Role of ABC transporters in pharmacoresistant epilepsy
Hanousková, Tereza ; Pávek, Petr (advisor) ; Trejtnar, František (referee)
The diploma thesis summarizes the recent knowledge on the possible role ABC transporters in mechanisms of refractory epilepsy. Approximately 30% of patients with epilepsy are refractory to treatment. An important characteristic of pharmacoresistant epilepsy (PE) is that most patients are at the same time resistant to several antiepileptics (AED) even though these drugs act by different mechanisms. It was suggested that one of the main cause of pharmacoresistance is the decreased drug uptake into the brain mediated by multidrug transporters in the blood-brain barier (BBB). The ABC transporters studied in relation to PE are the following: P-glycoprotein (Pgp, coded by MDR1 gene), some members of the multidrug resistance protein (MRP) family and the breast cancer resistance protein (BCRP). The over-expression of the ABC transporters was demonstrated in capillary endothelial cells of BBB, and astrocytes and neurons of epileptogenic brain tissue. Therefore modulation of ABC efflux transporters of the BBB could be a novel strategy to enhance the penetration of AED into the brain. Coadministration of inhibitors with less toxicity, higher potency and specifity to Pgp together with AED could thus reverse PE and achieve seizure control in epilepsy. The characterization of variations in the MDR1 gene and...

Fyzikální modelování změn difúze znečišťujících látek v oblasti křižovatky v závislosti na směru nabíhajícího proudění
Kukačka, Libor ; Jurčáková, Klára ; Kellnerová, Radka ; Jaňour, Zbyněk
The objective of this study is physical modelling of emissions from vehicle exhausts in street canyons within an urban area and the influence of the approach flow direction on contaminant spreading. The requirements of the similarity of the atmospheric boundary layer and a boundary layer in the wind tunnel are satisfied. For several flow directions mean velocity and concentration fields are measured within street canyon intersection inside symmetrical urban built-up area. Scalar fluxes are computed from measured data to quantify pollutant spreading. Results of the experiment show a significant sensitivity of the mean velocity field on the approach flow direction. The approach flow extensively influence contaminant spreading within the built-up area and therefore intensity of street canyons ventilation vary with the approach flow direction. The approximate flow direction with the most favourable dispersion conditions in the studied area is found out.

Charles University in Prague Faculty of Science Department of Organic and Nuclear Chemistry Carbocyclic analogues of nucleosides containing substituted bicyclic systems Michal Šála Ph.D. Thesis Abstract Prague 2010 Introduction Nucleoside and nucleotide analogues are of fundamental importance for all organisms. Therefore, nucleoside analogues are interesting target for drug discovery and development, mainly as potential antiviral and antitumor agents. A crucial disadvantage of natural nucleosides analogues is cleavage of the N-glycosidic bond by phosphorylases. Modification which increases resistance against enzymatic degradation is substitution of the sugar moiety furanose ring by a hydrocarbon ring. Many of such modified analogues - carbocyclic nucleosides1 - exhibit interesting antiviral activity. Several analogues containing conformationally locked bicyclic systems were also synthesized. Well known are carbocylic nucleosides with a fused cyclopropane moiety2 (bicyclo[3.1.0]hexane). Recently, novel conformationally locked carbocyclic nucleosides based on 2-oxabicyclo[2.2.1]heptane ring system were described3 (as precursors for carbocyclic locked nucleic acids). This thesis concerns the synthesis of biologically active compounds related to the carbocyclic nucleoside analogues. First part of the work is...

The narrative poetics of the novel La nieve del almirante by Álvaro Mutis
První kapitolu proto věnuji základním informaCÍm o životě a díle autora v kontextu kolumbijského literárního dění poloviny 20. století. Literární tvorba Álvara Mutise mne zaujala svou ambivalentní povahou v oblasti žánru, především pak přirozeností, s jakou propojuje svět poezie a výpravné tvorby, užívá prvků lyrických i epických a zachovává zároveň tematickou jednotu.

Sudden Cardiac Death - Social Impact on Family
JIROUŠKOVÁ, Alžběta
Abstract The Bachelor´s thesis "A Sudden cardiac death - and its social impact on the family" deals with a sudden cardiac death and its influence on functionality of a basic element of the state ? the family. The main task of this thesis is to determine a total psychosocial impact of a sudden cardiac death on the relationships in the family and verify which psychosocial rehabilitation options are offered to the bereaved persons. The expected application of this thesis in practice should improve living conditions for the bereaved persons and to provide them with more information about this diagnosis. In the theoretical part the diagnosis of a sudden cardiac death was described in detail. The death that occurs within one hour after the first signs is a frequently discussed topic in professional medical circles. The heart disease is considered as the main risk factor. Therefore, doctors primarily concentrate on this area in the treatment and prevention. Nevertheless 25 percent of patients who died of sudden cardiac disease had no signs of heart disease. So it´s important to think about familial syndrome. The major role in therapeutic activities is played by antiarhytmic therapy, kardiovertru-defibrillator implantation or catheter ablation. The main objective of the research was to determine how the current society is informed about the risk of NSS, its causes and possible prevention. The possibilities of psychosocial rehabilitation for families with psychosocial impact from diagnosed sudden cardiac death of a family member were analyzed here as well. The first set of questions was focused on lifestyle diseases that are closely connected with NSS and significantly increase the risk. The answers revealed the current society is quite well informed about lifestyle diseases, mainly cardiovascular diseases. Despite this knowledge the Czech population health situation has not changed significantly and civilization diseases are growing instantly. Another set of questions focused on the NSS and its causes. A lot of survey respondents put emphasis on SAC prevention in contradistinction to experts who already work with endangered patients and focus on managing and control of life threatening arrhythmias. Approximately half of the respondents identified themselves as endangered because of their health problems (cardiovascular disease, obesity, diabetes, etc.) More than 95 percent of the respondents expected impacts on the family and its relationships caused by the sudden cardiac death of a family member. Only 8 percent of the respondents put emphasis on potential social-economic changes. The research dealt with a problem whether the public is well-informed about possibilities of psychosocial rehabilitation. And the findings indicate it is not. More than 58 percent of the respondents try to solve problems themselves or with help of their family or friends. The responses to the survey confirmed the assumption that majority of the respondents (80 percent) know only about financial support from the state to the bereaved persons. And they would use only this state support if their family member died. Common people consider the using of psychosocial rehabilitation unavailable or they believe it is intended for the target group of psychologically ill people, mentally handicapped, alcoholics and drug addicts. A most of them don´t know that it could be used by the bereaved persons (91.16 percent). Due to the activity developed by the MMR as counseling for bereaved persons, it seems that it could get better in the near future. The offered service called ?consultancy for the bereaved persons? could get widely into the public knowledge.
