Study of stress factors in Candida albicans.
Vindušková, Renata ; Buchta, Vladimír (advisor) ; Podholová, Beata (referee)
Yeast cells, when growing on solid surfaces, form multicellular structures, colonies, with a characteristic organised morphologies, formed during the division of non- motile yeast cells. Organisation of yeast colonies should, therefore be ensured by signals transmitted and received by dividing cells within a colony In this regard, the yeast colony exhibits an analogy to embryogenesis of higher organisms, One of the characteristic attributes of multicellular organisms is their ability to emit and receive signals over long distances. Yeast colonies use for the long-range inter-colony signalling a simple volatile compound ammonia, produced by colonies in pulses. Ammonia action results in synchronisation of the development in neighbouring colonies. The transition of colonies from acidic phase to the phase of intense ammonia production (alkali phase) is connected with intensive metabolical changes. In this regard volatile ammonia acts as starvation signal between colonies. If we are able to influence cell communication for exampel by new antibiotics, we would protect ourselves against several mycoses.

Oppression symptoms in chldren's drawings - necessary knowledge for arts and crafts teachers
Skálová, Hana ; Klusák, Miroslav (referee) ; Jakubcová Hajdušková, Lucie (referee)
The work proceeds from the idea, that the knowledge of signs of emotional distress which can be found in children's drawings is important of arts and crafts teachers. There is more and more children living under stress and this is one of several possible ways of how to find them and provide them timely help. Children living under the stress (exposed to cruelty, too much demands, lack of emotional care etc.) show certain signs in their drawings and children psychologists can work with it. If this issue is taught in pedagogical faculties and in special lessons for arts and craft teachers, educators would be able to contact psychologists in time. The works is analysing available Czech psychological literature and determines whether someone has already worked with the issue, also introduces publications which might be useful for course of study of teachers, brings proposals of the idea of how to proceed in arts and crafts lessons, summarizes the problematic of CAN (syndrome of maltreated, neglected and abused child) and brings alarming tel. numbers of the Safety line. In the practical part, the teachers from kindergarten and grammar school prove that they are able to react to warning signs in children's drawings.

Study of the factors that influence setting time during current measurement.
Ježík, Jan ; Macháň, Ladislav (referee) ; Běťák, Petr (advisor)
This study is engaged in the effect of dielectric relaxation on very small direct current setting time after applying voltage of 600 V. Several relaxation phenomenons in dielectric materials stressed by electric field are discussed. Characteristics of three substrates of PCBs are evaluated by using time and frequency domain dielectric relaxation spectroscopy in order to find an optimal material for high speed tester UNISPOT S40 ACCEL construction.

Employment of gas chromatography in the field of drug analysis II.
Špalová, Klára ; Kučera, Radim (advisor) ; Kovaříková, Petra (referee)
Gas chromatography in drug analysis II. Thesis Klára Špalová Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Drug Control In this study, effect of various conditions on stability of 1,3-butanediol was studied. During these studies, 1,3-butanediol was found as a rather stable compound, susceptible only to oxidation using oxidative agents- including air oxygen or UV radiation. Various degradation products were detected during stress studies. There were peaks at retention times 6.0 and 6.9 at every chromatogram, which grew in time under some conditions. A new degradation product at retention time 3.0 was detected when UV radiation actuated on aqueous solution of 1,3- butanediol. The same peak was observed under oxidative conditions., Its concentration grew significantly in time.

Hybrid Composites Combining Short Deformable Fibers and Particulate Fillers in Polymeric Matrix
Smrčková, Markéta ; Pavelka,, Vladimír (referee) ; Poláček, Petr (advisor)
In this work, the effect of adding short deformable polyvinylalcohol fibers into dimethacrylate resins on mechanical properties of composite materials was investigated. Furthermore, micro- or nano-fillers were added into the matrix. The effect of filler/short fibers combination on mechanical properties of so-called hybrid composites was studied. Composites were characterized by differential compensation photocalorimetry (DPC), termogravimetric (TGA) and dynamic mechanical (DMA) analysis. The fracture surfaces were examined using scanning electron (SEM) and confocal laser (CLSM) microscopy. Elastic modulus and strength, the critical value of stress intensity factor and strain energy release rate and the viscoelastic properties of composite materials were measured. The mechanical properties of composite materials are affected not only by the type of filler/reinforcement, but also by properties of the matrix. These properties also depend on the way of network formation during photo-initiated polymerization. For this reason, heat of polymerization, degree of conversion and the maximum polymerization rate of mixture of dimethacrylate monomers were determined.

Assessment of the Impact of Irregular Work Regime in Healthcare Workers in České Budějovice Hospital, Inc., on Cholesterol Level and Weight
PAVLOVÁ, Martina
Nursing is a very demanding profession requiring not only expertise and erudition, but also a good mental and physical condition. Nurses are not only professionals with their rights and duties, but also persons with their needs (6). Health professionals must be able to communicate not only among themselves, but mainly with clients, family members, medical and nursing officials and departments and clinic management. After working some years in health care facilities, professional burnout and fatigue can appear as a result of experienced stress (24). The objective of this thesis was to determine total cholesterol levels and BMI values in the medical staff working shift and night work in the wards, and in the staff working in a single-shift in out-patient departments in České Budějovice hospital, Inc. Another objective was to ??compare these values in relation with the regime of workers, their food and biorhythms. To meet these objectives, quantitative research was chosen which was conducted through questionnaires. Qualitative research was also carried out by determination of total cholesterol levels in the blood, by measuring height and weight of the medical staff members who filled out questionnaires, and calculating their BMI. The research has concluded that the hypothesis 1: Irregular work regime has an effect on cholesterol levels and weight, was not confirmed. Hypothesis 2: Irregular eating has an effect on cholesterol levels and weight, was confirmed, and Hypothesis 3: Biorhythm of an individual, disturbed by irregular work regime, has an effect on cholesterol levels and BMI, was neither confirm or refuted. I would like to provide the results of my research to the České Budějovice hospital management, to individual wards and departments officials as well as nursing staff members.

Soil macrofauna (Diplopoda, Chilopoda, Oniscidea) under restored flood conditions in meadow and forest habitats in South Moravia, Czech Republic
Tajovský, Karel
Floodplain habitats in the alluvium of the Morava and Dyje rivers, Czech Republic, were strongly affected by changes in water regime conditioned by regulation measures in the past centuries. These negative changes resulted in the cessation of regular floods, decline of groundwater level and subsequently in dramatic changes of characteristic vegetation as well as animal assemblages. Therefore activities aimed at more thorough analysis and restoration and revitalization projects were undertaken in nineties. Specific extreme conditions in the regularly flooded biotopes are determining for the existence of the specific soil invertebrate assemblages including millipedes (Diplopoda), centipedes (Chilopoda) and terrestrial isopods (Oniscidea). Long termed pitfall trapping processed in forest and meadow habitats in the years 1993-2001 enable us to evaluate the impact of restoration of regular floods and improving water regime on these terrestrial invertebrates. While the animal assemblages in floodplain forest (Ranšpurk National Nature Reserve) showed to be more conservative towards restoration processes, those in flooded meadow (Křivé jezero National Nature Reserve) dramatically stressed by previous negative conditions well responded to improving water regime. Renewed inundations at this locality contributed to the return of parameters of natural wetland habitat. Low densities and the species typical for extreme flood conditions (centipede Lamyctes emarginatus, millipedes Julus scanicus and Leptoiulus cibdellus, terrestrial isopod Trachelipus rathkii) were monitored at this locality.

Psychological aspects of the gothic in Joseph Sheridan Le Fanu's fiction
Procházková, Ilona ; Pilný, Ondřej (referee) ; Wallace, Clare (advisor)
What is usually understood by the term "Gothic" is the distant and rather obscure period of Middle Ages connoting severe wars, fortified inhospitable castles and the burning of witches. Apart from that, the word is very often used specifically to describe the architecture of this time. However, especially in an English cultural context this word gained a secondary meaning which is not completely unrelated to the first one and which is to a large extent connected with literature. Under the influence of Romanticism, the second half of the eighteenth century bears witness to extended interest in uncanny Gothic castles or ruins, forlorn scenery and other melancholy places, with even greater stress put on its mysterious, obscure and frightening aspects that finally resulted in something which may be called the Gothic revival. Victor Sage writes that, "'Gothic' could connote any of a wide range of overlapping senses: horrid, barbarous, superstitious, Tudor, Druid, English, German, and even Oriental."1 One of the primary goals of Gothic literature was to create strong emotion of fear or even horror. Among the most popular settings of its fiction belong gloomy ancient chambers, buildings with rich pasts, abandoned decaying mansions, graveyards and similar places which help to establish the right atmosphere for...
