The influence of maternal care on the development of the circadian clock during ontogeny
Straková, Lucie ; Sumová, Alena (advisor) ; Spišská, Veronika (referee)
This bachelor thesis focuses on the importance of maternal care in early ontogeny on the development of circadian rhythms and further physiological development. The central clock oscillator (SCN) is most strongly synchronized by the external environment through the rhythms of light and dark. However, during ontogeny, when the main and peripheral oscillators first develop, maternal signals are the main synchronizers for the offspring. Thus, integration between mother and offspring is fundamental to the proper synchronization of the developing offspring's circadian clock, and the mother, through her behavior, can negatively or positively influence its setting. Attention is therefore paid to breastfeeding as a nowadays neglected factor of motherhood that has a strong influence. The composition of breast milk has proven beneficial effects over artificial diets, and breastfeeding has benefits for the health and circadian rhythm of mothers and their offspring. Premature infants are exposed to artificial conditions and weaned from maternal care, similar to offspring in cross-foster experiments. The observation of these effects on the development of their circadian rhythms again only confirms the importance of maternal care. Key words: maternal care, circadian rhythms, ontogeny, breastfeeding
Non-enzymatic roles of kinases and phosphatases - the case of MTMR9 and AAK1
Knop, Filip ; Macůrková, Marie (advisor) ; Harnoš, Jakub (referee) ; Soukup, Vladimír (referee)
Enzymatic roles of kinases and phosphatases in almost every aspect of cellular life are well described in a wide variety of examples. Lately the role of the same proteins independent of their catalytic activity is being increasingly appreciated. In this work, we focus on two proteins, mammalian MTMR9, and Caenorhabditis elegans SEL-5/AAK1. MTMR9 belongs to the myotubularin-related family of lipid phosphatases (MTMR) and is known to be a pseudophosphatase, a catalytically inactive member of the MTMR group. SEL-5/AAK1, on the other hand, is characterized by its kinase activity with at least two putative substrates identified so far. We described the localization of MTMR9 to early secretory pathway and its colocalization with known ER-to-GA compartment (ERGIC) markers. We also identified several possible MTMR9-interacting partners, such as RAB1 and MTMR6, whose localization and/or activity could be potentially regulated by MTMR9 binding. Disruption of proper MTMR9 levels led to an alteration in WNT3A secretion and subsequently to a reduced activity of the Wnt signaling pathway. Similarly, we identified SEL-5/AAK1 role in two separate Wnt-regulated developmental processes in C. elegans. Firstly, SEL-5 along with other members of the retromer complex regulate a proper QL.d migration. Secondly, excretory...

FROM THE END
Krausová, Radana ; Čapek, Jan (referee) ; Tobola, Ondřej (advisor)
The bachelor thesis focuses on designing a cover for a multisensor fire alarm. The project originated in collaboration with the company LITES, which specified the requirements for the alarm cover. The main objective is to create a product suitable for various interior types and capable of integration with other end elements in the space. The design is conceived for installing the alarm either directly on the ceiling or for recessing it into the ceiling. Manufacturers and sellers of these devices are increasingly recognizing the importance of visual communication in the user interface of fire alarms. If the end element is too conspicuous or detracts from the overall impression of the room, it can evoke considerable aversion from the user. The work is based on simple geometric shapes such as cubes, spheres, and cylinders, and their subsequent perforation. This decision stemmed from the effort to streamline the shape of the cover from unnecessary elements. For the alarm design, I chose the shape of a cylinder, which best meets the requirements for installing the alarm and its function in the space. In my opinion, it is important to perceive the alarm as an accessory in the space, not just as a technical element. This idea significantly changes the approach to the product and therefore also the approach to its design.

Harmonic Analysis of the Double Bass Bridge
Čalkovský, Lukáš ; Votava, Tomáš (referee) ; Hájek, Petr (advisor)
This bachelor's thesis focuses on the research of string instruments and their mechanical properties. It explores the double bass bridge and its importance. On a model created in the SolidWorks program, modal analysis was performed using the ANSYS software, which determined the frequencies and shapes of the individual modes. Harmonic analysis was also performed and analysed. Part of the investigation was to analyze the possible cause of the formation of so-called wolf tones. The double bass bridge does not exhibit any special characteristics in the double bass range, so it is not the cause of this anomaly.
Refractory high entropy alloys - preparation and properties
Urbancová, Kateřina ; Jan, Vít (referee) ; Dlouhý, Ivo (advisor)
Tato diplomová práce zkoumá mikrostrukturu a mechanické vlastnosti dvou vysokotavitelných vysokoteplotních slitin (RHEA), MoNbTaVW a HfNbTaTiZr, připravených mechanickým legováním a slinováním (SPS). Zavedení železa do MoNbTaVW významně ovlivnilo mikrostrukturu a mechanické vlastnosti. Vedlo to ke složitějšímu fázovému složení a zvýšené tvrdosti, která dosáhla maximální hodnoty 1077,8 HV v kombinaci s lomovou houževnatostí 4,3±0,3 MPa·m0,5. Tato slitina vykazuje rovnováhu mezi vysokou tvrdostí a střední lomovou houževnatostí, což dokazuje její potenciál pro aplikace vyžadující odolnost proti opotřebení. Tato práce zdůrazňuje důležitost vzájemného působení složení slitiny, délky mletí a podmínek spékání při úpravě mikrostruktury a vlastností RHEA pro pokročilé aplikace.
Friction of Titanium Surfaces in Implantology
Heto, Matyáš ; Odehnal, Lukáš (referee) ; Ranuša, Matúš (advisor)
This bachelor thesis focuses on the frictional properties and wear resistance of Ti6Al4V alloy. This alloy is suitable for use in orthopaedic implants such as the first metatarsophalangeal (MTP) joint in the big toe. The aim is to investigate different surface treatment methods that can reduce friction and wear of this material. The effect of applying DLC coatings and creating microtextures or microstructures on the surface is described. Metal on metal pair tests have been carried out using profilometer and tribometer in pin on plate configuration. The results show a noticeably higher coefficient of friction for the Ti6Al4V alloy than for the compared CoCrMo alloy. DLC coating significantly helped in reducing the friction and wear of Ti6Al4V. This cannot be said for the two microstructures tested, which did not have a significant effect on the observed tribological properties. The results indicate the importance of surface treatment of Ti6Al4V alloy for use in implants.

Šenkeříková, Veronika ; Tobiáš, Martin (referee) ; Fojtášek, Jan (advisor)
This bachelor's thesis is dedicated to suspension systems and their typology. Furthermore, the basic geometric parameters of these systems are discussed in the thesis. Types of suspensions (axles) are classified according to the interdependence of individual wheels. Subsequently, the thesis provides an overview of simulation software for chassis development and testing in the virtual phase of development. These are divided according to computational principles, result evaluation, and environment. The last section addresses issues related to chassis testing and its properties as a whole, testing individual chassis components to verify durability etc. The thesis includes specific representatives of testing conditions. For each chassis component, a representative from each chassis subsystem is selected. This thesis provides a current overview of methods and tools used in the automotive industry for development and testing. It demonstrates the importance of both virtual testing and testing on testbeds for already finished products.
