A study of the properties of tablets from the mixture of directly compressible xylitol and microcrystalline cellulose.

Název v anglictine: A study of the properties of tablets from the mixture of directly compressible xylitol and microcrystalline cellulose. Summary: The thesis deals with the study of properties of tablets from a mixture of directly compressible xylitol, namely Xylitab® 300, and microcrystalline cellulose, namely Microcel® MC-12, in the ratio of 1:1. (This ratio was arrived at after preceding experiments concerning the strength of tablets made only from Xylitab® 300, and from a mixture of Xylitab® 300 and Microcel® MC-12 in the ratio of 3:1. The strength was insufficient in these cases.) The focus of the study was the dependence of the tensile strength and disintegration time of the tablets on the compression force, the addition of lubricants, namely magnesium stearate and sodium stearyl fumarate (Pruv®), with concentrations of 0.5% and 1%, and a 50% addition of model active ingredients, namely acetylsalicylic acid and ascorbic acid. The compression forces used were 10, 12.5, and 15 kN; tablets containing the drugs were only compressed with a force of 15 kN. Tablets with a weight of 0.5 g and a diameter of 13 mm were compressed using material testing machine T1-FRO 50 TH.A1K Zwick/Roell and they were further subjected to a strength test using Schleuniger Tablet Tester 8M and to a disintegration time test...

Doxorubicin cytotoxicity and metabolism in MCF7 cell line

RNDr. Lenka Skálová, Ph.D. Title of a diploma work: Doxorubicin cytotoxicity and metabolism in MCF-7 cell line. Doxorubicin (DOX) is ranked among anthracycline chemoterapeutics, significantly participant in the treatment of solid tumors, inclusive breast carcinoma and haematologic malignancies. DOX is metabolized to 13-OH-doxorubicinol (DOX-OL) produced by carbonyl reductase 1 (CBR1). DOX-OL is lower antineoplastic, but more cardiotoxic compared to the parent drug. Aim of this work was research of reduction DOX and testing possible increasing cytotoxicity of DOX through inhibition his reductase. For experiments has been selected human breast cancer cell line MCF-7. Initially has been elicit optimal composition of cultural medium and optimalized process on sample preparation for HPLC. In the study of DOX metabolism in cytosol the MCF-7 cells act oracin (potentional chemoterapeutic) as significantly kompetition's inhibitor of DOX reductases. The aim of this master thesis is the study of adhesive properties of the polyesters of glycolic acid and lactic acid, branched with tripentaerythritol and plasticized with triacetin of various concentrations. The theoretical part deals with the bioadhesion, bioadhesive polymers, testing of bioadhesion, and plasticizers. The practical part is focused on the study of adhesiveness, measured as maximal force needed for the detachment of the polymer from the model substrate. The influence of model substrate, type of polyester, plasticizer concentration and incorporation of the drug on adhesiveness was studied. It was found, than 20 % of triacetin is sufficient for decreasing the viscosity of the polyesters. Even the lower concentration could be used in the further studies. The cork base can serve as model substrate for screening of the adhesivity. Lifestyle of the Straight Edge Suculture Youths

This thesis handles with the lifestyle of Straight Edge youth subculture. This subculture is identified by refusing alcohol, drugs, tobacco, sexual promiscuity and emphasizing moral compassion. First the thesis describes the lifestyle in general, including the typical attitudes, values and freetime activities. Further it reflects subcultures, their place within society, their specifics, and maps the academic studies of (post)subcultural theories. Following part describes the Straight Edge subculture in detail and is asking the question whether this subculture can fulfill the function of the substition of religion. The qualitative analysis then focuses on the lifestyle of the individual followers, their perception of the community, the relationship with society and selfpresentation. Additionaly the thesis presents the ethical and moral dimension of Straight Edge and delas with the problem of commercialism of subcultures.

Computational Study of the Interactions of Transition Metal Complexes with Biomolecules Using Quantum Chemical Methods.

It has been proven that platinum complexes are active in anticancer treatment as well as several other transition metals. There is an effort in recent medicine to replace cisplatin complexes by drugs with smaller side effects. This work focuses on the reaction of 5'-dGMP (2'-deoxyguanosine- 5'˗monophosphate) with a platinum complex PtIV (dach)Cl4 (dach=diaminocyclohexane) forming PtIV (dach)Cl3(N7-cGMP) complex. The explored reaction can be divided into two (independent) steps: cyclization of phosphate and substitution reaction where a new complex with coordinate- covalent bond between platinum atom and nitrogen N7 of guanine is formed releasing chloride particle. Here I studied geometry parameters which are important in the above mentioned reaction. The reaction course is observed from the thermodynamic point of view. The structures were optimized at the DFT level with B3LYP functional in basis set 6-31G* and PCM/UA0 solvation model. The energy parameters and electron density distribution were computed at the B3LYP/6˗311++G(2df,2pd) level in the D˗PCM/sUAKS solvation model.

Pharmaceuticals and their metabolites are excreted by patients into wastewater in considerable amounts. Conventional methods of wastewater treatment are often unable to fully eliminate these substances. Pharmaceuticals and their metabolites can get into the environment and affect the ecosystem. One way to streamline wastewater treatment is the application of phytoremediation technology in the form of root plants. In this work, the ability of plants to accumulate and degrade the cytostatic drug doxorubicin was studied. At the same time phytotoxic effects of this substance were monitored. For this purpose different systems of cultivation of plants were used: hydroponics in common reed (Phragmites australis (Cav.) Dále byl sledován vliv podávaného hypolipidemika atorvastatinu na změnu v expresi těchto endoteliálních markerů. Pro zobrazení exprese VCAM-1 a endoglinu byly využity imunohistochemické metody a ke kvantifikaci jejich exprese stereologické metody.
