This diploma thesis deals with the history of Rokytnice nad Rokytnou, a small town situated in the Bohemian-Moravian Highlands ten kilometres from the town of Třebíč. The focus is on the first half of the 20th century, which brought significant events in this village. The thesis itself deals with the history of Rokytnice nad Rokytnou from many points of view. The demographic development and development of the municipality, the functioning of local self-government, the activities of emerging associations and the operation of two age-old institutions, namely the school and the parish, are examined. There is also an insight into everyday life in the local village. The destinies of many Rokytnice families were also influenced by war conflicts and two totalitarian regimes. Rokytnice is largely affected by events from the early 50's in the Třebíč region. In connection with the so-called Babice case, the priest Jan Bula and the twenty years old Drahoslav Němec were executed, and nine of Rokytnice citizens and one native were also sent to prison. The diploma thesis also deals with the impact of the mentioned events on local citizens. The events in Rokytnice nad Rokytnou also influenced each other with the surrounding villages, especially Chlístov, Markvartice and Římov. For this reason, these villages are...

Grip strength is increasingly used as a bioindicator of quality of life in both older and younger population. That's why we decided to research methods for increasing grip strength. The first method involves using resistance rings for grip strength training, while the second method involves passive hanging. The aim of this study was to investigate two methods for training grip strength and compare their effects on the physiological parameters of the human body after a month of systematic involvement in the daily lives of study participants, with the goal of determining their potential for rehabilitation. Participants were healthy, active, recreational athletes aged 20-60 years. They were stratified and randomly divided into two groups: one trained with resistance rings, and the other was passive hanging. Training sessions lasted 7 minutes each day, and the study duration was 1 month. Input and output measurements included examination of ranges of motion in the shoulders and spine, evaluation of postural functions and grip strength assessment using a dynamometer. A total of 31 participants completed the study, with an average age of 40.7 years, including 15 women and 16 men. Most participants were right-handed, with only one left-handed and two ambidextrous individuals. Statistical analysis revealed...

Correlation of Molecular Biology and Imunohistochemistry Methods in the Diagnostics of Head and Neck Tumors
Klubíčková, Natálie ; Skálová, Alena (advisor) ; Mandys, Václav (referee) ; Stárek, Ivo (referee)
The increasing use of molecular-genetic methods in recent decades has facilitated precise classification of head and neck tumors. This has led to the identification of new entities among salivary, mesenchymal, and sinonasal tumors. Accurate classification not only aids in predicting disease course and determining appropriate treatment intensity but also holds significance in cases where mutations can be targeted with specific therapy. Additionally, newly available immunohistochemical markers can in some cases reliably replace part of the genetic analysis, which brings with it economic advantages and a shorter time needed to examine the biopsy material. The dissertation summarizes and comments on the publication activity of the author regarding salivary, sinonasal and mesenchymal tumors, with a particular focus on their molecular-genetic background and immunohistochemical findings. In the field of salivary tumors, the discussion revolves around the detection of NR4A3 and NR4A2 gene rearrangements in acinic cell carcinoma, as well as the genetic background of sclerosing polycystic adenoma and its implications on morphology and immunoprofile of individual cases. Furthermore, the dissertation lists four publications presenting various cases of sinonasal tumors, ranging from rare types of sinonasal...
Macromolecular systems for photodynamic therapy of tumors and inhibition of key proteins responsible for multidrug resistance
Kotalík, Kevin ; Etrych, Tomáš (advisor) ; Šálek, Petr (referee)
The research in the field of drug delivery systems has been extensively studied in the past decades. One of the fields where this principle is applied, is anticancer therapy. Many drug delivery systems (DDS) are based on macromolecular carriers, which are able to accumulate in the tumor tissue due to the enhanced permeability and retention (EPR) effect. Those macromolecular carriers are mostly based on polymer nanoparticles, polymerosomes, dendrimers, polymer micelles or water-soluble polymer conjugates. An interesting polymer material applicable in DDS is poly[N-(2-hydroxypropyl)methacrylamide] (PHPMA), which is highly hydrophilic, biocompatible and non-fouling to plasma proteins. PHPMA copolymers enable incorporation of functional groups for covalent drug attachment via stable or stimuli (e.g. pH or enzymatic activity) sensitive bonds. Modern approaches to anticancer therapy are e.g. photodynamic therapy (PDT), which uses photosensitisers, compounds able to generate cytotoxic reactive oxygen species (ROS) upon irradiation. Another trend is the usage of inhibitors of antiapoptotic proteins (IAP), which are able to increase the therapeutic effect in combination with classical cytostatics. In the treatment of advanced or highly resistant tumors, combination of such therapies is key to successful...
