Stimulant drugs and their physiological effects
Nováková, Pavlína ; Novotný, Jiří (advisor) ; Hejnová, Lucie (referee)
The category of stimulant drugs is a heterogenous group of substances which benefit-to-harm ratio varies widely. This work attempts to use a systemic approach in order to characterize the underlying basis of the physiological effects of their selected representatives in brain. A brief pharmacological description of caffeine, nicotine, cocaine, amphetamine and methamphetamine is followed by an identification of main molecular targets of these drugs in the context of respective neurotransmitter systems. Major attention is given to both acute and chronic molecular mechnisms of action of the selected stimulant drugs up to the level of selected relevant neural circuits. In the next section stimulant drugs are presented as Trojan horses that attack the motivational system of the brain. Initially, a theoretical analysis of reward together with a neuroanatomical dissection of reward-related circuitry serves as a basis for the formulation of contemporary theories of drug addiction. Finally, an effort is made to synthesize common molecular events with specific aspects of reward in order to draw a picture of the neurobiological view of addiction.

Petr Chelčický is one of the reformers from the first half of the 15th century. Little is known about him as a person. Of his 50 extant works, the most famous of which are O boji duchovním (About a Spiritual Fight), Replika proti Rokycanovi (A Repartee to Rokycana), Postila (Sermones) and Síť víry (A Net of Faith). The main topics of his treatises are a critique of the Catholic Church, an appeal against violence as a mode of action, and embracing non-violence. He often deals with dogmatic issues, i. e. Eucharist, purgatory, sacraments, the presence of an Anti-Christ, etc. Chelčický places an emphasis on the equality of all people, and therefore rejects the medieval model of society based on three classes of people. He felt that a society must be founded on love of one's neighbors and obedience to God through the examples set down by the apostles and the apostolic Church. Denying violence, he sets himself apart the Hussites. Chelčický's ideas of non-violence, human equality, truth, conscience, and suffering are still relevant in today's modern society.

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmacology and Toxicology Candidate: Mgr. Zuzana Šipulová Consultant: PharmDr. Jana Pourová, Ph.D. Title of Thesis: In vitro mechanism of action of selected quinazoline derivates on the respiratory system Theoretical part of the thesis summarizes basic facts about asthma bronchiale, its therapeutic options, and bronchodilating effect of quinazoline-containing agents. In experimental work, bronchodilating effect of selected synthetic derivatives of quinazoline (VN 014, VN 015) as well as their mechanism of action have been studied. We focused on a beta agonist effect, and used salbutamol (beta agonist) as a standard drug. The classical method of isolated quinea pig trachea in vitro was employed. We have found that both tested substances show a bronchodilating effect. The mechanism of action of VN 014 is probably not beta agonistic. Derivative VN 015 did not react as a competitive beta agonist, exact mechanism is not possible to determine based on the results obtained.
