ways they are applied and interpreted by parties of dispute and arbitrators in international commercial arbitration. The thesis is divided into two parts. In the fist part the basic principles and legal framework of international commercial arbitration are described. Differences between institutional arbitration and ad hoc arbitration are discussed and special chapters are dedicated to permanent arbitration courts in the Czech Republic and important international arbitration institutions. Second part begins by discussion of the UNCITRAL Commission history and describes historical development of the UNCITRAL Arbitration Rules. Analysis of individual articles follows. With some articles there is established common international practice regarding their application or interpretation in international commercial arbitration. In such cases these practices and interpretations are discussed in more detail. Some articles of the revised 2010 version of the UNCITRAL Arbitration Rules are significantly different compared to the 1976 version of the UNCITRAL Arbitration Rules. These differences are analysed comparatively. Legal-analytical, legal-descriptive and legal-...

Effect of temeperature stress and water shortage on thousand grain weight of selected winter wheat varietes
Hlaváčová, Marcela ; Pohanková, Eva ; Klem, Karel ; Trnka, Miroslav
The aim of the presented study was to assess the effect of high temperatures and water shortage during anthesis on thousand grain weight (TGW) of two winter wheat varieties (Tobak and Pannonia). In addition, numbers of grains per spike were also assessed. The six growth chambers were used to simulate heat stress conditions within following gradient of temperature maxima: 26 degrees C (control chamber), 29, 32, 35, 38 and 41 degrees C. The relative humidity (RH) course and photosynthetically active radiation (PAR) intensity were controlled via protocols. Additionally, drought stressed (dry) and well-watered (wet) treatments were established within each growth chamber. The plants were removed from the growth chambers after 14 days and they were left until a full maturity, exposed to actual weather conditions. The TGW at 14% moisture were evaluated for particular treatments within both winter wheat varieties. TGW was generally more affected by high temperatures under drought stress than in well-watered conditions. The results revealed that Pannonia TGW was much more affected by the water deficiency in combination with high temperature (particularly 38 and 41 degrees C) than Tobak TGW.

Transgenerational effect in \kur{Taraxacum brevicorniculatum}: test of a novel method of experimental plant DNA demethylation and its practical application in exploring the impact of maternal competition on progeny phenotype
DVOŘÁKOVÁ, Hana
Spray application of 5azacytidine on established plant seedlings was tested for its demethylating efficiency, as it represents a novel method for plant experimental demethylation with a potentially lower negative impact on plant development compared to the traditional application of the demethytaling agent through germination of seeds in its solution. Further, the 5azacytidine spray application was used in practice to erase the epigenetic memory in offspring of Taraxacum brevicorniculatum plants from different competitive conditions. The impact of parental competition on the juvenile phenotype was estimated by measuring growth related traits, while the experimental demethylation allowed for evaluating the significance of DNA methylation marks in bioticaly induced transgenerational effects in T. brevirorniculatum.
