Charles University in Prague University of Porto Faculty of Pharmacy in Hradec Králové Faculty of Pharmacy Department of Biochemical Sciences Department of Biology Candidate: Lenka Srbová Supervisor: PharmDr. Iva Boušová Ph.D. Prof. Alice Santos-Silva Title of diploma thesis: The influence of high concentrations of urea upon red blood cells and T-cells. Chronic kidney disease (CKD) is highly prevalent disorder worldwide with increasing number of patients. Several complications are associated with end-stage of CKD, namely infection, anemia and cardiovascular diseases. Infections are probably related to an acquired cellular immunodeficiency, essentially due to T-cell lymphopenia that may result in increased apoptotic susceptibility of T-cells. Anemia generally appears in patients with glomerular filtration rate lower than 60 mL/min/1.73 m2 . The red blood cells have shortened life span and are more susceptible to destruction in the uremic milieu. The lymphopenia and anemia have been associated with uremic toxins. The aim of this work was to evaluate the effect of urea on the viability of T-cells and erythrocytes (RBC), protein profile in RBC membranes, and induction of apoptosis in T-cells in vitro. RBCs and T-cells (alone or together) were incubated with pathological concentrations of urea (900...

Graphemics in the German Documents of Kutná Hora in the Period before the Hussites (1395-1420)
Janatová, Lucie ; Vodrážková, Lenka (advisor) ; Vaňková, Lenka (referee)
10. Summary In the area of Bohemia affected the German colonization in the 13th century the development of Czech and German language. The central Bohemia was under mining German colonist's influence, especially in Kutná Hora, in the historical medieval mining town, which became the second most important and the most powerful town in the area of Bohemia in the Middle Ages after the capital city Prague/Praha. The central theme of the dissertation is Graphemics in the German Documents of Kutná Hora (German: Kuttenberg) in the Period before the Hussites (1395 - 1420), in German: Der Schreibstand der Kuttenberger deutschsprachigen Urkunden aus vorhussitischer Zeit. The dissertation dealing with graphemics of twelve medieaval documents from Kutná Hora, of which original are deposited in the repository of municipal archive in Kutná Hora. I chose this theme because of my relation to this town, in relation to history and to privileged distinctive position among others medieval cities. The first chapter discribes the history of Kutná Hora, cultural development and economic expansion from the 13th century, after the revelation of silver mine. The follow-up chapters refer to the formation of new urban scribe and of the progress administration. The study of medieval documents require a detailed aquaitance of historical...

The Motivation of Pupils during German Lessons, and the Use of a "Language Portfolio"
Powered by TCPDF (www.tcpdf.org)

Orphan Nuclear Receptor TLX (NR2E1) in Regulation of Cell Reproduction and Differentiation
Raška, Otakar ; Kostrouch, Zdeněk (advisor) ; Otová, Berta (referee) ; Nedvídková, Jara (referee)
Nuclear receptors constitute a large family of transcription factors that are powerful regulators of animal tissue metabolism, homeostasis, tissue maintenance and development. They are particularly attractive for their ability to respond to the binding of hormones, metabolites, xenobiotics and artificially prepared molecules and transmit the interaction with these small lipophylic molecules to specific regulatory potential. In search for nuclear receptors that are likely to be critical for neural tissues in invertebrates and conserved during the evolution of animals, we have identified a close homologue of vertebrate TLX in a planarian Schmidtea mediterranea. Planaria represent very promising biological model systems for studies on tissue maintenance and regeneration. Planaria are able to resorb their tissues and use them as sources of energy during fasting and they re-build their bodies from neoblasts when food is plentiful. Our search in Schmidtea mediterranea's publicly accessible genome sequencing data indicated that planarian genome contains at least one gene with a high degree of similarity to vertebrate TLX. We cloned full length CDS (coding DNA sequence of cDNA) and characterized the gene functionally. This showed that the planarian and vertebrate NR2E1 are highly similar...

Study of cosmic muons in Tile calorimeter
Slovák, Radim ; Davídek, Tomáš (advisor) ; Drásal, Zbyněk (referee)
Main goal of this thesis is to learn about detector ATLAS, mainly about his hadron calorimeter Tile Calorimeter. Next step is get knowledge about processing date with program ROOT. It is necessary to test and monitor detector for example by muons from cosmic rays before his initiation. We analysed real experimental date from single module of Tile Calorimeter here. We found out signal-to-noise ratio. Then we separed out signal and noise and we found cells which muons flew through.

Microtubule-associated proteins in plants
Benáková, Martina ; Krtková, Jana (advisor) ; Vinopal, Stanislav (referee)
1. Abstract and key words MTs are one of the basic cellular protein structure. Their features and function are influenced and modified by group of other proteins, i.e. microtubule-associated proteins (MAPs). In the last decades, an extensive research on MAPs and their wide range of functions has been carried out. Therefore we are aware of the involvement of some of the MAPs in MT dynamics, other have been shown to have rather structural function. They bundle MTs with various cell structures, such as the other MTs, proteins, organelles, actin cytoskeleton or plasma membrane. Many described MAPs are homologous in the whole eukaryotic domain, for example MAP65 or EB1 (END BINDING 1) family, therefore it is interesting to follow if and how the functions of plant MAPs differ from their animal counterparts. On the other hand, there are many specific MAPs with unique functions in plants, e.g. ATK5 or SPR1 (SPIRAL 1). This Bachelor thesis is a survey on current knowledge of plant MAPs and it makes an effort to present their characteristic and functions in plant cell and organism. Key words: cytoskeleton, microtubules, microtubule-associated proteins, plant cell, growth and development

Disorders of iron metabolism in skin and chronic liver diseases
Krátká, Karolína ; Horák, Jiří (advisor) ; Ehrmann, Jiří (referee) ; Lata, Jan (referee) ; Martásek, Pavel (referee)
Iron is one of the important biogenic trace elements and its role in the mammalian body is indispensable. In nature there is another element with similar characteristics. Iron is part of a series of compounds that provide key functions such as cellular respiration and oxygen transport to tissues. It is also important for cell proliferation and differentiation, the regulation of gene expression and applies also in the immune system. Given that the effects of iron accumulation in genetic hemochromatosis have been investigated in detail, in recent years, increasing attention and concern about the consequences of iron accumulation also in other diseases. Because the results of previous studies are inconclusive and often mutually contradictory, the aim of this work to analyze and clarify the relationship between HFE gene mutations and iron metabolism in the pathogenesis and progression of some skin and chronic liver disease among genetic hemochromatosis.

Educators activities with hyperactive children
Matulová, Veronika ; Kovaříková, Miroslava (advisor) ; Hanušová, Jaroslava (referee)
The aim of thesis is to familiarize teachers and potentially parents with the issue of children suffering with concentration disorder. This should make a rather uneasy job using a handbook, which summarizes all communication procedures, methodologies and possibilities, much easier. From my personal experience, I'm aware of that teacher struggle working with these children; they don't know how to approach them efficiently. Teachers do have certain awareness from their universities; however, they still need to deal with resolving various situations to which they have not been prepared to. The theoretical part describes the terminology, aspects and gives overall information relating to ADD/ADHD clarifying the whole issue. It is indeed impossible to mention everything; however, I have chosen and included such information that will help the teachers, teaching staff assistants and parents to orientate within the whole thing. The experimental part, which is a quality research, contains case studies of boys diagnosed with ADHD. These quite clearly suggest teachers' work is not optimal. As for the output of this thesis, it is a handbook concluding from learning I have gained while working on mentioned case studies. It contains practical recommendations as well as suggestions useful when working with ADD/ADHD children.
