Individualization of the treatment of prostate cancer patients based on the immunocytochemical detection of circulating tumor cells

Introduction: Together with the introduction of new therapeutic options in castration- resistant prostate cancer (CRPC), an advance in individual disease characterization is required. Since common biopsy methods are not suitable for the majority of CRPC patients, one possible solution is the liquid biopsy that is, the analysis of circulating tumor cells (CTCs) isolated from the cancer patients' blood. Methods: A method based on the immunomagnetic enrichment of CTCs and subsequent PCR detection of tumor-associated genes (AdnaTest, Qiagen) was characterized and used for the detection of CTCs in 41 CRPC patients. Each patient was screened at the time of CRPC diagnosis and after the 3rd cycle of docetaxel therapy. A panel of genes associated with therapeutic decision-making was established and validated. Quantitative PCR (qPCR) method on a BioMark platform (Fluidigm, USA) was used to determine the expression of the gene panel in the CTC-enriched and primary tumor samples and the results were analyzed. Results: CTCs were found in 85% and 45% of CRPC patients before and during the therapy, respectively. The presence of CTCs, as well as EGFR and AR PCR fragments, was associated with a decreased sPSA response and lower survival. Application for Time Management

This work presents a Windows application developed to increase user focus and effici- ency through effective time and schedule management strategies. The software integrates established methods such as Eat That Frog!, the Eisenhower Matrix, the 80-20 Rule, and the modified Eat That Frog! along with the Pomodoro Technique to help users create and stick to optimized schedules. After successfully mastering the activity in the schedule, the user has the opportunity to share his result with other users, which also increases motivation. The study underscores the app's role in promoting improved work habits and concentration. The development process, from design to implementation, prioritizes practical functions in the creation of a schedule.
CSV file validator according to the CSV on the Web W3C recommendations

The goal of this thesis was to create a validator of CSV files according to the CSV on the Web recommendations, which enhance CSV data format. This validation is needed to increase the quality of data on the web. Validator offers multiple user-friendly interfaces in the form of CLI app, web service and web app. The validator itself is a library implemented in the popular object-oriented programming language C#. In this thesis we will describe the development of the validator, starting with the recommendations and analysis of existing solutions, through the design of architecture and user interfaces, and finish with the description of implementation, testing, and evaluation of software solution. 1
Analysis of variations of stochastic integrals

In this thesis, we study the 1/H-variations of stochastic integrals, where the integrators are the fractional Brownian motion and Rosenblatt process (with the Hurst parameter H> 1/2). The considered stochastic integrals are defined as the Skorokhod integrals within the framework of Malliavin calculus. We summarize the already established results about the 1/H-variation of the integral with respect to the fractional Brownian motion and then apply the techniques used therein to obtain the form of the 1/H-variation of the integral with respect to the Rosenblatt process. 1
Usage of a luminance analyzer for analysis of natural light sources

This bachelor thesis is focused on the description of photometric quantities, presentation of methods of measuring illuminance and luminance, and explanation of the principle of devices used to measure these quantities. It also deals with the functions of the eye, its sensitivity, and changes in vision due to the alternation of day and night. The last chapter describes the origin of light in the natural sources of light, the Sun, the Moon, and fire, and then presents the measured values with their analysis.
Network Infrastructure Design for Family House

This thesis deals with the network infrastructure design for family house. The default aspects for the design of network infrastructure are house plans and requirements of the client. The work presents the theoretical knowledge necessary for further analysis of client requirements and design optimal solution. Part of the work are cabling plans, location of active elements and cost calculations to build the network infrastructure.
Microcosm on the Road

Although there is an international vehicular consensus about signing, the maximum speed allowed or connecting the most significant regions together, their nature is different all based on geographical, cultural or social conditions. Rest stops along the roads reflect each country dispositions. Rest stops are the place where different social and national clusters meet. At this moment building up the rest stops is on behalf of the Czech republic which is not that much interested in it. Instead of taking care and maintain them, because the current state of the reststops is not satisfactory, the Czech republic is letting them to the third parties - supranational corporations. This leads to unpleasant lines along the highways accompanying by petrol stations and cheesy fastfoods. There is no place for rest stops without the need of consummation. The Czech republic has currently begun planning and building up new kilometers of highways and the new opportunities of rest stops arise. Because the existing site is overloaded with petrol stations, supranational corporations are no longer interested in these plots. State has an opportunity to take over and redesign these neglected plots on his own.
The effect of physical properties of building materials in buildings in low-energy construction

The theme of the diploma thesis is the study of physical properties of various building materials in buildings in low-energy construction. The work is mainly focused on the influence of the thermal accumulation properties of building materials used in buildings in order to achieve low energy consumption for heating. The thesis compares the energy characteristics of five different constructional material variants of the house.
High speed steel milling cutters made by powder matallurgy

Bachelor thesis is a research work describing milling cutters made of high speed steel by powder metallurgy (HSS PM milling cutters). In chapters one, two and three are generally described milling, high speed steels and powder metallurgy. In chapter four are introduced particular HSS PM milling cutters of a few producers. The summary contains mechanical properties of cutters and suitability for machining. It is focused on main type of HSS PM cutters, which are the end mills.
Pozitive and negative selection of mitochondrial genome

This bachelor thesis deals with positive and negative selection detection in mitochondrial genome of invertebrates and vertebrates. The component of this paper is to interpret the theory of evolution and evolution models. The main objective of this bachelor thesis is to formulate the algorithm package for a positive and negative selection detection in a program environment, RStudio. Consequently, outcomes of algorithm are compared to values acquired by tool PAML.
