Z-scan Fluorescence Correlation Spectroscopy Applied in Studies of Model Phospholipid Membranes
Kulakowska, Anna ; Hof, Martin (advisor) ; Konopásek, Ivo (referee) ; Dejneka, Alexander (referee)
6. Summary First of all, the issue whether and to what extent lipid mobility in the bilayer is altered when the bilayer is being deposited on the solid support was addressed. We designed a method that allows a direct comparison of the diffusion coefficients of labelled-lipids in the free-standing lipid bilayer of GUVs and that of the bilayer interacting with the mica surface (SPBs) in the glucose solution. The lateral diffusion analysis strongly indicate that the observed phenomenon is an unrestricted 2D diffusion. The results clearly show that the diffusion is slowed by more than 2 times for the interaction with the support. Moreover, we believe that the quantitative comparison of lipid diffusion in two frequently used model membranes can be helpful when comparing data reported in the literature. Additionally, the obtained data clearly indicate the existence of an inter-leaflet coupling. The lateral diffusion of investigated membrane probes and phospholipid analogues depend on the character and structure of the used fluorescent probe. The obtained lateral diffusion of the flavone probe, F2N12S is significantly faster than that of the fluorescent lipid analogues under study. Moreover, flavone probe exhibits approximately an order of magnitude faster flip-flop in comparison to the Atto633-DOPE lipid analogue...

Monitoring of the pain of the patient after the operation from nurse´s view
ŠIMÁČKOVÁ, Alexandra
The bachelor paper aims at monitoring pain during the post surgery period and it is concentrated on the attitude of nurse to it. When a nurse is capable of finding out pain, its quality and intensity in the shortest time possible, he/she promotes the comfort of a patient, improves his/her psychical state and can thus help significantly to successful healing of the surgery wound and total improvement of the health condition of the patient. A nurse{\crq}s active attitude to pain monitoring is in her/his own interest. She/he should be able to offer alternative ways of calming pain and to opt for an individual attitude to every patient. The theoretic part describes the origin of pain, its kinds, the nursing attitude towards it and different modes of monitoring of pain.Within the practical part we have set up two goals: to find out whether nurses monitor pain by patients after surgery and whether they use in practice the evaluating scale of pain. On this basis we have set up three hypotheses. H1: nurses at post surgery units monitor pain in patients having been operated on; H2: Nurses have no knowledge on scales evaluating pain; H3: Pain in patients hospitalized at post surgery units is not evaluated according to the evaluating scale. A quantitative method of research was chosen in the form of questioning and data collection using anonymous questionnaires.

Conflict in Colombia and its impacts on the neighbour countries
Lipková, Alena ; Kochan, Jan (advisor) ; Jeníčková, Martina (referee)
This work is dedicated to Colombia's conflict. Its objective is to sum up its implications on neighbour states. Venezuela, Ecuador, Brazil, Peru and Panama have to cope with the invasions of Colombia's guerrillas and paramilitary groups, waves of refugees who look for asylum in other countries in order to run away from acts of violence of their homeland or with increasing trend in cultivation of illicit crops. The United States of American intervenes significantly in the situation by trying with Plan Colombia to lower the quantity of cocaine coming from this region to American's markets. Brief conflict's development and characteristics of its actors, analysis of the relations among countries within the conflict and 3 possible scenarios of future development are used to achieve the main objective of the work.

Cannabis drugs among students of Prague medical faculties: prevalence of use and related risk behaviour
Bošková, Milena ; Vacek, Jaroslav (advisor) ; Gabrhelík, Roman (referee)
Objectives : The main goal of this work is to find out prevalence of use cannabis drugs among students of medical faculties in Prague and also find out a relationship between cannabis drugs abuse and risk behaviour. Sample : The research sample consisted of 686 medical students First, Second and Third faculty of Medecine of Charles University in Prague. The main criterium for entry into the research was that students had to be enrolled in first through fifth year of medicine. Students of medicine sixth grade were excluded from research because of their limited time schedule, which related to their studies. Methods : Research was provided by quantitative methods - questionnaire, which was structured and anonymous Results : It was found that lifetime prevalence of cannabis use among medical students compared with the earlier results of different studies significantly increased. The prevalence of cannabis use over the past 12 months was higher in men than in women. It was confirmed that men on average achieve higher scores on the CAST questionnaire, which maps the risk behavior regarding the use of cannabis. Those in the questionnaire CAST achieve higher scores, their risk behaviour was more related with cannabis drugs use. Conclusion : Lifetime prevalence of cannabis use among young people, including...

Influence of temperature and stimulus intensity on FAD transience in rat hippocampal slices in vitro
Rous, Matouš ; Otáhal, Jakub (advisor) ; Konopková, Renata (referee)
Aims: The purpose of this study was to characterize the influence of temperature and stimulus intensity on flavoprotein autofluorescence in rat hippocampal slices. Methods: Experiments were performed in hippocampal slices of adult male Wistar rats. Slices of 400 µm of thickness were maintained in submerged recording chamber, through which oxygenated ACSF was perfused (2 ml/min). Synaptic activation was being made by bipolar stimulating electrode (10s stimulus train 20Hz) placed in hillus of the dentate gyrus (Mossy fibers). FAD signals were detected by cooled 12-bit CCD-camera (RETIGA2000R). Results: Our findings demonstrate that FAD autofluorescence signals were significantly decreased at higher temperature. The data confirmed that FAD autofluorescence signals showed smaller light intensity in different layers of CA3 under the same conditions at 36řC compared to 26řC. Our data approved that under defined conditions FAD fluorescence signals have risen during stimulation. Keywords: Mitochondria, FAD autofluorescence, temperature, stimulus intensity, hippocampus, rat.
