Identifikace genů nezbytně nutných pro normální průběh oplození a preimplantačního vývoje skotu v podmínkách in vitro

The aim of this work was to find and characterize genes that seem to be important for normal preimplantation development. We characterized three genes in more detail - centromeric protein F (CENPF; mitosin), ubiquitin C-terminal hydrolase-L1 (UCHL1) and nucleophosmin (NPM1; B23; numatrin; NO38). CENPF and nucleophosmin were shown to start their expression at late 8-cell stage, i.e. at major embryonic genome activation (EGA), and were hence supposed to be important during bovine preimplantation development. CENPF plays crucial role during cell division, especially by mediating the interaction of kinetochores and microtubules. Nucleophosmin is a multifunctional nucleolar phosphoprotein, whose most important roles are rRNA processing, chaperoning, ribosome biogenesis and centriole duplication control. Further, we characterized the role of UCHL1 during fertilization of bovine oocytes. UCHL1 is a deubiquitinating enzyme that controls cytoplasmic protein degradation, recycling of free ubiquitin from proteasome products and is involved in regulation of physiological apoptosis. More over CD is known to participate in a range of physiological processes such as apoptosis and tissue homeostasis, as well as in the regulation of angiogenesis and the production of peptidic antigens. The role of CD in pathophysiology is associated with several diseases such as Alzheimer's disease and cancer. Alzheimer's disease is a neurodegenerative disorder, which is generally considered to be the most common form of dementia. Progression of this disease is accompanied with the deposition of amyloid plaques (AP) in the brain, which leads to neurodegeneration. The AP is a fragment released from amyloid precursor protein (APP) cleaved by secretases1 . High levels of CD were found in cerebrospinal fluid of the Alzheimer's patients2 . The aim of this bachelor work is to create a laboratory training which is made for teaching and demonstration laboratory furnace temperature control with applications created in Control Web 6.1 The role includes the regulation and capture waveforms measured and desired temperatures, allows the timing of the required temperature. The application is processed for example burning of small products. Allows archiving of temperature distribution and also monitoring the instantaneous values of the desired and measured temperatures with the possibility of switching heating via the Internet. Regulation of lobbying in the Visegrad Four

The subject of this thesis is regulation of lobbying in the countries of the Visegrad Group (V4) -- specifically statutory regulation of lobbying. The work gives a comprehensive overview of the development of regulation of lobbying in V4 member states from the very beginning until now. Main aim is to answer questions related to lobbying such as: "is there a lobbying regulation in V4 members", "what is the quality of that regulation" and "is there any common signs shared across the national regulation of lobbying". To achieve the desired objective thesis uses descriptive, comparative and analytical methods. Thesis consists of two main logical units, which are divided into five chapters, an introduction and a conclusion. The first unit contains three introductory chapters. This section is devoted to the history of lobbying, the definition of lobbying and theoretical concepts of regulation of lobbying. The second unit is completely dedicated to the legal regulation of lobbying in the V4 countries. Part of this unit is also the final comparison and analysis of the legal regulation of lobbying in these countries and answers to questions mentioned in the introduction.

Application of local approximators for control of real mechatronic system

Cieľom práce je aplikácia lokálnych aproximátorov pre riadenie reálnych mechatronických sústav pomocou metódy dopredného riadenia predstavujúcej zaujímavú alternatívu k metódam využívajúcim globálne aproximátory. Po ukážkových príkladoch funkcie lokálnych aproximátorov bol navrhnutý algoritmus implementovaný pre riadenie dvoch sústav, elektronickej škrtiacej klapky a výukového modelu magnetickej levitácie, predstavujúcich vysoko nelineárne a nestabilné sústavy. Skúmali sme, či riadiaci algoritmus bude mať pozitívny vplyv na presnosť regulácie, ďalej bola skúmaná jeho schopnosť prispôsobiť sa zmene parametrov sústavy a tiež prípadná možnosť jeho implementácie pre mikrokontrolér znížením vzorkovacej frekvencie. Výsledky ukázali, že riadenie založené na lokálnych modeloch zlepšilo riadenie v porovnaní s jednoduchým PID regulátorom a že má schopnosť adaptability. Veľmi výhodné sa zdá byť jeho použitie pre zariadenia umožnujúce vzorkovaciu frekvenciu do 1 kHz.

Modern methods of control of heating systems

The aims of the thesis are modern methods of control of heating systems. Basic principles of regulations are explained and methods of control of heating sources and exchangers are explored. Heating system of family building is projected with two types of control – equithermal regulation with thermostatic heads and individual room control (IRC). Both variants are simulated in program TRNSYS 16.1. From the results are concluded advantages and disadvantages of examined systems of control.

Thermoelectric cooler construction

Bachelor thesis will concern description of Peltier modules and detailed description of thermoelectric phenomena’s on which is principle of these modules based on. Thermoelectric materials and their properties will be mentioned as well as different applications of Peltier modules. Heat losses of the cooler will be described. Onwards it describes mathematical model of Peltier modules and their coefficient of performance. Also mounting of modules, types of power supply and types of regulation are mentioned. Experimental part consists of calculation and designing of cooling assembly.

The effect of bailiff territoriality on the reclamation of receivables, the length of debt recovery and other aspects of court-ordered distraints

The area of individual reclamation of receivables in the form of court-ordered distraints has undergone significant changes since the year 2000. The initial practice, when the claims used to be enforced directly by judicial officers, proved to be largely ineffective and with difficult to apply. The Ministry of Justice has responded by adopting an amendment to the Act (Act no. 120/2001 Coll., on bailiffs and executor activities), which introduced the independent bailiff offices. Then, the law itself understood the bailiff authorities as a separate entrepreneurs. Currently, in the Czech Republic, there can be seen the distribution of the market with distraints according to the Pareto rule. Approximately 20% of bailiff offices manage over 80% of distraints. It is obvious that this is an unsatisfactory condition for the majority of bailiff offices. In response to this fact, the Czech Chamber of Bailiff offices calls for the introduction of territoriality, which would eliminate by law the market environment from the market with distraints and every single bailiff office would have a stable allocation of distraint proceedings. The analysis carried out clearly shows that there are significant differences between bailiff offices in terms of speed and in success of the debt recovery. The actual demand for the introduction of territoriality would not merely be a reallocation of the market with distraints. Putting it into a practice would greatly influence the transactional costs of business and non-business entities in the Czech economy. The thesis comprehensively describes the issue of territoriality and quantifies how much the debt collection will decrease in case of its introduction. In the conclusion, an alternative draft is formulated, which might, according to the author, allow effective regulation and supervision of the activities of bailiffs' offices without compromising the inherent efficiency of the reclamation of receivables in terms of market environment.

THE LATEST TRENDS IN PUBLIC PROCUREMENT LAW

This thesis deals with public procurement law in the Czech Republic. It captures the latest trends in public procurement law, together with the amendment to the Public Procurement Act, which entered into force on 1 April 2012. The opening chapter introduces the concepts that are closely related to the issue of public procurement and corruption. There are explained key institutional control mechanisms in the Czech Republic and criminal liability of corruption. The analytical part of this thesis describes the changes, which the amendment brought into the Public Procurement Law. These changes are mainly assessed in terms of impact on the suppression of corruption. This thesis presents and analyzes the arguments of proponents and opponents of the amendment. The analysis is accompanied by a brief look at the development of legislation on public procurement in the European Union in general and in two selected countries of the European Union. The conclusion contains a summary assessment of the issue along with an outlook for a possible further developments in the legal regulation of public procurement. Based on the analysis, the thesis leads to the conclusion that the amendment of public contracts, despite some shortcomings, is leading to a more transparent procurement environment.

Possibilities for developing sustainable transport in Prague

This diploma thesis is concerned with the issue of sustainable transport. In the theoretical part, the emphasis is placed on setting the concept of sustainable transport into a broader context; it also describes various factors influencing the choice of the means of transport, which is crucial for transport planning. Furthermore, it describes a set of diverse tools used for transport regulation. At the end of the theoretical part, it mentions specific approaches implemented in selected cities all over the world. The practical part is based on a significant personal research, which helps to understand the issues of transport in a selected area of Prague. On the basis of this research, it subsequently suggests possible measures that might lead to a decrease in the intensity of automobile transport in the aforementioned area. Most of those measures can be similarly applied in other city areas as well.
