Serological evidence of Lyme borreliosis

This bachelor thesis deals with Lyme borreliosis (Lyme disease). Lyme disease has a worldwide distribution. It is caused by several species of the Borrelia bacteria that vary according to geographic occurrence. In our country, this disease is transmitted, above all, by hard ticks of the genus Ixodes ricinus (Other genera are prevalent in other parts of the world.). These ticks are present in a wet environment, e.g. in the woods or in high grass. This poses a higher risk for people who stay or work in similar sites; i.e. lumberjacks, but also for children who spend their free time in the great outdoors. According to symptoms Lyme disease can be divided into three stages: stage 1 is called early localized Lyme borreliosis, stage 2 is called early disseminated Lyme disease, and stage 3 is called late disseminated, or persistent Lyme disease. The most frequently reported typical early sign is a rash called erythema migrans (EM). EM is an oval reddish skin eruption created at the site of the tick bite; this skin condition may gradually disappear. Other symptoms are: borrelial lymphocytoma, conjunctivitis, neuroborreliosis, Lyme carditis, etc. Therefore, generally recommended preventive measures should be used before each trip to an area infested with ticks in order to avoid contact with the ticks. Lyme disease is treated with antibiotics that are administered to a patient upon confirmation of the disease. As a rule the following antibiotics are prescribed: cephalosporin, macrolide, tetracycline or penicillin, depending on at what stage of the disease medication is prescribed and whether the patient shows typical Lyme disease symptoms. Laboratory diagnostics of Lyme disease relies on direct and indirect methods. The direct methods include: cultivation of Borrelia, hybridization, immunoelectron microscopy, and the PCR method (polymerase chain reaction) applied to prove the presence of borrelial DNA. Of all the methods, PCR is most often the method of choice. The indirect diagnostic methods are: indirect immunofluorescence, Western blot (WB) and ELISA tests (enzyme linked Immunosorbent Assay). These methods are used mainly to establish specific antibodies of the class IgM and IgG. The aim of this thesis was to analyze patients' samples for the presence of Lyme disease by means of the ELISA test and the Western blot, to test these methods, and to evaluate their results. The individual samples of serums, liquors and punctuates to be analyzed were provided by the Teaching Hospital in Pilsen, specifically by the Department of Serology and Parasitology. The samples must be diluted before examination. The method ELISA involved the use of a device manufactured by Dynex, DSX (an automated ELISA system). This device determines IgM and IgG antibodies in samples and facilitates analysis of up to 96 patient samples during a single measurement. The author of this thesis used this opportunity to try the manual determination of the ELISA test whose use is rather infrequent, especially in situations where a large number of samples need analyzing. The further examination of the IgM and IgG antibodies was accomplished with a Western blot, specifically by means of a commercial set for manual determination. The author applied the ELISA test to a total of 90 samples in the IgM class. 4 samples were borderline and 4 were positive. The same method was used to analyze the same 90 samples in the IgG class where 4 samples were positive. The Western blot method was used to examine 98 samples. The IgM class was borderline in 5 samples and positive in 18 samples. The IgG class was borderline in 5 samples and positive in 30 samples. Following laboratory examination the results of serological tests and clinical symptoms must be considered by a physician who suggests a suitable method to prove Lyme disease and exclude others.

Mapping appopriate Prague parks and trails for leisure activities focused on in-line skating for the pupils of a primary school

In the introduction of theoretical part, a sport discipline In line skating is described. Legislative aspect together with use of cycle paths is considered.The second chapter describes system of cyclopaths in the cities. Following chapters are describing the concept of parks and cyclopaths development in the rural area until 2020. Next, the work establishes criteria for evaluation of locations discussed in charter 5. Theoretical part of this work discusses in chapter six general rules for cyclopaths and park use. In chapter seven traffic signalization and in line skaters' equipment are discussed. Further chapters focus on sport and safety equipment. In the end of theoretical part of chapter ten and eleven I comment on particularities of in line skating education for children 7- 10 years old. Practical part describes mapping and evaluation of 10 selected cyclopaths located on the left side of Vltava River and close surroundings in chapter 12-13. Chapter 14 is dedicated to in-line skating and its categories. In the end of the practical part, the structure and methodology of teaching lesson of in-line skating is depicted together with description of basics of in line skating. Readiness of the beginning teacher for the work with an integrated pupil

This thesis deals with the issues of the work with an integrated pupil. It is particularly dedicated to the beginning teachers. It explains the important terms concerning the integration of the handicapped pupils into the basic school education. It describes the development, historical attitudes and explains the basic legal principles. Besides that, it is focused on the arranging of individual educational schedule, oral assessment and cooperation with the special institutions. A substantial part of this piece of work is the survey in the area of preparedness of the students of the field of study "Teaching for the 1.stage of the basic school", aimed at the subjective opinion of the readiness and skillfulness to educate the integrated pupil in her / his class.

Diagnostic of musical appreciation of primary school's pupils. Comparison of auditory and audio-visual versions of the tests

The thesis deals with the possibilities of the measurement of the musical abilities and skills of the pupils through the collective tests of musicality. The test was prepared in two version. The first was "classical" - only auditive, but the second version was "experimental", it use the visualization in all subtests. Following a concise overview of the several tests of the musicality for last 100 years, the author defines the fundamental terms as musicality, musical abilities and skills, test, visualization. Following this introduction, she concentrates on the methodology of the construct of the tests for pupils of 1st , 4th and 6th classes of the primary school. The most extensive section of the thesis is devoted to the evaluation, description and interpretation of results of both versions of tests in several classes with use of many tables and graphs. This research verified most of hypothesis and confirmed the needfulness of the utilization of visualization not only for the testing procedure, but also for the teaching of music education in school. The interconnection of visual and acoustic perception helps pupils to understand subject matter faster and easier and also concretizes abstract musical concepts, what is necessary in case of younger age above all. 1 1 1 1 1 1

Violin and violin playing have actually been existing only a few centuries. From a historical standpoint it is a very short time, yet extremely rich in a number of personalities, events and trends. When passing through the history of vilolin playing it would be interesting to stop at a number of important milestones. The overall development has been shaped by prominent personalities, who impressed to timeless memories of this instrument, their own contibution. In this way, thr most important violinists influenced the instrumental and expressive interpretation from a technical point of view. Some of them influenced, enriched or changed it even in a giute complex way.As a topic of my bachelor work I chose an essay on one of these outstanding personalities, a violinist Ruggiero Ricci. His artistic activity was not limited to concerts. Especially later he used stage experience to teaching and publishing activities. That´s why after a brief introduction, resume, and the chapter on the art of violin, I insert a chapter on Ruggiero Ricci´s pedagogical and methodological viewpoints. I had an opportunity to recognize it as his student at the International Music Academy in Pilsen. After the final summary I attach also a brief review of his most important recordings and a list of literature, as well as some interesting facts from my personal contact with this distinctive artistic personality.

Effective usage of telecommunication education environment

The topic of this bachelor´s thesis was effective usage of telecommunication‘s educational interface. The main aim of my thesis was to teach software and hardware versions of telecommunication‘s educational environment TIMS from Emona Instruments company. Also the goal of this paper was to design effective usage of educational environment in laboratories in the subject called Access and transport nets. There was a selection of three exercises suitable for these laboratories. These exercises were designed and implemented. Each of these exercises contains extensive theoretical introduction to solve problems, exercise assignment, block diagrams for visual understanding and for insight into measurement and detailed instructions to measurements. After all exercises have been completed exemplary protocols were created. The first exercise carried out was PCM coding and encoding in which students would be introduced with pulse code modulation, its advantages and disadvantages in way of acquiring high-quality broadcast. Second exercise was to focus on the introduction of quadrature amplitude modulation and phase shift keying. Target of the last exercise is to introduce students with binary data transmission via voiceband and factors affecting this transmission.

Gambling in Juveniles

I have chosen the topic of my bachelor paper ``Gambling in Juveniles{\crq}q because of its seriousness and topicality. The pathologic gambling has been more and more penetrating among juveniles to whom it causes, in the same way as in the adult population, more and more serious problems in the socio-economic sphere. This serious social pathologic phenomenon can be compared to drug addiction even though it does not damage human body in such fatal way. However, it affects the sphere of human psyche as well as the sphere of one{\crq}s social relations. And in the same way as drug addiction it has, in many cases, fatal impact on human life. My bachelor paper aims at finding out the extent and triggers of gambling in the population of high school juveniles in the area of Havlíčkův Brod. In the theoretic part of the paper specifics of gambling in children and juveniles are described as well as possible prevention of this phenomenon. Further diagnostic criteria of a pathologic gambler are described, the personality of a gambler is characterized, stages of development of pathologic gambling are defined and the corresponding legislature is dealt with. To fulfill the outlined goals, data were collected by means of the method of quantitative survey using anonymous questionnaires at six high school type schools (gymnasiums, professionally oriented high schools, craft oriented high schools). The questionnaires monitor students{\crq} experience with gambling machines and their reasons for gambling. The questionnaires further monitor students{\crq} awarness of the issue as well as prevention carried out at school they study at. On the basis of the survey I have checked three presumed hypotheses. Hypothesis 1: Students of craft oriented high schools yield more frequently to gambling than students of other high school types and gymnasiums. This hypothesis proved right. Hypothesis 2: Boys tend to yield to gambling more than girls. Also this hypothesis proved right. Hypothesis 3: Juveniles yield to gambling machines and to gambling more often under the influence of another person than from their own tending. This hypothesis proved wrong. The results of my bachelor paper can be used for the needs of further survey, for the needs of teaching and preventive measures at educational institutions and to increase the awareness concerning the gambling problematics in juveniles.

Author Jan Rendl in his thesis attempts to look at the world of ideas and educator Jan Gogola ml. through the eleven chapters in which each chapter somehow characterizes itself by Jan Gogola ml. and each of them somehow determines its creative ideas of it through the metaphor of a football match when Jan Gogola, with its characters, movies himself a teammate, as well as defensively. It gives goals with their situations as well as occasionally digging his opponents ankles. Jan Gogola ml. thus embodies one stage of the Department of Documentary Film at FAMU, which often stands at the intersection between teaching activities and Karel Vachek among students who applied by them during their seminars psychological methods that work must be peculiarly associated with the author of the film.

Upbringing and education of children with autism

The theme of this thesis is upbringing and education of children with autism. The text is divided into five parts. The first chapter is primarily devoted to the diagnosis. There are introduced the types of autism spectrum disorders and disorders, which can be associated with autism. Furthermore, I describe the typical symptoms of autism in social interaction, communication, imagination, and more. The following chapter deals with possible treatments and therapies and various educational methods, such as TEACCH program or applied behavioral analysis. The third part describes the possible ways of teaching and educating children with autism spectrum disorder. This extensive chapter informs about the types of educational facilities for children with autism and the importance of a special needs teacher and teacher's assistant. It also offers view of the family of a child with autism and a variety of educational strategies used in the home. The fourth chapter informs about organizations helping children with autism. The last part is devoted to case report of a boy who attends a special class for children with disabilities in kindergarten.
