Independent predictors of prognosis in patients after out-of-hospital cardiac arrest

The dissertation refers to the up to date topic of Out-of-Hospital Cardiac Arrest (OHCA). The clinical research is divided into two parts. 1. Characteristics and Survival Determinants in Patients After Out-of-Hospital Cardiac Arrest in The Era of 24/7 Coronary Intervention Facilities Background. Survival rates and outcomes after out-of-hospital cardiac arrest (OHCA) remain low despite investments of time and money. The goal of this analysis was to identify variables related to survival of patients transferred to our coronary care unit (CCU) after an OHCA. Methods. 102 consecutive OHCA patients, mean age 64.6 (SD 13.3), 70.6 % men, between January, 2011 and December, 2013, who were transferred to our tertiary care CCU, were studied. Results. Cardiac-cause OHCA was present in 84 patients (82.4 %). Of these 60.7 % had an acute coronary syndrome (ACS) - STEMI 35.7 %; NSTEMI 23.8 %. Coronary angiography was performed in 73 (71.6 %) patients - 81.0 % with cardiac- and 31.3 % (5/16) with a non-cardiac cause. Percutaneous coronary intervention (PCI) was performed in 50 patients (68.5 %), 49 with cardiac-cause, and succeeded in 92.0 %. In-hospital mortality was 38.2 %, one-year mortality was 51.5 %. Kinamatic analyses of tennis serve in relation to succesfull serves

Title: Kinematic analysis of the toss in relation to the success of serving in tennis Objectives: The aim of this master's thesis is to conduct a kinematic analysis of the toss, determine the impact the serve will have on the success of the first serve among competitive and recreational female players and compare these data with results of male serves obtained in the bachelor's thesis. Methods: To obtain the results, a method of indirect observation was used, during which video recordings were made of 8 competitive and 2 recreational female players. Each player executed a total of 40 first direct serves, and the partial videos were then analyzed using Dartfish 10 software. We observed the maximum point of the serve and the ball contact, and each serve was categorized as successful, net, out - on the side, or out - long. Data obtained from the video recordings were subsequently processed using effect size analysis (Cohen's d) and statistical significance (ANOVA). Results: Analysis of variance for repeated measures showed a statistically significant difference between individual serve categories on the Y-axis, i.e., maximum toss height, F (3,21) = 7.16, p = 0.002, ηš = 0.013. Evaluation of breast milk production in relation to body composition in Czech lactating women.

The aim of this study was to evaluate the production of breast milk in relation to the body composition of Czech breastfeeding women and to compare the results to similar studies. Six breastfeeding women participated in this specific part of the longitudinal study, undergoing 4 examinations during a 9-month lactation period. Anthropometric methods were used to measure basic body parameters. Furthermore, body composition was assessed using dual bioimpedance analysis with a digital scale and spectroscopic bioimpedance analysis using the Body Composition Monitor device. According to Friedman's nonparametric paired test, statistically significant differences between the examined periods were found in BMI values (p = 0.029), BSA (p = 0.019), waist circumference (p = 0.048) and hip circumference (p = 0.031). These values fall below the significance level of 0.05. Preparation of Cosmetic Product Based on Microbial Extracts

This thesis is focused on the preparation of a cosmetic product based on microbial extracts. The thesis is divided into two main parts, theoretical and practical. The theoretical part describes the types of cultured microorganisms, microbial metabolites, legislation and safety of cosmetic products, composition of human skin and last but not least the applied analyses. The experimental part deals with the use of waste products of the food industry, specifically coffee grounds as a substrate for culture media. Furthermore, the cultivation is studied with a focus on the gain of metabolites useful in the cosmetic industry, the analysis of these metabolites by GC/FID and HPLC/PDA. The work addresses the preparation of microbial extracts, their testing including determination of polyphenol and flavonoid content and characterization of antioxidant activity and antimicrobial activity tested on bacterial cultures of Esterichia coli and Staphylococus epidermidis. The application of the analysed extracts and their application to the final product is also addressed. The last chapter discussed in this thesis to test the final product using Visioscope PC–35, instrument and performing sensory analysis. The cultured strains were Phaffia rhodozyma CCY 77-1-1, Cystophilobasidium macerans CCY 10-1-2, Rhodosporidium toruloides CCY 062-002-004 Chlorella vulgaris CCALA 924. The biomass obtained by cocultivation of Phaffia rhodozyma CCY 77-1-1 and Chlorella vulgaris CCALA 924 as well as the biomass of these two microorganisms cultured separately were used for the preparation of microbial extracts.

Technology of injection molding of thermoplastic test specimens

The diploma thesis focuses on the optimization of technological parameters of plastic injection molding and the study of the influence of technological parameters on the quality of molded test specimens’ type 1A. The quality of molded parts for 3 types of polypropylene (PP) with different melt flow rate (Mosten GB 002, Mosten GB 218, Mosten MA 230) and 1 type of polystyrene (PS) (Krasten PS GP 154) was evaluated in terms of dimensional stability and weight. The contribution of software for modeling the plastic injection molding process was evaluated in this work. SOLIDWORKS Plastics software was used to optimize technological parameters. The construction of the bodies, mold and cooling system was constructed, and test bodies were produced on the basis of parameters obtained from the simulation of the injection molding process. Their quality parameters were compared with a 3D model and for each of the studied materials the optimal technological parameters were selected in terms of quality and the degree of influence of individual injection parameters on the quality of moldings was evaluated. The accordance of the results of the theoretical simulation with the real experiment was proved and a computational module independent of the optimized quality parameters, generally suitable for optimizing the quality parameters of the injected parts, was developed.
Optimization of cultivation of carotenogenic yeasts on mixed waste substrates

The master thesis addresses the issue of cultivation of selected strains of carotenogenic yeasts on waste materials of the food industry using a laboratory bioreactor. Carotenogenic yeasts are able to produce highly valuable metabolites during cultivation, which are located predominantly in the lipid part of the cells. Particularly, they are carotenoids, ergosterol, coenzyme Q and fatty acids. The thesis is divided into two main parts, the theoretical part and the practical part. The theoretical part describes individual yeast strains, types of waste materials, produced metabolites and methods of their analysis. The experimental part deals with the processing of waste materials of the food industry, specifically animal fat, whey and spent coffee grounds into the form of substrates usable as nutrition sources for yeast cultivation. Furthermore, cultivations focused on the recovery of the monitored metabolites and their analysis by using HPLC/PDA and GC/FID assemblies were studied as well. The yeast strains Rhodotorula mucilaginosa (CCY 19-4-6), Rhodotorula kratochvilae (CCY 20-2-26), Rhodosporidium toruloides (CCY 062-002-001), Sporidiobolus pararoseus (CCY 19-9-6) a Cystofilobasidium macerans (CCY 10-1-2) were used in this work. As one of the best producing strains Sporidiobolus pararoseus (CCY 19-9-6) was found, which achieved very high productions of carotenoids, coenzyme Q and ergosterol.

The Bachelor's thesis is focused on the practical tests of standard spiral drills with a new geometry CZ 002 and CZ 004. The first part is devoted to a short description of a history and current state of the art and in particular, the analysis of core according to the standard DIN 1414-1. Another part deals with the description of the test loading. The last part of the work is devoted to test the geometric quality and precision of the bored holes.
Identification DNA of Plant and Animal Species in Food by Polymerase Chain Reaction

We were developing detection methods for three food allergens of plant origin. We used real-time PCR for soy detection in food oriented on gene lec, that is coding lektine specific for soy. On this target sequence were oriented PCR system with primers Le2F and Le2R and TaqMan probe Le2P. Detection limit (2,75 pg), practical detection limit (0,02 %), inclusivity and exclusivity were determined. Whole system were quantified. Real-time PCR for pistachio detection were based on primers and probe for gene COR. Detection limit (3,5 pg), practical detection limit (0,002 %), inclusivity and exclusivity were determined. For almond detection we were not succeed system, that fulfil all qualitative parametres.

The aim of the thesis is to design a two-storey gallery and café building in Chrudim. The thesis has three parts. The first part presents the architectural and structural designs for the building permit. The building consists of two parts: a two -storey gallery and a one-storey café with necessary facilities. There is a green roof above the gallery and a terrace above the café. The envelope wall is insulated with mineral wool and loadbearing structures are designed from reinforced concrete. Internal non-load-bearing structures are designed of aerated concrete and plasterboard. The floors are monolithic reinforced concrete. I evaluated the building for Energy Performance Certificate. The second part proposes the design of selected building services: HVAC, artificial lighting and rainwater management system. There are two mechanical ventilation unit with heat recovery, which partly provide heating and cooling. The remaining heat is provided by local district heating grid. The designs also include concept of a photovoltaic plant on the roof. Third part assesses the life cycle assessment (LCA) of the structures. All the proposed compositions have the same main function - to ensure the stability of the building, fire safety, weather protection and thermal comfort. Six similar compositions of the vertical wall structure are proposed, combining different types of load-bearing masonry with ETICS system, ventilated facade, variant of aerated concrete blocks or VELOX system formwork. For the selected compositions, the functional unit is 1 m2 with a service life of 50 years and a heat transfer coefficient of U=0.216 ± 0.002 W/m2.K.
