This bachelor thesis discusses polymers that are used in medicine and healthcare. In general, it addresses the field’s already known products and their possible further research and spreading of their use. Most people imagine medical polymer applications mainly in the forms of syringes, catheters, latex gloves, and the like. This text’s goal is to point out how much potential there is in further research of the material allowing for qualities as varied as biodegradable pill coating and high resistant dental implant.
Bioproduction, Characterization and Application of Selected Biopolymers

Proposed doctoral thesis deals with microbial production of biopolymers (polysaccharides and polyesters) using renewableble inexpensive resources as carbon and nitrogen source. At first, production of extracellular metabolites, mainly pullulan and poly-L-malic acid, by polymorphic microorganism Aureobasisium pullulans was studied. Further part of thesis was focused on production of bacterial biopolyesters polyhydroxyalkanoates (PHA). PHA are produced in the form of intracellular granules by various bacteria species. During the study of PHA production, sythesis of poly(3-hydroxybutyrate) and technologically significant copolymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) was investigated as well with regard to waste substrate utilization in biorafinery concept. Spent coffee ground seemed to be applicable waste in biorefinery proces. One of the coffee wastes used for PHA production was spent coffee ground hydrolysate which was converted to PHA by Burkholderia cepacia. Moreover, copolymer of 3HB and 3HV was accumulated when SCGH was used as a sole carbon source, no precursor had to be added. Another coffee waste, which was utillized by Cupriavidus necator H16, is represented by oil extracted from spent coffee grounds. Hydrolysis proces of waste substrates had to be optimized, to reach the highest possible saccharide concentration. Due to the hydrolysis process several toxic compounds can be formed, hence, influence of hydrolysis procesings on polyphenols content in hydrolysates was monitored. Waste substrates were also tested as potentional complex nitrogen source for PHA production. Hydrolysates of cheese whey and chicken feather were used as inexpensive complex nitrogen source for PHA production by C. necator, when waste frying oil was used as a carbon substrate. The finnal part of the thesis is dealing with possibilities how the PHA can be processed into atractive aplication forms. Liposomes with partial content of PHB had been prepared, moreover, for improving of antioxidant activity and antimicrobial effect the waste coffee oil was added. Micro-fibrous PHB materials were prepared for the purposes of water filtration. Higher filtration effectivity was observed when composite PHB fibrous material containing active charcoal or metal oxides was used. Important part of thesis is represented by study of degradation processes of prepared PHA-based aplication forms. Various microorganisms were tested on PHA depolymerases production. Biodegradability of PHA materials was tested in of composting conditions by standard composting test IS/ISO 20200. Changes in mechanical properties of polyurethan, where polyol was partialy replaced by PHB, were tested depending on the exposure of composting condition.
Preparation and Characterisation of Polyelectrolyte Complexes in the System Dextran-Surfactant

This bachelor thesis deals with the study on preparation and characterization of the polyelectrolyte complex formation in the dextran-surfactant system. The aim of the thesis was the investigation of the influence of the ionic strenght on the formation of the interactions in the studied system. For these purposes, components having opposite charges (DEAE-dextran hydrochloride and anion-active sodium dodecyl sulphate) were selected. For individual measurements mainly the methods of light scattering, dynamic and electrophoretic light scattering in particular, have been used. In the thesis have been studied interactions and aggregation concentrations of individual components having the potential to form polyelectrolyte complexes. It was verified in all the studied samples, by measuring the intensity of scattered light and the size of Z-diameter of particles, that the increase in sizes of particles is caused by the formation of these complexes. There were also measured additional characteristicts of the system such as pH or conductivity. Their dependences provide comprehensive view on the formation of complexes in studied system and can be utilized for other measurements and possible future applications, especially for purposes in medical applications.
Biotechnological production of selected biopolymers employing Haloferax mediterranei

Aim of this bachelor thesis is to study of production of selected biopolymers, polyhydroxyalkanoates (PHA) and extracellular polysaccharides (EPS) by Haloferax mediterranei. The first part contains theoretical background including properties of biopolymers, advantages and purposes of commercial production, characteristics and positive aspects of cultivation of extreme halophile archaea Haloferax mediterranei. The experimental part is focused on study of effects of concentration of waste substrates, such as proteolytic whey hydrolysate and feather hydrolysate on PHA and EPS production. The effect of medium volume and its correlation with acceess of oxygen on biopolymer production were studied as well. PHA, in particul copolymer P(3HB-co/3HV) was analysed by gas chromatography with FID detection. The content of 3HV in PHA to 15,09 % was achieved using medium which contains 25 % proteolytic whey hydrolysate without glucose supplementation. Medium with 10 % of feather hydrolysate and 10 % of feather hydrolysate with 30 g/l glucose proved to be suitable in volume 50 ml in 250 ml Erlenmeyer flask. Haloferax mediterranei was able to produce biomass and accumulate PHA using protein substrate without need of saccharide supplementation.
Preparation of mixed colloids of humic acids with biopolymers

This bachelor’s thesis discusses an applicability of innovative colloidal forms of humic acids, prepared by a complexation with biopolymer chitosan. In experimental part, suitable procedures have been proposed and optimized for the preparation of hydrogels and xerogels made of these abundant polymeric substrates. In characterization of resulting materials, major attention was paid to the determination of the water content and the swelling ability. Besides, mechanical properties were studied as well. The development of a desired material aims at an industrial or agricultural utilization of sorption abilities and a tendency to immobilize various pollutants in the environment. Another advantage is represented by the ability to control the release of humic substances from the gel to environments.
Extraction of Poly(3-hydroxybutyrate) from Biomas

Polyhydroxyalkanoates are a group of biopolymers which occur in cells of microorganisms. These biopolymers can be considered as alternatives to commonly used petrochemical materials such as polypropylene and polyethylene due to their physical and chemical properties. This bachelor thesis focuses on isolation of particular kind of PHA from biomass and contains both experimental and theoretical part. It consists of information about the origin and usage of PHA, challenges and problems of their biotechnological production and mainly various ways of downstream. Experimental part includes comparison of PHA extraction from biomass done by two ester solvents. Studied parameters were molecular weight of the polymer, solvent extraction ability and their resistance against hydrolysis. The main goal of this thesis was to verify the best process conditions of isolation done by nonhalogenated bio-solvent.
Dual-focus FCS in colloidal research

Tato práce se zabývá studiem fluorescenčně značeného hyaluronanu, konkrétně rhodaminylamino hyaluronátu sodného (Hya-Rh, 40 kDa), pomocí dvouohniskové fluorescenční korelační spektroskopie (2f-FCS). Nejdříve byla prostudována literatura týkající se využití FCS techniky v koloidní chemii a při studiu polymerů, přičemž následně byly shrnuty veškeré poznatky o využití 2f-FCS metody. Na základě prvotních měření byl zjištěn vhodný postup přípravy a způsob uchovávání vzorku Hya-Rh používaném pro následující experimenty. Záhy byly prostudovány možné vlivy koncentrace Hya-Rh na jeho difúzní charakteristiky jak ve vodě, tak ve fyziologickém roztoku. Následně bylo studováno chování Hya-Rh a vliv koncentrace solí alkalických kovů ve vodných roztocích těchto solí a fluorescenčně značeného hyaluronanu. Poté bylo sledováno chování Hya-Rh v závislosti na koncentraci velmi nízkomolekulárního hyaluronanu (VLMW HA, 404 kDa) v čisté vodě i ve fyziologickém roztoku, přičemž získané výsledky byly mezi sebou porovnány. Nakonec bylo využití 2f-FCS metody celkově zhodnoceno a popsáno z hlediska studia chování fluorescenčně značeného hyaluronanu v roztocích.
Characterization of polyelectrolyte complexes using structural and thermal analysis

This master thesis deals with study of chitosan-lignohumate, chitosan-polystyrenesulfonate, chitosan-alginate and chitosan-carrageenan polyelectrolyte complexes. The work was motivated by research of finding suitable alternative substance for lignohumate. The molecular weights of substances were characterized using SEC-MALLS. A degree and a character of the interactions between polyelectrolyte were studied by isothermal titration calorimetry and dynamic light scattering method. The calorimetric experiments proved that decreasing concentration of samples causes decreasing of heat flow. The best calorimetric measurements were provided by adding chitosan into polymer solution. The interactions between chitosan and polyanions and influence of mixing order were proved also by measuring intensity of zeta potential, Z-average of particle size and turbidity. New chitosan-based materials have a big potential in agriculture and medicine.
Hydration of biopolymers

Presented bachelor's thesis focuses on the study of hydration of selected biopolymers – humic acids, chitosan, hyaluronic acid (low molecular weight and high molecular weight) and dextran. Thermogravimetric analysis TGA is used t determine the original moisture content of the samples. Two calorimetric methods were used for subsequent observations of changes in their chemical and mechanical properties in the presence of water – differential scanning calorimetry DSC and perfusion calorimetry.
Preparation of methacrylated hydroxybutyrates monomers and polymers

The aim of this work is the study, design, and preparation of methacrylated derivatives of poly(3-hydroxybutyrate) and the subsequent preparation of polymers from the prepared precursors. A total of six compounds were prepared. The prepared compounds include methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate prepared by alcoholytic depolymerization with monofunctional alcohol and their methacrylated derivatives. The preparation of ethylene glycol terminated oligomer of 3-hydroxybutyrate prepared via depolymerization using ethylene glycol in two ways is also an essential part of the work. The first way involves depolymerization using conventional heating and the second one explores the preparation of a series of oligomers with different molecular weights using microwave heating. Methacrylated derivatives of the prepared oligomers were also prepared. The prepared compounds and their actual preparation are characterized using multiple analytical methods including VPO, NMR, GPC, FT-IR, LC-MS, and DSC. A series of polymers were obtained from the prepared compounds by radical polymerization using purple light (with wavelength 405 nm) as the energy source. The prepared polymer networks were characterized by DSC and their tensile strengths were tested.
