Utilization of biomass for the energy purposes

The theoretical part of work focuses on the issue of biomass used for energy purposes, in particular agricultural waste, which can be used as a substrate for biogas station. Furthermore, examines the composition of polymer substrates, which affects most biogas process. The aim of this work was to measure each sample of enzyme activity of biomass, which is used as a raw material for biogas plants.
Cultivation of microalgae and carotenogenic yeasts under stress conditions

In biotechnological industry we can register greater use of microorganisms like microalgae and carotenogenic yeast for production of selected substances or for valorisation of waste materials. Among metabolites produced by these microorganisms are carotenoids, which are natural pigments with antioxidant and other biological effects. Other significant produced substances are lipids, which represent large range of substances in living organisms. They form cellular membranes and serve as a reservoir of energy. These substances can be used in food, cosmetic and pharmaceutical industry and also in production of biofuels. Theoretical part is focused on description of selected genera of microorganisms, their cultivation and produced metabolites. Stress factors to which they were exposed are also mentioned, as are the analytical methods used for detection of produced metabolites. Next part is dedicated to the comparison of production properties of selected microorganisms depending on used stress factor. Increased production of lipids in yeast is observed in C. macerans and S. metroseus with higher concentrations of FeCl3. While using whey as a source of ccarbon, yeasts show decreased production of carotenoids. Highest production of lipids in algae and cyanobacteria was observed in S. acutus and D. quadricaudea. Production of carotenoids depended on applien stress factor and highest productions was observed in C. sorokiniana, C. reinhardtii, D quadricaudea and Coccomyxa sp.
Use of stress factors for the production of lipid substances by yeasts of the genus Metschnikowia

This Master´s thesis consists of two parts. The theoretical part is focused on the influence of various stress factors in the cultivation process, such as osmotic stress, oxidative stress, dehydration, cultivation time, pH or temperature on yeasts of the Metschnikowia genus. The practical part deals with osmotic and oxidative stress. Oxidative stress was performed under two different conditions (various cultivation time and temperature). The properties of yeast production were monitored mainly by gas chromatography and marginally also by flow cytometry. The analysis was performed for Metschnikowia pulcherrima, Metschnikowia andauensis, Metschnikowia chrysoperlae, Metschnikowia sinensis, Metschnikowia zizyphicola and Metschnikowia shanxiensis strains. The results showed that both oxidative stress and osmotic stress induce better lipids production. The amount of lipids, especially the fatty acid composition, also varied depending on the strains studied and the culture conditions used. The production of unsaturated fatty acids was further demonstrated in this work. The most suitable medium for the production of lipids and unsaturated fatty acids was the salt medium. It is clear that yeasts of the genus Metschnikowia are highly adaptable yeasts. Therefore, they might be potentially auspicious biotechnology producers.
The application of microorganisms to the degradation of lipids

This bachelor thesis is focused on the study of commercial products for degradation of lipids in waste water. This commercial products contain microorganisms with lipolytic activity. Theoretical part of this thesis describes lipids, lipases, mikroorganisms which produce lipolytic enzymes and basical principles of cleaning of waste water. In this part analytical methods of lipids isolation are presented too. The practical part of this thesis is concerned with the study of three commercial products. These products were tested for the content of microflora. Subsequently the restriction analysis was performed. Using this method the microorganisms were identified as Bacillus sp. Furthermore the growth curves were determined. This curves show growth of biomass. Then the pH during cultivation and lipolytic activity of microorganisms were determined. Lipolytic activity was determined by spectrofotometric method with using of p-nitrophenyllaurate.
Monitoring of fatty acids content during cheese production

This master thesis deals with the monitoring of fatty acids during production of natural Edam cheeses, i.e. natural hard cheeses with low heat curd, and of processed cheeses, which were produced from them. The theoretical part briefly characterizes the natural Edam cheese and processed cheese, the main attention is paid to lipids, fatty acids and the possibility of their determination. In the experimental part the content of fatty acids in the model samples of natural and processed cheese produced at Tomas Bata University in Zlín was determined. The method according to ISO 1735 was chosen for extraction of lipids from the samples, the method according to ISO 5509 using methanol solution of potassium hydroxide was chosen for the esterification of fatty acids. The resulting methyl esters were determined by gas chromatography with FID detection.
Assessment of fatty acids in processed cheese

This bachelor thesis deals with determination of fatty acids in samples of processed cheeses made at Tomas Bata University in Zlín. In the theoretical part, lipids and fatty acids are characterized. Subsequently, the issue of production, distribution and composition of natural and processed cheeses is discussed. The conclusion of the theoretical part is focused on the assessment of fatty acids, i.e. their extraction and the determination by gas chromatography. In the experimental part, the fatty acid content of the seven samples of processed cheese was compared on the basis of different temperatures and the time of their production. The ČSN EN ISO 1735 method was used for extraction of fat. The method with methanolic potassium hydroxide solution was used for esterification. The determination of fatty acid methyl esters alone was accomplished by gas chromatography with a flame ionization detector. Based on the comparison with the standards, 17 fatty acids were identified in the samples.
Valorization of waste animal fat by carotenognic yeasts

This diploma thesis deals with valorization of waste animal fat using carotenogenic yeast and conversion to biomass enriched with valuable substances. These microorganisms have the ability to utilize various waste substrates and convert them into interesting products such as carotenoids, lipids, ergosterol and ubiquinone. Four strains of yeast (Rhodotorula glutinis, Cystofilobasidium macerans, Rhodotorula mucilaginosa, Sporidiobolus pararoseus) were used. The theoretical part is focused on characterization of yeasts, enzymes and lipases, produced metabolites, yeast and description of analytical methods. In the experimental part, cultures were performed at four C/N ratios on media with different carbon sources, which were glucose, glycerol, fat, fat with added emulsifier, enzymatically hydrolyzed fat and fat with enzyme addition. Biomass was determined gravimetrically for all samples. Carotenoids, ergosterol and ubiquinone were analyzed on an HPLC/PDA equipment. The intracellular lipid content was determined by GC/FID. The production of these substances was different in individual strains depending on the carbon source and the C/N ratio. Rhodotorula glutinis is the most suitable for producing biomass and carotenoids. The highest amounts of ergosterol and ubiquinone were produced by Cystofilobasidium macerans. Sporidiobolus pararoseus had the largest lipid content in biomass. This diploma thesis was elaborated within the international project "LipoFungi".
Phase-separated systems biopolymer-lipid

This diploma thesis deals with experimental study of interaction based on polymer-lipid system. The main goal was to investigate interaction leading to phase separation. Three anionic polyelectrolytes were selected for this purpose: sodium hyaluronate, sodium chondroitin-6-sulfate and sodium poly(4-styrenesulfonate). The liposomes were formulated by sonication of lipid – cationic 1,2-dipalmitoyl-3-trimethylamonium-propane (chloride salt) and zwitterionic 1,2-dipalmitoyl-sn-glycero-phosphocholine. It was found that the phase separation occured at specific ratio of DPTAP and selected polymer. It was also explored that the strong electrostatic interaction which leads to the phase separation can be weaken by increasing the ionic strength in the sample. As we aspected the systems contain the hydrophobic domain and therefore these are able to incorporate hydrophobic substances in their structure.
Optimization of extraction of metabolites produced by selected strains of microalgae and carotenogenic yeast.

This bachelor thesis is focused on the production of microbial biomass using selected strains of cyanobacteria and carotenogenic yeasts. Subsequent preparation of extracts and study of their stability under different temperature conditions with the addition of the antioxidant tocopherol. In the experiment, 2 series of extracts with hexane and ethanol were prepared. The theoretical part of the work describes the studied metabolites carotenoids, chlorophylls, ubiquinone, ergosterol and microbial lipids. Furthermore, the used strains of cyanobacteria and carotenogenic yeasts are described here. Finally, methods for extraction and analysis of metabolites are described. The experimental part describes the used cultivation techniques, preparation of microbial extracts and used analytical methods. The results part of the work then contains a summary of the results of long-term stability tests of extracts. The results of the work clearly confirm the temperature dependence of the stability of individual metabolites. This phenomenon was most evident in chlorophyll and carotenoid pigments. Hexane appears to be the best solvent in terms of the stability of the substances used. The results of this work provide important information in terms of the economics of storage of these valuable substances usable in the food, pharmaceutical or feed industries.
Comparison of fatty acids profile of selected type of cheese

This master thesis deals with the determination of bound and free fatty acids in selected types of cheeses with/without „Protected designation of the Europen Union“ and their mutual comparison. The theoretical part focuses on a milk fat, a brief overview of cheeses, protected designation of the Europen Union, lipids, fatty acids and a possibility of their determination. In the experimental part, free and bound fatty acids profile in 12 selected samples of white mold cheeses was determined. The declared amount of total fat was verified. The cheeses were compared by a content of bound and free fatty acids, the profile of fatty acids was compared based on a type of milk used. Finally, the results were statistically treated by PCA method. The method according to ČSN EN ISO 1735 was chosen for extraction of lipids from the cheese samples. A methanolic solution of boron trifluoride was used for fatty acids esterification. The fatty acids methyl esters were determined by gas chromatography with flame ionization detection.
