Thao Tranová Student: Lukáš Ficek Title of Thesis: The preparation of tablets by laser sintering This work focuses on the preparation of orally dispersible tablets by selective laser sintering (SLS). The present project is a pilot study conducted on the 3D printer Sintratec Kit at the Department of Pharmaceutical Technology. SLS is a 3D printing method based on the fusion of powder particles using a laser beam that sinters the individual particles together in the layers of material.1-2 The matrix forming polymer used in work was Kollidon® VA64 which was combined with two pigments namely Candurin® Gold Sheen and NTX Ruby Red. In addition, mixtures of the polymer with a co-processed dry binder Prosolv® ODT G2 and a physical mixture of mannitol, silicified microcrystalline cellulose, and crospovidone at the concentrations of 20%, 40%, and 60% were used. The influence of printing parameters, especially laser speed, on tablet quality was tested. NTX Ruby Red pigment was selected for final tablet printing. Storytelling according to the TAT in the State of Moderate Stress: The Effect of Defense Mechanisms on Coping with Moderate Stress.

This diploma thesis is focused on storytelling in the state of moderate stress. The theoretical part includes an introduction to the issue of stress, defense mechanisms, Thematic Apperception Test and then explains in detail theory of defense mechanism according to Phebe Cramer and also Defense Mechanism Manual which is used for evaluation of TAT defenses. The aim of empirical part of this thesis is to find out whether moderate pain which is caused by the fact that participants holds their hand in front of them (in position of "staying power") has effect on number of occurrence of defense mechanisms in narratives according to TAT. The stories are narrated during the staying power position. The second aim is to verify whether number and quality of used defenses has effect on the duration of the staying power position. The simple regression analysis was used to find out that there is no significant relationship between number of used defense mechanisms and the level of experienced pain. However, it is shown that higher usage of defense mechanisms has positive effect on duration of staying power position. This effect is even stronger if only matured defense mechanisms are included in the model, which is projection and identification.
Regenerative Medicine in Ocular Surface Reconstruction: Advancing Cell-Based Therapies for Limbal Stem Cell Deficiency

Limbal stem cell deficiency is a disease caused by the impairment of limbal epithelial stem cells (LESCs), leading to the replacement of the corneal surface with nontransparent conjunctiva, and its treatment is not standardly available worldwide. This work focuses on preparing cells for treating both uni- and bilateral forms of the disease using advanced cell- based therapy. It investigates LESCs in various scaffolds (fibrin, nanofibers) and explores non-limbal cell sources like oral mucosal epithelial cells (OMECs) under standard complex and xenobiotic-free culture conditions. The study uses immunofluorescence and gene expression to detect stem cell markers, proliferation, and differentiation capacity of cultured cells. Moreover, long-term OMECs storage with different media and cryoprotective agents and the healing properties of the amniotic membrane (AM) were also examined. We found that adding interleukin-13 to the culture media enhanced LESCs' stemness. LESCs on fibrin gels showed higher expression of stemness markers, while those on polymers expressed more mesenchymal ones. Using standard and xeno-free media, we successfully prepared OMEC-containing cell sheets on fibrin gel substrates. Behavior and Complexity in Household Finance

This thesis explores mechanisms underlying differences in financial decision-making, includ- ing retirement saving and mortgage choice. Chapter 1 investigates the long-term effect of extrapolative expectations on retirement savings in subsidized employer-matched 401(k) ac- counts in the U.S. The chapter introduces a deviation from rational expectations in reproduc- ing worker-level retirement contribution rates over their tenure. Chapter 2 generates a novel U.S. data set and examines the correlation between financial literacy, mortgage rate attain- ment, and refinancing. Empirical estimates motivate the introduction of financial literacy and the level of search effort as dimensions of heterogeneity that generate differences in mort- gage repayments. I find that losses from low financial skill levels and search effort amount to almost 10 percent of the total loan, implying significant effects on the budgets of financially unskilled and inexperienced U.S. households. Chapter 3 leverages these empirical findings and embeds a micro-founded mortgage search framework in a standard heterogeneous agent model of consumption and saving. Music as a Value of Human's life

The thesis considers an issue of the value of music in a human life. In the theoretical section the author interprets the term music and its roles in human life using available literature. Moreover, she focuses on the influence of a music on human psyche, in context of emotions, thinking, fantasy and motivation. The author also deals with the description of musical development of a personality. The author is also attentive to issues of values and value orientation in a human life. In the empirical section the author works with three groups of respondents; namely, with active musicians (i. e. playing a musical instrument or singing). She included professional musicians (group of students of music) and amateur musicians (group of working people with a musical hobby and a group of university students with a musical hobby). There are 33 respondents in the sample aged from 20 to 30. The comparison of these groups is based on the method of halfstructured interview. Issues of the development of musical activities and influences that mattered in this development, a motivation to musical activities, an influence of musical activity on personal life of the respondents (especially in context of relationships), a value and function of a music in an overall value orientation of respondents are compared. Public Policy to Prevent Childhood Obesity and the Role of Food Production and Marketing

This thesis is a case study focused on a relationship between public policy to prevent childhood obesity and the role of the food production and marketing in the Czech Republic. The emphasis is especially given to the institutional framework of food marketed and produced for children and its social concequences, because childhood obesity is one of them. The results from the analysis are then confronted with views and opinions of children and their parents, with how they feel about food advertising. Conclusion: advertising and production of food designated for children is not regulated in the Czech Republic. On governmental level only basic standards and rules are given and self-regulation is given precedence. Parents are aware of the huge influence of advertising on their children and that is why they are not satistied with the current situation. They would appreciate stronger legislative regulation of marketing and production of food for children.
Surface ozone influence on native vegetation: results based on ozone visible symptoms and stomatal flux

Regarding the vegetation, the most affected areas by high levels of surface ozone (O3) are the mountain ridges. Our study has been carried out in the Jizerske hory Mts. High O3 levels together with the convenient environmental conditions for stomatal conductance could be a threat for the health of recovering ecosystems in this area. The aims of this study was both to assess the influence of O3 on vegetation in the Czech mountains and to provide recommendations and outlooks for possible future using of relatively new methods (visible symptoms and stomatal O3 flux modelling) used for O3 impact assessment on native vegetation in the field; that means physiologically relevant methods for the determination of O3 influence. During 2006 and 2007, O3-like visible symptoms were assessed on the leaves of seven species at four sites. Symptoms on only two species (Fagus sylvatica L. and Rubus idaeus L.) have been determined as O3-induced. To our knowledge, it is the first study in the Czech Republic in which the O3-like symptoms on native plants have been verified by the Ozone Validation Centre for Central Europe. Binding of trace elements to colloids in MSW landfill leachates

1 SUMMARY The association of trace elements with colloids in leachates from closed and active municipal solid waste (MSW) was studied using the cascade filtration/ultrafiltration method. A study was carried out after long-term monitoring of seasonal variations of the leachate quality. Both landfills are situated in the vicinity of Prague and the closed landfill site represents a serious environmental risk for part of this city. In the first step, the evaluation of the long-term seasonal changes in the main and trace element contents in leachate from the closed landfill was performed and supplemented by investigation of precipitation/dissolution processes in the solid phases. It was shown that the quality of the leachate is strongly influenced by rain events, which are responsible for the dissolution of calcite and Fe oxides in the landfill body. This process leads to the release of Cu, Zn and Cd bound to these solid phases and to an increase in their concentrations in the leachate. The contents of the other element are significantly decreased by simple dilution. Therefore, the mobility of all the studied elements increases substantially during rain events. This doctoral dissertation summarizes the effects of statins and inflammation on TGF-β1 signaling both in vivo and in vitro, especially the role of endoglin in the endothelial cells. Transforming growth factor-β (TGF-β) is a multifunctional factor that regulates various cell specific functions in atherogenesis. Endoglin (TGF-βRIII, CD105) is known as accessory TGF β receptor, and it is able to modulate activity of TGF-β1 and TGF-β receptors. Previous studies demonstrated relation between endoglin and eNOS expression in blood vessels suggesting crucial role of endoglin in endothelial cell functions. SUR Theory - Psychotherapeutic Training Community as a Tertiary Family

Objectives: Theoretical contribution of the research is to extend knowledge about the impact of psychotherapeutic training on its participants. Community and group psychotherapeutic training is an endemic phenomenon which does not appear out of the Czech Republic and has not been systematically investigated yet. Goal: The main goal of my dissertation is to detect the influence of psychotherapeutic community training on its members. Subjects: The subject of investigation is 168 introspectional texts which participants of psychotherapeutic community wrote immediately after completing the training. The texts were developed from the diaries which the members wrote during the 5 years persistence of training. Research methods: Qualitative analyses of documents based on application of methods, techniques and procedures of "grounded theory". Content analysis and focus groups were used as complementary methods. Conclusions: This study formulates an integrated theory of psychotherapeutic approaches including cognitive behavioral methods, psychodynamic psychotherapy and expressive therapeutic techniques concentrated in a system of community and psychotherapy group training.
