HOW TO MOVE ON WHEN THE STATION IS GONE?

The diploma thesis is based on the pre-diploma project, which deals with the vacant buildings after the railway is moved from the center of Brno to the newly proposed location of the transport hub according to the KAM study. The diploma project focuses specifically on the heritage-listed building of the Brno main railway station. The aim is to find a suitable function for this building and to integrate it into the context between the historic center and the newly built southern quarter. The proposal also solves the building's surroundings, which include the area of the vacant railway track and the station forecourt with its complex traffic situation, which has been one of the least popular public spaces in the city for many years.
A new life for the factory = a new life for Portlaw

Master´s thesis describes the revitalization of a former cotton factory site and later tannery. The abandoned factory building, the chimney, the former engine room, the canal, and other secondary buildings are uncontrollably absorbed by nature. The place has an exceptional charm thanks to the contrast of raw materials and the amount of greenery, but the lack of maintenance of individual structures shortens their lifespan. The main idea of the design is the conservation of the preserved objects of the factory, support of historical values, and preservation of nature in a man-made space. The proposal describes the creation of a multi-purpose area that creates employment opportunities, a form of recreation and public space for the residents of Portlaw, as well as an attraction within the region and for tourism. The functional content is a botanical garden with an exterior and interior part, a public space with a market, small spaces for rent, a food court, accommodation, and areas with hydroponics. Each functional unit works independently, and at the same time, complements each other - grown products from hydroponics are sold at the market, visitors to the botanical garden use accommodation, and people who rent spaces visit the food court. The goal was to bring new life to the factory and thus revive the town of Portlaw. The functional content of the entire area uplifts the history as well as the current state of the place.

Analysis of the impact of conservation on the price of real estate in comparison with the real estate now located outside the reservation in Mikulov

The aim of the thesis is to asses an influence of a conservation on real estate price determined using comparative method and regular price and to define potential complications connected to using such real estate. Theoretical part of the work is focused on description of selected evaluation methods, basic concepts related to conservation and issues connected to real estate located in conservation. Following part of the thesis is description of selected area, specifically description of Mikulov city, Mikulov city conservation and protection zone of Mikulov city conservation. Practical part of the thesis is calculation of price of the real estate, subsequent comparison of the prices and evaluation of influence of conservation on real estate price.
Evaluation of industrial heritage

The main subject is evaluation of effective use in terms of care (not only within protection of monuments) in order to save and protect industrial heritage. Pointing out on critical issues connected with the end of production of traditional branches which played the key role in historical development of our cities. Nowadays abandoned buildings and sites are represented by memory of the place or its prosperity. New owners often don't share the positive approach to the industrial heritage, developers are seeking new sites, cities and inhabitants have developed new needs through the time. The work will demonstrate examples within European context and will make recommendations in monitoring and classification of industrial heritage to prevent undo able losses.
Evaluation criteria of industrial heritage - practical application

The protection of monuments in Slovakia is mostly concentrated on „classical“ examples of cultural heritage (castles, historical city centers). It is not paying much attention to industrial buildings, which are important documents of national development in European context from the era of industrialization. These buildings are often also on the edge of public interest, due to their brownfield character, what places then into the great risk. Because many of these buildings are in private hands, it is often complicated to define the protection policy. It is in the interest of healthy development of society to protect cultural heritage as part of its own identity. The thesis is monitoring approaches to protection of monuments in Slovakia, as well as in global context and offers alternative approaches in terms of protection of industrial heritage.
Plasma cleaning of corroded plumb objects

This bachelor thesis deals with the application of low-pressure hydrogen plasma to lead corrosion products. There are many historical objects and these objects require careful removal of corrosion layers. Therefore, it is necessary to optimize the treatment process in order not to destroy real historical artefacts in the future. This work is focused on lead, which has not been explored by this method yet. Model samples with corrosion were prepared for comparison and these samples corrode in vapors of concentrated acetic acid and formic acid. There were three working temperatures for plasma chemical treatment because of the low lead melting point and the atomic heating. Argon-hydrogen plasma and hydrogen plasma was applied to 12 corroded samples during the experiment. Process progress was monitored by optical emission spectrometry and surface analysis was performed by SEM, XRD and EDX analysis. The results of these analyzes showed that the most appropriate temperature for plasma chemical treatment was 130 ° C. Based on the obtained results many suggesstions and ideas were obtained for the future research like using of pulse mode and gas composition.
Sequences forming G-quadruplexes in the amyloid beta precursor human gene and its homologues

The APP gene encodes the transmembrane protein amyloid beta precursor, which is expressed in many cell types, including neurons. Its functions have not yet been fully described, but it is clear that it is being cleaved before being exported to the extracellular space. It is degraded by various degradation pathways also undergoes homodimerization, which can produce particles with protective neuronal function as well as fragments that are toxic and cause nerve cell death. The formation of harmful amyloid beta plaques and their accumulation among neurons in the brain is closely linked to the onset and progression of Alzheimer's disease, a neurodegenerative brain disease manifested by death and loss of neurons, which leads to dementia, i.e. loss of cognitive functions. There is currently a lot of research that deals with the links between neurodegenerative diseases and the occurrence of G-quadruplexes in genes that are involved in disease manifestations. G-quadruplexes are non-canonical DNA and RNA nucleic acid secondary structures that arise in guanine-rich regions. They are important mainly in terms of their connection with biological processes such as the regulation of gene expression in genes and mainly in oncogenes because they occur in important regions of the gene such as the promoter. It is possible to stabilize them with small molecules, and it is this ability that is used in research into the therapeutic treatment of various diseases. A bioinformatics analysis of both the human gene and 346 other gene homologs was performed to determine the importance of G-quadruplexes localization and conservation in the human APP gene. For this purpose, the G4Hunter program was used, which provided information about the found sequences with the potential to form a G-quadruplex, such as their location or G4Hunter score. In vitro analysis was performed using thioflavin T reagent, which tested the ability of the found sequences to form G-quadruplexes under physiological conditions. The results confirmed the presence and evolutionary importance of G-quadruplexes found in the APP gene of Homo sapiens and their ability to assemble into quadruplex structures in the presence of salts such as sodium and potassium.

The goal of this doctoral thesis is to map and evaluate the current situation of folk architecture in the region around the city of Uherské Hradiště on the example of selected villages. Later, these findings will be compared with the archive of prof. Antonin Kurial. The text has been extended to include currently listed buildings and output of this work is a new catalogue of buildings of folk architecture in the above mentioned region.
The ruins and the possibilities of their conservation

This bachelor´s thesis deals with a question of conservation of castle ruins. The theoretical part discusses generally building materials used in their construction, sort of material´s degradation and corrosion of materials, possibilities of ruin´s conservation by using suitable building materials, and processes in realization of historical property restoration. Work focuses on a history and conteporary condition of two historical buildings, the castle Cimburk in Czech republic and the castle Lietava in Slovakia. As a part of my work was collection and subsequent analysis of samples of historical mortars of these castle. Resulting response curves differential thermal analysis, RTG diffraction analysis and their mutual comparison.
