Analysis of Historical Pharmaceutical Preparations of Caffeine and Santonin.

Historical pharmaceutical preparations of caffeine and santonin were analyzed by mass spectrometry, the sample of caffeine was quantified by spectrophotometry, the sample of santonin was quantified by acid-base titration. In the analyzed historical injection solution "Coffeinum natriosalicylicum" (dated circa 1945) was determined 110 ± 1 % of the declared amount of caffeine and 94.33 ± 0.02 % of the declared amount of sodium salicylate. There were observed neither degradation products of caffeine nor sodium salicylate. The content of santonin in the historical sample (dated prior 1918) was determined by titration to 104 ± 4 %, sample was pure santotnin with no additions. No degradation products of santonin was observed. The analyzed samples were stable for more than seventy, ninety resp., years of storage.
Liquid Chromatography Methods for Analysis of Actinomycete Secondary Metabolites - Potential Antibiotics

(EN) This dissertation thesis contains scientific results achieved in the field of analytical chemistry, particularly liquid chromatography. The major part of the results has been published in prestigious international journals in five papers. In addition to that, relevant yet unpublished results have been included as well. In general terms, the work presented here contributed to the concerted efforts to tackle the current lack of novel antibiotics. Specifically, high-performance liquid chromatography (HPLC) and ultra high-performance liquid chromatography (UHPLC) techniques coupled to a variety of detection systems have been employed for analysis of antibiotics and actinomycete secondary metabolites. The first thematic part describes the development of liquid chromatography methods for analysis of lincomycin precursors, lincomycin precursor analogues, and lincomycin derivatives. The methods have been applied to study lincomycin biosynthetic pathway and obtain improved lincomycin derivatives by mutasynthesis. The second thematic part aims at investigating alternative approaches for analysis of antibiotics. Firstly, the core-shell particle and the sub-2 μm particle chromatographic columns were compared. Quantitative Structure-Retention Relationships of Salicylthioamide Derivatives

The aim of the thesis is to study the relationship between the structure of N-benzylsalicylthioamide derivatives and their retention behavior in high-pressure liquid chromatography. Furthermore, the influence of various substituents on electron absorption spectra in ultraviolet spectral range was studied. All studied derivatives have two absorption maxima in ultraviolet spectral range at 260 nm and 293 nm. The substitution of an auxochrome on the acyl ring leads to bathrochromic shift and hyperchromic shift, especially of the first absorption maxima. The presence of an auxochrome on the amide ring leads to hyperchromic shift as well. HPLC reversed-phase chromatography, using XDB-C18 ZORBAX column and mobile phase with various composition of acetonitrile, was used to obtain retention times of the derivatives. Using this data, the correlation equations between retention factor (mobile phase with no acetonitrile), resp. slope of the dependence of the logarithm of retention factor on the volume fracture of acetonitrile in mobile phase, and partition coefficient octanol-water, resp. hydrophobic substituent constants, were derived. Voltammetric detection of stigmasterol at boron-doped diamond electrode in mixed media

Cyclic voltammetry in mixed media of water and organic solvents was used for this purpose. First of all, the effect of organic solvent on potential window of boron doped diamond electrode was studied. Solvents used were methanole, isopropanole, N-dimethylformamide and acetonitrile; the water components was phosphate buffer (0,075 mol.l-1, pH 3,0). It was deduced from the results, that suitable solvent for detection of stigmasterol on was the acetonitrile, which provided sufficiently large potential window in anodic area.
