Utilization of mouse polyomavirus derived virus-like particles for cargo delivery into cells

and key words Mouse polyomavirus-derived virus-like particles composed from major capsid protein VP1 (MPyV VP1-VLPs) are interesting structures for use as a delivery system of various cargos into cells. VP1 protein self-assembles into icosahedral particles of 45 nm in diameter that are hollow highly regular nanoparticles. In this work, model small molecule cargo, Cyclodextrin-Based Bimodal Fluorescence/MRI Contrast Agent, was encapsidated into MPyV VP1-VLPs. The cargo was stably associated with VLPs and was delivered into mammalian cells using these VLPs. To prevent VLPs entrapment in endolysosomal compartments and increase the potential of VLPs applications, MPyV VP1 protein was modified by insertion of histidine-tag (6 histidine long sequence surrounded by glycine and serine) sequences into VP1 surface loop DE, because histidine modification of synthetic systems had enhancing effect on endosome escape and cargo delivery. With the use of in Bac-to-Bac® baculovirus expression system His-VP1 protein was expressed in insect cells and a variety of VP1-assemblies was obtained: long tubules and small 20nm VLPs formed from VP1 with 4 histidine-tags in DE loop, and novel VP1 nanostructure, which we named nano-jumpers, formed from VP1 with 2 histidine-tags. Robot for Robotour 2010

The objective of this master's thesis was to get acquainted with an autonomous outdoor robot competition - The Robotour and further to study the capabilities and information about a robot designed on the DITS FIT Brno VUT. Next part of the project was to study various planning and localization algorithms and to make a proposal of a functional solution, allowing the robot to participate in the competition. To be specific, the goal of this work was to use the acquired knowledge for implementation of an antonomous software so the robot can start in the year 2010 race. The solution also included several related mechanical modifications. The final solution fulfills all the competiton rules and is capable of taking part in this year's plant.

Evaluation design of civil airports effectivity indices

Introductory part of this thesis concludes the development of air traffic, division of airports in the Czech Republic according to the Ministry of Transport, according to the Law on Civil Aviation and according to the Coding of the airports (Requirement L 14). The aim was to realize theoretical definition and division of output and economic indicators. After that comes choice of appropriate airports from the European Union. The choice has been made on the basis of the number of check-in passengers at the airport in Prague-Ruzyně. Eight most suitable airports have been chosen from all twenty-seven member countries of the EU. The following step is comparative analysis of these chosen airports on the basis of specified output and economic indicators for the years 2005-2009. The year 2010 has not been included, because the chosen airports did not complete their annual reports, balances and statements of profit and loss. From these sources the values for calculation of individual indicators have been chosen. These indicators have been shown in charts, evaluated by means of graphs in Excel Programme and further they have been characterized in words. The aim of the thesis was also the evaluation and comparison of output and economic indicators of effectiveness and after evaluation, proposal of methods of evaluations has followed.
Air Cargo

The bachelor thesis looks at the system of air cargo. The first part deals with division of aircraft cargo compartments into classes in accordance with safety rules. The following part describes the usage of air containers and pallets and refers to a possibility of transportation of special kinds of cargo. The main ways of cargo transportation by various kinds of aircraft are explained. Some special cargo aircraft are described in more detail. The last part of the thesis is concerned with ground support. The requirements which have to be supplied by an airport are mentioned and also the technical equipment for handling of cargo.
The New Cargo Aircraft for ACR

The purpose of this bachelor project is selection the new cargo aircraft for ACR. There is charakterized present condition of military cargo aircraft in Armed Forces of the Czech republic and causes of its renovation. The part of a project is exhaustive listing of demands on the new aircraft. Each parts contains subareas of this issue. Major part of this project is paid to choosing inner cabinet of aircraft from this category and their brief description. Last part solves choosing of the best candidate from the existent group.
Innovation of information system in rail freight transport in the Czech Republic

This thesis deals with innovations in freight transport, especially from the perspective of information and communication technologies. It identifies key strengths and weaknesses, opportunities and threats in an implementation of innovations. Its main contribution is the linking of current informatics trends with specific trends of the rail freight sector in the Czech Republic in relation to the regulation and harmonization of the European Union. One of the important objectives of today's enterprise informatics is support of the strategic management. Based on the test the validity of the working hypothesis, according to which companies in the rail freight don't use strategic management tools enough, it makes recommendations for innovation in the sector. The thesis also briefly presents some innovative applications of Czech IT developers. It is a comprehensive material from which the Chief information officer (CIO) can get basic orientation in the rail freight sector. It may also serve as an inspiration for companies that want to enter the market, or are already operating on it and want to achieve competitive advantage through innovation.
