Study of physiological functions of betaine homocysteine S-methyltransferase and betaine homocysteine S-methyltransferase 2

Betaine homocysteine S-methyltransferase (BHMT) and betaine homocysteine S-methyltransferase 2 (BHMT-2) are mammalian cytosolic metalloenzymes. They both participate in the metabolism of homocysteine (Hcy), specifically Hcy remethylation, mainly in liver and kidney cells. BHMT catalyzes the transfer of a methyl group from betaine to L-Hcy, yielding L-methionine and dimethylglycine (DMG). BHMT-2 catalyzes the transfer of a methyl group from S-methylmethionine (SMM) to L-Hcy as well, yielding two molecules of L-methionine. Disorders in Hcy metabolism could lead to the so called hyper- homocysteinemia and homocystinuria, which can be connected with several pathological conditions. BHMT is already relatively well characterized enzyme. Its crystal structure and reaction mechanism have been described and a series of BHMT inhibitors have been prepared. The specific inhibitors enabled further in vivo studies and, recently, Bhmt-/- mice model has been successfully developed. In contrast, the research of BHMT-2 is still at the beginning and physiological functions of the enzyme are unknown so far. Investigation of the protein interactions involved in the acylation of Escherichia coli α-hemolysin

Uropathogenic bacteria Escherichia coli are the primary cause of urinary tract infections. One of the virulence factors of these bacteria is α-hemolysin (HlyA), a protein belonging to the cytolytic RTX (Repeats in ToXin) toxins secreted by some gram-negative pathogenic bacteria. RTX toxins share several characteristic structural and functional domains and segments: (1) an N-terminal hydrophobic pore-forming domain, (2) an acylated segment where two lysine residues are modified by fatty acid chains, (3) a repetitive (RTX) domain binding calcium ions, and (4) a C-terminal secretion signal recognized by the type 1 secretion system. HlyA is synthesized as an inactive protoxin (proHlyA), which is activated by covalent acylation of the ε-amino groups of two conserved lysine residues, K564 and K690, by the co-expressed acyltransferase HlyC. Acyl-acyl carrier protein (acyl-ACP) serves as the acyl chain donor. However, the molecular mechanism by which the acyltransferase HlyC interacts with acyl-ACP and proHlyA is currently poorly understood. The thesis "System for Monitoring the Technical Condition of Irradiators" focuses on developing a system that simplifies the display, interpretation, and subsequent processing of data obtained from stability tests on linear accelerators. The thesis is divided into theoretical and practical parts. The theoretical part deals with radiotherapy and the technical tests conducted. The practical part describes the development and implementation of the system. The resulting product will be used in clinical practice at the Radiation Oncology Clinic of the Masaryk Memorial Cancer Institute.
Návrh algoritmu jízdní stability pro laboratorní model vozidla

This Master's thesis deals with a research study in the area of modelling the dynamics of a four-wheel drive (4WD) and four-wheel steer (4WS) vehicle, including a tyre model together with the design of vehicle stability control algorithms. The work also includes making the necessary modifications to the experimental vehicle, including the design of new control electronics. Based on the proposed dynamic model, the estimation of the necessary vehicle parameters is carried out. On the simulation model, ABS/ASR driving stability algorithms are designed, which are than implemented on the experimental vehicle.

The analysis for the extension of the control system and proposal of controlling program for surveillance cameras and related theoretical analysis necessary for understanding problems. Proposal communication protocol to access all functions via TCP/IP communication. Subsequent protocol implementation into control program and a programmable controller. Programming application with a user interface for camera control and basic image processing. The program also represents the server for TCP/IP communication with clients over the proposed protocol. The proposed application has to fulfill high pretensions of data streaming from cameras. Extension of the existing control system of the telescope, dome and other systems. The addition of analog measurement and control of digital inputs and outputs. Furthermore, control of stepper motors to control focusers. The embedded communication protocol will be expanded of the new instruction.

Subsequent characterization of all elements from the perspective of the control system and extension of technological scheme of measuring and regulating signs. For the section of the brewery brewhouse writing technological sequence with specifying the steps. Based on this description, create a control program for Simatic S7-300 from Siemens. The program is written in STL which takes the form of structured text. Visualization and process control is provided by SCADA system TomPack from the company ProjectSoft. The system communicates with the PLC via DDE server.
