Upconverting nanoparticles as contrast agents for biomedicine

Upconverting nanoparticles (UCNP) represent an interesting class of contrast agents for biomedical applications due to their ability to convert infrared light into the visible or even UV region of the spectrum. The aim of this work was to prepare UCNP with bound hyaluronan (HA), a natural polysaccharide, and to investigate their interaction with cells in vitro. The reproducibility of the synthesis of β-Na(RE)F4 UCNP was demonstrated in a series of 15 consecutive preparations. Particles with different ratios of RE elements (Yb, Gd, Y, Lu, Er, Tm) with emission spectra covering the range from NIR to UV and advanced core/shell type UCNP with different combinations of core/shell material were prepared. The layer-by-layer shell growth method allowed to control the particle size. Hydrophobic UCNP were incorporated into oleyl-HA micelles. The UCNP@oleyl-HA micelles were stable in buffers and in cell culture media, where they effectively limited the quenching of UCNP luminescence. The second method of UCNP modification consisted of sorption of HA-furanyl onto particles followed by photochemical crosslinking. Subject C - reactive protein in microbial wor/d ofburnt Thesis Aim: Familiarization with burn injury - highlighting of importance of complex care of burnt patients, familiarization with specific characters of infection of the burn trauma Monitoring of most frequent bacteria of the burnt patients, monitoring of CRP values Detection of the CRP IeveI differences of sportsmen and of humans who do not do sport for selected bacteria Method: Thesis is divided in three parts. Burn trauma, possible infection complications and most frequent types of bacteria are described in the clinical part. Personal assistance in supported living for people with learning disability

The main topíc of this thesis is assistance in supported living for people with mental retardation. The theoretical part of this essay defines these concepts in the context of the new trends in the care for people with mental retardation. Then we focus on persona! assistent himself; we describe his personality, demands on his professional skills and his approach towards the clients. Then there are introduced some psychological aspects of assistance. We pay attention on motives for this helping profession, on burn-out syndrom and ethical aspects of assistanť s work. The aim of the practical part is to explore the current situation in assistance in Czech republic. The goal of my investigation was to find out using mainly qualitative research methods, who is the typical assistant in supprorted living, why did he choose his profession and how does he experience his work. The results pointed out, that there is a large specific group of young women working in this profession as well the lack of other age groups of assistants and the lack male assistants. The most frequented motives for assistance are the posibility of using the skills obtained by previous education and experience, the meaningfullness of this work and the wish to provide high quality service for mentally handicaped people. Preparation, characterization and testing of blood derivatives for applications in regenerative medicine

Platelet products can be used, thanks to the broad range of bioactive molecules, either as a supplement for cell cultering in vitro alone or for development of cell- or cell-free scaffolds in diverse fields in regenerative medicine. The aim of this study was to prepare several types of platelet products. The concentration of selected molecules were observed. Subsequently, these products were tested with cell cultures in vitro alone or in combination with nanofibres scaffolds prepared by electrospinning or centrifugal spinning. It was found out, that platelets products contains chemokine RANTES and growth factor PDGF in the highest concentrations. It was further discovered the content of pro and antiinflammatory in terleukins and other growth factors. Platelet lysat in concentration 7% is sufficient to replace FBS in keratinocytes and fibroblasts cultures. In the other experiments, platelets in different concentrations were adhered to the scaffolds prepared by electrospinning and centrifugal spinning. Thus prepared scaffolds promote the proliferation and viability of all tested cell types in dose-dependent manner. Imaging of Soft Tissues in Micro-CT

X-ray radiography is a noninvasive imaging technique for a visualization of internal structures of investigated samples. In past years, imaging of biological samples in a micro-CT is gaining in popularity. The disadvantage of this technology is a low ability to display soft tissues like muscles, fat tissue or nerves due to their low intrinsic contrast. The aim of this study was to modify a conventional micro-CT MARS in order to improve the scanning of the soft tissues and to create a protocol for simple and cheap fixation of ex-vivo soft tissues for micro-CT scanning. In the study, a modified micro-CT MARS was used together with a photon-counting detector Timepix Quad with the resolution 30µm. The micro-CT was afterward tested on the phantom and also on the real soft tissues. For scanning soft tissues in the micro-CT, the ethanol fixation method was invented. Hearts and lungs from laboratory mice were either fixated in 97%, 50% ethanol solution or in a series of ascending ethanol concentrations. Images were acquired after 72, 168 and 336 hours. The resulting images were compared among themselves and with the native specimens. How precede to civilisation diseases (complex education and motivation text about health life style)

Název: Jak předejít civilizačním chorobám How to prevent civilization diseases Cíle práce: Mým cílem bylo posoudit názorovou orientaci a informovanost studentů gymnázií a odborných učilišť v oblasti prevence civilizačních chorob. Toto šetření mělo být východiskem pro přípravu informačního výukového materiálu. Metodika: Metodou výzkumu bylo dotazníkové šetření, mapující jak názorovou orientaci a postoje respondentů v problematice životního stylu, tak i informovanost v oblastech: pohyb, strava, škodliviny a psychohygiena. Dotazník vyplnily 2 skupiny 17-18 letých respondentů, kterými bylo123 gymnazistů (60 dívek a 63 chlapců) a 122 učňů (60 dívek a 62 chlapců) z Prahy a okolí. Výsledky: Výsledky výzkumu ukázaly nedostatečný stupeň informovanosti v obou skupinách respondentů, i když studenti gymnázií projevili dle předpokladů lepší znalosti než studenti z odborných učilišť. Nebyla však potvrzena hypotéza, že dívky mají ve srovnání s chlapci lepší znalosti o zdravé výživě.
