Figallová, Kateřina ; Klusák, Miroslav (advisor) ; Kučera, Miloš (referee)
As their café training programme comes to a close, the mentally disabled persons involved face an important decision: "What next?" They usually enter a follow-up transit programme, which helps them to deal with the transformation from the café environment to a different one. The objective of this dissertation was to map, understand and gain a deeper view into the process o f career choices among mentally disabled people who have made use o f the transit programme. The research group consisted of a group o f five subjects who entered the transit programme in 2003 and 2004. Data was collected until the end of 2005 when the transit programme was completed by the last of the five subjects. The data processing proceduře was inspired by the qualitative grounded theory of Strausse and Corbin (1999). The data were obtained particularly through active observation and ethnographic interviews with the subjects, and an employee of the transit programme (working consultant). The process of a career choice is considered to be an issue of professional identity. All subjects entered the transit programme with a certain idea about their future careers. We generally speak about a desirable professional identity, represented by jobs which the subjects find attractive for some reason. However, such attractive jobs are often...

The Citizens with Officially Cancelled Permanent Residency on Territory the Ceske Budejovice
HOLZÄPFELOVÁ, Vlasta
Permanent address is very important information for each citizen of the Czech Republic as it is closely linked to some rights and obligations exercised. A citizen of the Czech Republic may only be registered for permanent address at one place, usually where he/she has his/her family, flat or job. The permanent address location however does not have to be connected with the real place of residence and the bachelor thesis focuses on this reality, which often complicates citizens? lives. The origins and purpose of permanent address is examined from the historical point of view with links to the present situation. The bachelor thesis deals with the exact definition of the institute of permanent address of citizens of the Czech Republic and with a relatively new phenomenon of citizen registration, the possibility of so called official permanent address cancellation if the address does not correspond with the place where a citizen really lives. A house owner or a new lessor then exercises his/her right and files an application for cancellation of permanent address of a former lessee that is still registered at the address. During the administration procedure the lessee is asked to re-register voluntarily to another address upon valid right of use, if he/she fails to do that within a set deadline the address is officially cancelled, he/she is formally registered to a registration address, and the registration office of the municipality in whose territory the permanent address was cancelled then becomes the permanent address. The issue of permanent address cancellation brings numerous aspects not only in the context of the phenomenon of homelessness. The purpose is to point out the fact that not all the people with officially cancelled permanent address are homeless and still they are people who solve urgent living conditions and other existential anxieties. The bachelor thesis focuses on the territory of České Budějovice, its goal is to map the structure of the citizens with officially cancelled permanent address in the territory from the gender point of view. Another aim of the thesis is to find out in which parts of České Budějovice applications for permanent address cancellation are the most frequent and except others to propose a prognosis of the number of citizens with officially cancelled permanent address in České Budějovice. The fact showing certain relation between officially cancelled permanent address and escalated crime in selected area of České Budějovice seems to be a very interesting indicator. Demographic data processing is based on quantitative research performed upon own work experience by means of analysis of secondary data and trends from documents of opened administration procedures, their course and from already issued administration decisions on permanent address cancellation at the territory of České Budějovice.

Průšová, Pavlína ; Rýparová, Dominika (advisor) ; Mareš, Petr (referee)
Integration of catch-phrases of Jara Cimrman's plays into the everyday communication This bachelor thesis deals with a usage of catch-phrases from the Jára Cimrman Theatre plays in the context of our everyday conversation. In the theoretical part it explains fundamental basic terms, discusses the communication in general, defines common conversation. Furthermore, the text discusses the comic and its forms and briefly introduces the Jára Cimrman Theatre and its unique poetics. In the end of this part are presented conclusions about knowledge of these catch-phrases among common Czech speakers. The research answers three questions: What was the trigger to use the catch-phrase? What was the speaker's intention? What kind of influence the catch-phrase had on the listeners and the situation as a whole? Research questions are answered based on the method of participant observation and the results have shown that the main triggers are similarity of the communicative situation, linked meanings of the situation in a play and in reality or specific use of words. Speaker's meaning is the most often to entertain the addressees or to downplay the situation. This usually corresponds with the real effect of the catch-phrase usage. However, very important role plays the knowledge of the plays of Jara Cimrman - the better...

Studies on NKR-P1A and NKR-P1B receptors expressed in eukaryotic organisms
Ivanova, Lyubina ; Rychlovský, Petr (referee) ; Bezouška, Karel (advisor)
NK (natural killer) cells, with their ability to identify antigens and extraneous substances, available in the organism through various moleculary receptors, are an important component of the immune system. The NKR-P1A and NKR-P1B proteins belong to the lectin receptors of natural killer cells. Primary ligands of lectin receptors comprise terminal oligosaccharides of glycoproteins on the surface of target (e.g. tumor) cells. The interaction between carbohydrate structures on the surface of antigens and their binding partners on NK receptors is followed by triggering the effector function of NK cells against the targets. The NK cells and NK receptors findings and their interactions with ligands are greatly utilized in the treatment of cancer, viral and autoimmune diseases. Heterologous protein production in the eukaryotic organism brings a lot of advantages. Unlike the prokaryotic organism, the methylotrophic yeast Pichia pastoris has the capability of performing many posttranslational modifications resulting in production of biological active protein molecule. Usually, the P. pastoris expression system disposes of high level protein expression and is also generally regarded as being faster, easier, and less expensive to use than expression systems derived from other eukaryotes. In this thesis, I...

New findings of physiological and ecological aspects of hibernating bats
Ungrová, Zdislava ; Jahelková, Helena (advisor) ; Lučan, Radek (referee)
Bats can be found in almost all climatic zones. They have to deal with unfavorable conditions of environment to survive and reproduce sucessfully. Especially in temperate zones they have to deal with cold temperatures and lack of food during a winter season. To save energy bats enter a short, diurnal torpor or a hibernation. Usually, hibernation lasts all winter and is disturbed by short bouts of arousals. During both hibernation and torpor a physiological changes occur in bodies of bats. Body temperature decreases from endothermic levels, metabolic rates are reduced as well as respiratory rate and pulse. These changes are mainly influenced by weight and body size, diet, ambient temperature, sex and reproductive status. With the developement of new technologies, these changes can be measured more accurately than they have been in past, whether in a field or in a laboratory. But not all physiological processes are well explained. We have little knowledge especially about mechanism of arousal from hibernation. During awakening bats are infected by fungus Geomyces destructans that affects their immune system. In Norh America fungus causes a disease called White- Nose syndrome (WNS), that is responsible for death of millions of bats, especially endangered is Myotis lucifugus. In Europe, this disease is reffered...
