Variability in extracellular phosphatase activity of dominant phytoplankton species in the Bohemian Forest lakes

Phosphorus (P) is considered as the most frequently limiting factor for phytoplankton growth in freshwater environments. One of the ways, how to solve the lack of P in algae and cyanobacteria, is the production of extracellular phosphatases. Extracellular phosphatase activity (PA) of phytoplankton at the single cell level was investigated in three acidified mountain lakes - Čertovo, Prášilské and Plešné in the Bohemian Forest (Czech Republic) from May to September 2007. PA of phytoplankton was detected directly at the site of enzyme activity using the fluorescence labelled enzyme activity technique (FLEA) and epifluorescence microscopy. The FLEA technique is based on colourless substrate ELF phosphate (ELFP) that is converted by enzymatic hydrolysis to the insoluble fluorescent product ELF alcohol (ELFA), which marks the site of PA. This thesis is divided into two parts. The first part (in Czech) is aimed at a general comparison of PA of phytoplankton in the three Bohemian Forest lakes. PA was characteristic for dinoflagellates Gymnodinium uberrimum, Gymnodinium sp. and Peridinium umbonatum that were ELFA labelled in all lakes almost during the whole season. Changes in spatial distribution of gelatinous zooplankton: causes and consequences

During the day, changes in the spatial distributions of organisms in the water column occur repeatedly, especially in diurnal vertical migrations, which affect abiotic and biotic components of ecosystems. The migratory behaviour occurs worldwide in the oceans and in fresh water, in different habitats and under different physical conditions. The main migrants are gelatinous zooplankton, which are a heterogeneous group of invertebrate animals, although phytoplankton, nekton and other representatives of zooplankton also migrate. The main focus of the study of diurnal vertical migrations is proximate and ultimate factors that influence the migration. Furthermore, occurrence and density of organisms are examined. To study vertical migrations of gelatinous zooplankton, it is the easiest to study jellyfish because of their size. The distribution behaviour might also be observed in freshwater zooplankton. Two university studies of diurnal vertical migration of freshwater jellyfish were executed in previous years. Methane emissions from temperate freshwater reservoirs

Methane (CH4) emissions from temperate freshwater reservoirs have only been evaluated for the last several decades. Freshwater ecosystems (including water reservoirs) release crucial quantities of this important greenhouse gas. Ebullition that releases methane into the atmosphere from bubbles originating from sediment is important source of methane. Regardless of the importance of these emissions, a very limited number of scientific research papers monitoring this flux into the atmosphere are available. Estimated values of methane emissions from freshwater temperate reservoirs vary considerably. There are many different methods for monitoring methane emissions. It seems that the freshwater reservoir stimulates methane formation in increasing scales. The size of the sum of methane emitted from freshwater reservoirs is also difficult to say.
Morphogenesis of the bacterial colonies and their mutually influencing

This thesis follows previous works of our group (Rieger T. et al., 2008; Cepl J. et al., 2010 and Patkova I. et al., 2012), where we focused on the morphology of the bacterial colonies Serratia marcescens and its variety caused by changing of the inoculation conditions on nutrient agar. When bacterial colonies S. marcescens are grown on nutrient agar enriched with glucose isolated enough from other colonies in its living space, it can form coloured structured colonies, which we named morphotype "fountain" (F). This morpotype becomes ideal for following studies of mutual influencing of the bacterial colonies, because of its ability of pigmentation change or structure loss caused by altering surrounding inoculation conditions. We noticed in normal sowed agar plates, that bacterial colonies, which grows in the close distance with other colonies develop their pigmentation sooner, than colonies, that grows more isolated. We studied how is this influencing happening and what are the necessary conditions for it. We proved, that different species of bacterial macrocolonies (S. marcescens - morphotype (M), S. rubidea and E. coli) emits into the nutrient agar informative signal, which makes the recipient colonies S. marcescens reacts on this signal with the same manner (X structure). Dispersal ability of anostracans and its impact on genetic structure of populations

1 ABSTRACT The coldstenothermal species Eubranchipus grubii (Dybowski, 1860) is restricted only to vernal temporary pools in floodplain and does not occur in any other type of water habitat. Organisms living in these habitats have to be well adapted to low temperatures, desiccation, low nutrient levels, and have the ability to migrate among habitats and colonize newly established ones. E. grubii survives periods of desiccation in the form of cyst. These cysts serve as a mean of dispersion. In the Czech Republic E. grubii occurs in watersheds of rivers Labe, Morava, Odra and Dyje. We collected 95 populations in these different regions. The extent of genetic divergence among populations was assessed by analyzing sequence variation in mitochondrial gene (COI). Comparing levels of mtDNA sequence divergence divided studied populations into three main groups according to watersheds. Genetic relationships among populations inhabiting different watersheds showed 1,7 - 5,7 % sequence divergence. Sequence divergence within populations is very small. It seems that E. grubii colonized the area of today's Czech Republic from different refugia.

Fytoplankton aluviálních tůní

Phytoplankton of alluvial pools at Lužnice alluvium have been investigated by following methodological approaches: seasonal course of phytoplankton in two pools, extensive sampling (quarterly) of 30 pools, and phytoplankton diurnal vertical migration studies in several pools. Prevailing dominance of phytoflagellates, especially Cryptophyceae and Euglenophyceae and low contribution of coccal forms as Chlorococcales and Cyanophyte has been found as a main feature of phytoplankton community. This is caused by poor mixing of water column (large relative depth, wind protectio), where th ecoccal forms sediment out and are outcompeted by motile forms, able to determinate the position within the water column.
