The multimodality of the narrative in the video game Kentucky Route Zero

Tato diplomová práce zkoumá narativ videohry Kentucky Route Zero na základě multimodálního přístupu. Cílem práce je popsat funkce a specifika jednotlivých sémiotických modů a analyzovat způsob, jakým se tyto mody podílejí na formování multimodálního narativu. První část práce poskytuje teoretický základ pro následnou analýzu. Nejprve popisuje problematiku vztahu narativů a videoher. Následně představuje teorii multimodality, multimodální koncepty a s nimi spojené přístupy. Nakonec se věnuje také videohře Kentucky Route Zero, jejímu příběhu, hernímu i uměleckému žánru a záměru herních designérů. Výzkumná část práce se nejprve věnuje analýze jednotlivých sémiotických modů jako je jazyk, obraz a zvuk. V kontextu celé videohry zkoumá jejich specifika, funkce a limity pro vytváření významů v rámci narativu. Kromě toho popisuje možnosti interakce a jejich vliv na narativ. Tyto poznatky následně využívá v rámci analýzy narativních elementů, přičemž se zaměřuje na způsoby kombinace jednotlivých modů, na jejichž základě je narativ videohry Kentucky Route Zero formován.
Preparation, characterization and testing of blood derivatives for applications in regenerative medicine

Platelet products can be used, thanks to the broad range of bioactive molecules, either as a supplement for cell cultering in vitro alone or for development of cell- or cell-free scaffolds in diverse fields in regenerative medicine. The aim of this study was to prepare several types of platelet products. The concentration of selected molecules were observed. Subsequently, these products were tested with cell cultures in vitro alone or in combination with nanofibres scaffolds prepared by electrospinning or centrifugal spinning. It was found out, that platelets products contains chemokine RANTES and growth factor PDGF in the highest concentrations. It was further discovered the content of pro and antiinflammatory in terleukins and other growth factors. Platelet lysat in concentration 7% is sufficient to replace FBS in keratinocytes and fibroblasts cultures. In the other experiments, platelets in different concentrations were adhered to the scaffolds prepared by electrospinning and centrifugal spinning. Thus prepared scaffolds promote the proliferation and viability of all tested cell types in dose-dependent manner. Polystyrene photoactive nanomaterials producing singlet oxygen

The increasing number of multidrug-resistant strains of bacteria call for alternatives to antibiotic therapy and, more generally, for the antimicrobial material as a component of prevention. Of particular interest is the photodynamic inactivation of bacteria and other pathogens caused by photogenerated singlet oxygen. This work is focused on the field of photoactive polymer nanofiber membranes and nanoparticles, generating singlet oxygen, suitable for medical applications. We prepared different types of photoactive modified polystyrene nanofiber membranes with encapsulated or externally bound porphyrin photosensitizers. These materials efficiently produce highly reactive and cytotoxic singlet oxygen capable of restricted diffusion into to the external environment. Our results demonstrate the crucial role of wettability for materials of this type with a short diffusion length of generated singlet oxygen, illustrate the effect of temperature and indicate their potential use as multifunctional materials. Due to their antimicrobial properties, these materials are suitable alternative to antibiotics and local antiseptics. Detection and visualization of native and artificial chondrogenous tissue using biophysical methods

Tissue engineering is a rapidly developing field, based on using scaffolds made from natural or synthetic materials in combination with cells and stimulating factors for the replacement of damaged or lost tissue. It is very important to evaluate qualities of these scaffolds, which are necessary for cell proliferation and their mechanical support. The aim of this study was to develop a suitable scaffold for chondrocyte proliferation, scaffold functionalization and detection of cells and proteins of extracellular matrix (ECM) using methods of fluorescence and confocal microscopy. Another aim was to test an appropriate scaffold on a big animal model in vivo. Several scaffolds from natural and synthetic materials, in the form of microfibers, non-woven textiles, gels and foams were prepared for this study. Scaffolds were seeded with chondrocytes and cell adhesion, proliferation and synthesis of ECM proteins were detected. Methods of fluorescence microscopy, confocal microscopy and second harmonic generation (SHG) were used for visualization of cells and proteins. A hydrogel based on fibrin and hyaluronan was used as an scaffold for osteochondral defect regeneration in minipigs study. Study of the photodynamic inactivation of prions by phthalocyanines.

Transmissive spongiform encephalopathies, also called prion disorders, are fatal neurodegenerative diseases affecting mammals. In patients, the pathological prion protein (PrPTSE ) accumulates in CNS and causes death. Prions possess high binding affinity to surfaces. Moreover, they are highly resistant to conventional sterilization procedures which rise the risk of nosocomial transmission from patients in subclinical stage of prion disease through medical tools. In the thesis, we evaluate the efficiency of photodynamic inactivation (PDI) for prion decontamination. The PDI is induced by photoactivation of phthalocyanine (Pc) derivates AlPcOH(SO3)2, SiPc(OH)2(SO3)1-3 or ZnPc(SO3)1-3. Pc exposed to light generate reactive oxygen species, mainly singlet oxygen (O2(1 ∆g)). Production of O2(1 ∆g) in aqueous solution was confirmed by iodide method, quenching by NaN3 and oxidative degradation of uric acid. The photoactivation of Pc in infectious brain homogenate led to elimination of PrPres signal (= proteinase K-resistant PrPTSE fragment) below the detection limit of western blot by using nanomolar AlPcOH(SO3)2 concentration. Determination of endocrine disruption potential by early identification of selected substances with health risk, used in consumer products

Substances of natural or synthetic origin, capable of activating or inhibiting the functions of cellular receptors and thus contributing to the modulation of the neuroendocrine system through the pathways of hormonal signaling ("disrupting" the endocrine system) are referred to in the literature as "endocrine disruptors" (ED). Endocrine disruption as a form of mild systemic toxicity of chemicals and their mixtures is intensively addressed at both scientific and legislative levels. In 2018, progress has been made towards the introduction of a comprehensive European Union legislative framework for ED when a consensus was reached on the scientific criteria for their assessment and identification. ED research is supported by the European Union as a significant task of preventive medicine. Under the various Framework Programs for Research and Innovation, the EU has provided funding to dozens of projects and has supported projects under Horizon 2020 to develop appropriate methods for testing ED. The legislation on e.g. pesticides and biocides, chemicals (REACH), medical devices, cosmetics, food contact materials, water, toys, and workplace protection has been updated. The aim of this bachelor's thesis is to analyze the image of the USA and Americans in North Korean propaganda. The thesis briefly defines the concept of propaganda, describes its characteristics and categorizes its types and purposes. It deals with the specifics of North Korean propaganda and examines its techniques and effectiveness. The core of the thesis focuses on the image of the USA and Americans in relation to historical events which form North Korean Anti-Americanism. Furthermore, it thoroughly analyzes the visual aspects of the portrayal of the USA and Americans in North Korean propaganda and searches for generally valid principles and the way they are applied. Comparison of badminton match in men's and women's singles

Title: Comparison of the badminton match in men's and women's singles Objectives: The goal is identify the most effective a choice combinations of the first three shots, leading to the acquisition point in singles between the best Czech male and female players. And also to discover determine and compare which was the usage of different types of serve, return and the third shots. Methods: Comparison of differences in badminton match between genders was based on indirect observation - video analysis. Statistics were used for data processing. Results: The results shower huge difference in tactical approaches among men and women players. Women preferred long high service and most of the service returns were played into back backhand court area and the third shot usually routed into back forehand court area. It means that the most played shot was a clear shot and the most occupied part of court was a back backhand corner. Despite more successful was when women players played drop by their third shot. Domination of short backhand service was seen by men players, the second shot routed the same direction like women players second shot - to the back backhand court, but also to the front part of the court. Developmental, pathobiochemical and molecular aspects of selected inborn errors of metabolism

Inborn errors of metabolism represent a heterogenous group of rare conditions, most having an incidence of less than 1 in 100,000 births. Because of their low prevalence, they are on the margin of attention of general research and even more so of large pharmaceutical companies. Study of rare diseases is the only way to design therapeutic options in order to improve quality of life of affected patients. Present Thesis particularly focuses on disturbances in mitochondrial energy metabolism. The main goals were the characterization of mitochondrial biogenesis within foetal development, as well as in childhood and adulthood. Another aim was to define clinical, biochemical and molecular aspects of mitochondrial optic neuropathies in childhood and adulthood. This work supported the earlier observations that gestational week 22 is the edge of viability, which has to be taken into account in upcoming discussions about guidelines on resuscitation of preterm neonates. Secondly, over last four years, we managed to examine and describe large cohort of patients with optic neuropathies based on a mitochondrial dysfunction.
